A solid-state 2H NMR study of methyl-d3-cobalamin has been performed as a function of temperature to provide information concerning the character and energetics of the motion performed by this unique bioorganometallic methyl group. Analysis of the 2H NMR line shape indicates that the methyl group undergoes rapid three-fold rotation, and that the Co-C-2H angle lies between 105.9 and 109.5 degrees. Determination of the spin-lattice relaxation times T1 shows that the relaxation is anisotropic, consistent with a "jumping" motion of the methyl group rather than rotational diffusion. This also provides the activation energy to methyl jumps as 8.3 +/- 1.3 kJ/mol. It is proposed that this energetic barrier may be a useful probe of changes in the electronic character of the Co-C bond that accompany the biological role of this molecule in such enzymes as methionine synthase.  相似文献

Normal diffusion of interstitial oxygen atoms (Oi ) accounts for the rate of oxygen aggregation in silicon for T > 500C. There is evidence for the dissociation of SiO2 precipitates (Ostwald ripening) and the formation of self-interstitials (I-atoms) to accommodate the local increase in volume. For T < 500 C, measurements of the loss of oxygen atoms from solution indicate that O2 dimer formation is the rate-limiting process, but dissociation of dimers must be taken into account when modelling this process. Large clusters of up to 10–20 Oi atoms, usually assigned to thermal donor (TD) defects cannot form unless dimer diffusion is much greater (by a factor of 104 to 107 ) than diffusion of Oi atoms and unless there is dissociation of clusters with the emission of dimers. Hydrogen impurities enhance Oi diffusion by a catalytic process and speed up donor formation. Infrared absorption measurements reveal H-Oi complexes and there is also partial passivation of TD defects to produce shallow thermal donors (STDs).  相似文献

On-section immunocytochemistry is divided into two parts: (i) processing of biological tissue for section microscopy and (ii) immunolabelling of sections. Many of the more successful microscopical methods employ delicate aldehyde fixation of biological tissue followed by "sympathetic" processing into an acrylic resin. Processing regimens do not have to be complicated. Simple and cost effective room temperature protocols utilising partial dehydration have been devised and they can be as effective as the more complex low temperature techniques in preserving both ultrastructure and antigenic reactivity. The embedded material can be investigated by either light or electron microscopy. Frozen sections can be cut and immunolabelled but only if the tissue is chemically fixed first, as in resin embedding. Fixation with low concentrations of aldehyde will normally better preserve tissue immunoreactivity but this may be at the expense of good ultrastructure with these protocols. If so, low temperature resin embedding methods or rapid freezing and cryosubstitution can be tried. The choice of processing protocol will determine which acrylic resin to use, as will the preference for subsequent immunolabelling with either colloidal gold or peroxidase/diaminobenzidine (DAB). Both types of labelling system offer advantages to localisation studies and can be used in combination for double or even triple labelling. Silver enhancement of the colloidal gold or DAB allows for improved observation by light microscopy.  相似文献

Despite the seeming ubiquity of young people's Internet use, there are still many for whom access to the Internet and online social networking remains inequitable and patterned by disadvantage. The connection between information technology and young people with disabilities is particularly under‐researched. This article contributes to the field of critical information systems research by exposing significant barriers and facilitators to Internet accessibility for young people with disabilities. It uses Bourdieu's critical theory to explore how the unequal distribution of resources shapes processes of digital inclusion for young people with disabilities. It highlights access needs and experiences that are both disability and non‐disability related. The article draws on interviews in South Australia with 18 young people aged 10–18 years with a physical disability (such as cerebral palsy) or acquired brain injury and with 17 of their family members. Interviews evaluated participants' and parents' reflections on the benefits of a home‐based, goal‐oriented intervention to increase the young person's Internet use for social participation purposes. The Bourdieuian analysis demonstrated how varying levels of accrued individual and family offline capital resources are related to digital/online resources and disability‐specific online resources. This revealed how unequal resources of capital can influence technology use and hence digital inclusion for young people with disabilities. Our study demonstrates that young people with particular types of disabilities require intensive, personalised and long‐term support from within and beyond the family to ‘get online’. We conclude that Internet studies need to more frequently adopt critical approaches to investigate the needs of users and barriers to information technology use within sub‐groups, such as young people with disabilities. © 2016 John Wiley & Sons Ltd  相似文献

The possibility of developing a simple, inexpensive and specific personal passive “real-time” air sampler incorporating a biosensor for formic acid was investigated. The sensor is based on the enzymatic reaction between formic acid and formate dehydrogenase (FDH) with nicotinamide adenine dinucleotide (NAD+) as a co-factor and Meldola's blue as mediator. An effective way to immobilise the enzyme, co-factor and Meldola's blue on screen-printed, disposable, electrodes was found to be in a mixture of glycerol and phosphate buffer covered with a gas-permeable membrane. Steady-state current was reached after 4–15 min and the limit of detection was calculated to be below 1 mg/m3. However, the response decreased by 50% after storage at −15°C for 1 day.  相似文献

