X-ray diffraction patterns from human arterial specimens containing atherosclerotic fatty streak lesions exhibited a single sharp reflection, corresponding to a structural spacing of about 35 A. Specimens without lesions did not. When specimens with fatty streaks were heated, an order-to-disorder phase transition was revealed by the disappearance of the sharp reflection. The transition was thermally reversible and its temperature varied from aorta to aorta over a range from 28 degrees to 42 degrees C. Since cholesteryl ester droplets are a major component of fatty streaks, comparison studies were made of the diffraction behavior from pure cholesteryl esters. We found that the diffraction patterns of the fatty streak material could be accounted for by the organization of the cholesteryl esters into a liquid-crystalline smectic phase that melts from the smectic to a less ordered phase upon heating. When combined with the conclusions of others from polarized light microscopy, our study shows that a droplet in the smectic phase has well-defined concentric layers of lipid molecules. In each layer, the long axes of the molecules have a net radial orientation with respect to the droplet, but the side-to-side organization is disordered. We suggest that the accessibility of portions of the lipids for specific binding to enzymes or transport proteins may be restricted when they are in the smectic state, and that exchange of lipids with surrounding membranes or other potential binding sites may likewise be inhibited. Regulators of G protein signaling (RGS) proteins act as GTPase-activating proteins (GAPs) toward the alpha subunits of heterotrimeric, signal-transducing G proteins. RGS11 contains a G protein gamma subunit-like (GGL) domain between its Dishevelled/Egl-10/Pleckstrin and RGS domains. GGL domains are also found in RGS6, RGS7, RGS9, and the Caenorhabditis elegans protein EGL-10. Coexpression of RGS11 with different Gbeta subunits reveals specific interaction between RGS11 and Gbeta5. The expression of mRNA for RGS11 and Gbeta5 in human tissues overlaps. The Gbeta5/RGS11 heterodimer acts as a GAP on Galphao, apparently selectively. RGS proteins that contain GGL domains appear to act as GAPs for Galpha proteins and form complexes with specific Gbeta subunits, adding to the combinatorial complexity of G protein-mediated signaling pathways. 相似文献

The NMR solution structure of an immunoglobulin superfamily module of twitchin (Ig 18') has been determined and the kinetic and equilibrium folding behaviour characterised. Thirty molecular coordinates were calculated using a hybrid distance geometry-simulated annealing protocol based on 1207 distance and 48 dihedral restraints. The atomic rms distributions about the mean coordinate for the ensemble of structures is 0.55( +/- 0.09) A for backbone atoms and 1.10( +/- 0.08) A for all heavy atoms. The protein has a topology very similar to that of telokin and the titin Ig domains and thus it falls into the I set of the immunoglobulin superfamily. The close agreement between the predicted and observed structures of Ig 18' demonstrates clearly that the I set profile can be applied in the structure prediction of immunoglobulin-like domains of diverse modular proteins. Folding studies reveal that the protein has relatively low thermodynamic stability, deltaG(H2O)U-F = 4.0 kcal mol(-1) at physiological pH. Unfolding studies suggest that the protein has considerable kinetic stability, the half life of the unfolding is greater than 40 minutes in the absence of denaturant. 相似文献

Fragile histidine triad protein (Fhit) is a diadenosine triphosphate
(ApppA) hydrolase encoded at the human chromosome 3 fragile site which is
frequently disrupted in tumors. Reintroduction of FHIT coding sequences to
cancer cell lines with FHIT deletions suppressed the ability of these cell
lines to form tumors in nude mice even when the reintroduced FHIT gene had
been mutated to allow ApppA binding but not hydrolysis. Because this
suggested that the tumor suppressor activity of Fhit protein depends on
substrate-dependent signaling rather than ApppA catabolism, we prepared two
crystalline forms of Fhit protein that are expected to model its
biologically active, substrate-bound state. Wild-type and the His96Asn
forms of Fhit were overexpressed in Escherichia coli, purified to
homogeneity and crystallized in the presence and absence of ApppA and an
ApppA analog. Single crystals obtained by vapor diffusion against ammonium
sulfate diffracted X-rays to beyond 2.75 A resolution. High quality native
synchrotron X-ray data were collected for an orthorhombic and a hexagonal
crystal form.

Finite element mesh adaptation methods can be used to improve the efficiency and accuracy of solutions to computational modeling
problems. In many applications involving hexahedral meshes, localized modifications which preserve a conforming all-hexahedral
mesh are desired. Effective hexahedral refinement methods that satisfy these criteria have recently become available; however,
due to hexahedral mesh topology constraints, little progress has been made in the area of hexahedral coarsening. This paper
presents a new method to locally coarsen conforming all-hexahedral meshes. The method works on both structured and unstructured
meshes and is not based on undoing previous refinement. Building upon recent developments in quadrilateral coarsening, the
method utilizes hexahedral sheet and column operations, including pillowing, column collapsing, and sheet extraction. A general
algorithm for automated coarsening is presented and examples of models that have been coarsened with this new algorithm are

Abstract This paper discusses the applicability of the ideas of fuzzy sets and grades of membership to problems encountered in the quantification of clinical (i.e., diagnostic and prognostic) judgment. The methods of constrained maximum likelihood are used to derive consensus estimates of grades of membership given a set of categorical data and an a priori set of specified pure types.
