A 1.3-μm continuous wave lasing operation is demonstrated, for the first time, in a GaInNAs quantum-well laser at room temperature. This lasing performance is achieved by increasing the nitrogen content (up to 1%) in GaInNAs quantum layer. The authors demonstrate a spatially chirped emission wavelength in vertical cavity surface emitting laser (VCSEL) arrays grown by molecular beam epitaxy. The wavelength shift is due to a lateral thickness variation in the Al0.2Ga0.8As cavity, which is induced by a substrate temperature profile during growth. The objective of this study was to assess the effectiveness of alkaline hydrolysis as an alternative ex situ technology for remediating groundwater contaminated with hexahydro-1,3,5-trinitro-1,3,5-triazine (RDX). Treatment in both batch reactor and continuous stirred tank reactor (CSTR) was investigated. RDX reactivity was strongly dependent on the reaction pH investigated (11–13). The batch system achieved pseudo-first-order RDX reaction rates in the range of (0.8–27.7)×10?3?min?1, corresponding to half-life periods of 17.9?to?0.5?h, respectively. In the CSTR system operated at the initial RDX concentration of 4.5×10?3?mM, 99% RDX removal was achieved with the hydraulic retention time of 2?days and the reaction pH of 11.9. Formate and nitrite were produced as the major hydrolysates in the CSTR system, indicating a simultaneous reaction mechanism involving RDX ring cleavage and elimination of the ring nitrogen. The net OH? demand used only for RDX removal in the CSTR was found to be 1.5, 390, and 130?M OH?/M RDXremoved at pH values of 11.9, 11.5, and 11.0, respectively. A conceptual cost analysis indicated that the expense of alkaline treatment may be comparable to the expense of granular activated carbon treatment for long treatment periods (30?years or more), due to the potentially lower annual operational cost of alkali treatment.  相似文献

A number of hypercube-variant networks attempt to improve the hypercube by adding extra connections and thus reducing the diameter of the constructed network. We briefly outline a model which describes these variant networks. Further, we show that by restricting this model, we can describe hypercube variants with exactly the same number of edges as the hypercube. We mention several such networks which all have diameter about n/2. We describe a new network within this class that has diameter about 2n/5, thus improving the best known previous bound by a constant factor. We show that within a limited construction paradigm our network is best possible. This revised version was published online in June 2006 with corrections to the Cover Date.  相似文献

Experience of scintigraphic detection of bone lesion and active bone marrow involvement of multiple myeloma, especially with sestamibi and FDG-PET scans is in evolution. We report a case of intense sestamibi uptake in bone marrow correlating with the extent of the disease, while FDG-PET scans showed activity only in areas of active disease progression associated with pain. Technetium-99m-sestamibi appears to indicate the extent of the disease, while [18F]FDG-PET scans show sites of active tumor proliferation and may be useful in directing local therapy such as radiation.  相似文献

The Lewis alpha(1,3/1,4)-fucosyltransferase, Fuc-TIII, encoded by the FUT3 gene is responsible for the final synthesis of Lea and Leb antigens. Various point mutations have been described explaining the Lewis negative phenotype, Le(a-b-), on erythrocytes and secretions. Two of these, T202C and C314T originally described in a Swedish population, have not been found as single isolated point mutations so far. To define the relative contribution of each of these two mutations to the Lewis negative phenotype, we cloned and made chimeric FUT3 constructs separating the T202C mutation responsible for the amino acid change Trp68 --> Arg, from the C314T mutation leading to the Thr105 --> Met shift. COS-7 cells were transfected and the expression of Fuc-TIII enzyme activity and the presence of Lewis antigens were determined. There was no decrease in enzyme activity nor of immunofluorescence staining on cells transfected with the construct containing the isolated C314T mutation compared with cells transfected with a wild type FUT3 allele control. No enzyme activity nor immunoreactivity for Lewis antigens was detected in FUT3 constructs containing both mutations in combination. The T202C mutation alone decreased the enzyme activity to less than 1% of the activity of the wild type FUT3 allele. These results demonstrate, that the Trp68 --> Arg substitution in human Fuc-TIII is the capital amino acid change responsible for the appearance of the Le(a-b-) phenotype on human erythrocytes in individuals homozygous for both the T202C and C314T mutations.  相似文献

BACKGROUND: Protein conformational modifications and water‐protein interactions are two major factors believed to induce instability of protein and eventually affect the solubility of milk protein concentrate (MPC) powder. To test these hypotheses, MPC was stored at different water activities (aw 0.0–0.85) and temperatures (25 and 45 °C) for up to 12 weeks. Samples were examined periodically to determine solubility, change in protein conformation by Fourier transform infrared (FTIR) spectroscopy and water status (interaction of water with the protein molecule/surface) by measuring the transverse relaxation time (T2) with proton nuclear magnetic resonance (1H NMR). RESULTS: The solubility of MPC decreased significantly with ageing and this process was enhanced by increasing water activity (aw) and temperature. Minor changes in protein secondary structure were observed with FTIR which indicated some degree of unfolding of protein molecules. The NMR T2 results indicated the presence of three distinct populations of water molecules and the proton signal intensity and T2 values of proton fractions varied with storage condition (humidity) and ageing. CONCLUSION: Results suggest that protein/protein interactions may be initiated by unfolding of protein molecules that eventually affects solubility. Results of the operation and evaluation of the grid-connected Coolidge solar thermal-electric power plant are reported. Performance was determined for each of the major subsystems—line-focus collector array, thermal energy storage and 200 kW organic fluid Rankine cycle engine and generator. Daily collector array efficiency (based on total direct solar radiation) was nominally 32% in June, 26% in September and 9% in December. Rankine cycle engine-generator conversion efficiency was about 20%; electrical parasitic losses reduced the efficiency by 12%. Operation and maintenance required about 90 h/mo, only 20% requiring special skills or training. Operating supplies and repair services cost about $6300 per year.  相似文献

The compatibility of superficially porous (SP) resin for label-free intact protein analysis with online capillary LC/MS is demonstrated to give improved chromatographic resolution, sensitivity, and reproducibility. The robustness of the platform was measured against several samples of varying complexity and sample loading amount. The results indicate that capillary SP columns provide high loading capacities and that ~6 s chromatographic peak widths are typical for standard proteins in simple mixtures and proteins isolated from cell and tissue lysates. Subfemtomole detection limits for standard proteins were consistently observed, with the lowest levels at 12 amol for ubiquitin. The analysis of total heart homogenates shows that capillary SP columns provide theoretical peak capacity of 106 protein forms with 30 min total analysis time and enabled detection of proteins from complex mixtures with a single high-resolution scan. The SPLC/MS platform also detected 343 protein forms from two HeLa acid extract replicate analyses that consumed 5 × 10(4) cells and 30 min analysis time, each. Comparison of all the species observed in each HeLa replicate showed 90% overlap (309 forms) with a Pearson correlation coefficient of 89.9% for the common forms observed in the replicates. Efficient acid extract of 1 × 10(4) HeLa cells allowed reproducible detection of common modification states and members from all five of the histone families and demonstrated that capillary SPLC/MS supports reproducible label-free profiling of histones in <15 min total analysis time. The data presented demonstrate that a capillary LC/MS platform utilizing superficially porous stationary phase and a LTQ-Orbitrap FT-MS is fast, sensitive, and reproducible for intact protein profiling from small tissue and cell amounts.  相似文献