A number of human-centred methodologies--subjective, objective, and combined--are used for slipperiness measurement. They comprise a variety of approaches from biomechanically-oriented experiments to psychophysical tests and subjective evaluations. The objective of this paper is to review some of the research done in the field, including such topics as awareness and perception of slipperiness, postural and balance control, rating scales for balance, adaptation to slippery conditions, measurement of unexpected movements, kinematics of slipping, and protective movements during falling. The role of human factors in slips and falls will be discussed. Strengths and weaknesses of human-centred approaches in relation to mechanical slip test methodologies are considered. Current friction-based criteria and thresholds for walking without slipping are reviewed for a number of work tasks. These include activities such as walking on a level or an inclined surface, running, stopping and jumping, as well as stair ascent and descent, manual exertion (pushing and pulling, load carrying, lifting) and particular concerns of the elderly and mobility disabled persons. Some future directions for slipperiness measurement and research in the field of slips and falls are outlined. Human-centred approaches for slipperiness measurement do have many applications. First, they are utilized to develop research hypotheses and models to predict workplace risks caused by slipping. Second, they are important alternatives to apparatus-based friction measurements and are used to validate such methodologies. Third, they are used as practical tools for evaluating and monitoring slip resistance properties of footwear, anti-skid devices and floor surfaces.  相似文献

High-T c superconductivity has generated a great deal of interest because of the challenges it presents in the fields of material science, condensed matter physics, and electrical engineering, and because of the potential applications which may result from these research efforts. Thin-film passive microwave components may become the first high-temperature superconducting (HTS) devices available for widespread use and commercialization. In this article, we review aspects of material science, physics, and engineering which directly impact high-T c superconducting microwave devices and discuss issues which determine the performance of these devices. Methods of growing HTS thin films on large-area substrates, techniques for fabricating single-level HTS passive microwave components, and the relevant properties of high-T c superconducting films are discussed, with a focus on thin films of the HTS material YBa2Cu3O7–. Several known mechanisms for microwave loss in both the superconductor and the dielectric substrate are presented. An overview of the general classes of superconducting passive microwave devices is given, and representative microwave devices which have been recently demonstrated are described in detail. Examples of a select few HTS active microwave components are also presented. Potential microwave applications are illustrated with comparisons to current technology.  相似文献

In this paper we describe anO(logN)-bit-step randomized algorithm for bit-serial message routing on a hypercube. The result is asymptotically optimal, and improves upon the best previously known algorithms by a logarithmic factor. The result also solves the problem of on-line circuit switching in anO(1)-dilated hypercube (i.e., the problem of establishing edge-disjoint paths between the nodes of the dilated hypercube for any one-to-one mapping).Our algorithm is adaptive and we show that this is necessary to achieve the logarithmic speedup. We generalize the Borodin-Hopcroft lower bound on oblivious routing by proving that any randomized oblivious algorithm on a polylogarithmic degree network requires at least (log2 N/log logN) bit steps with high probability for almost all permutations.This research was supported by the Defense Advanced Research Projects Agency under Contracts N00014-87-K-825 and N00014-89-J-1988, the Air Force under Contract AFOSR-89-0271, and the Army under Contract DAAL-03-86-K-0171. The role and importance of enterogastric secretion in the disposition and elimination of the weak base, meperidine (pKa 8.63), was studied after intravenous administration (50 mg) of the drug to 6 normal volunteers. Continuous collection of the gastric fluid over a 4-hr period demonstrated the establishment of high gastric fluid/plasma concentration ratios for meperidine (mean about 50, range, 10 to 200). However, the total amount of drug recovered, even after correction for incomplete collection, was only a small percentage of the administered dose. Under basal conditions a mean +/- SE of 1.9 +/- 0.3 mg, equivalent to 3.7% of the administered dose, was found in the total gastric aspirate. Stimulation of gastric secretion by subcutaneous injection of betazole (1.5 mg/kg) increased this recovery to 3.6 +/- 0.3 mg (7.2%) primarily due to the increase in gastric volumen output. Aspiration of the gastric fluid in either the basal or stimulated situation had no observable effect upon the plasma concentration/time profile of meperidine whether assessed by the terminal half-life, t 1/2 beta, or the plasma clearance; control values were 3.8 +/- hr and 1,190 +/- 130 ml/min, respectively. In 2 subjects "bile fluid" was also collected for 2.5 hr and found to contain less than 0.2% of the administered dose. Enterosystemic recycling is therefore of minor importance in the disposition and elimination of meperidine in man.  相似文献
