The genes Never ripe (Nr) and ripening inhibitor (rin) were introduced by backcross programmes into the tomato variety Ailsa Craig to give nearly isogenic lines. The effect of each of these genes on the major constituents of fully-developed tomato fruit has been studied as part of a programme to select breeding lines bearing fruit having prolonged shelf-life. The Nr gene produced high acid, low sugar, slow-ripening tomatoes that were very firm. Because of the adverse effect of this gene on tomato composition, even in heterozygous form, it has little or no commercial potential. The rin gene also led to the production of poor quality fruit. The heterozygote (Rin rin) gave fruit that were similar in compositon to that of the recurrent line, but despite possessing lower polygalacturonase activity were not significantly firmer. The distribution of isoenzymes in the normal and mutant tomato tissues was examined by gel electrophoresis. The majority of the enzymes investigated showed a diminished activity in Nr and rin fruit compared with normal. The very low activities of polygalacturonase, phosphofructokinase and NADP+-malic enzyme in the fully-developed mutant lines offers an explanation for the weak climacteric respiration rise and slow ripening of Nr fruit and the non-climacteric behaviour of rin fruit.  相似文献

The pollen beetle, Meligethes aeneus, which is an important pest of oilseed rape, Brassica napus, and turnip rape, B. rapa var. campestris, does not oviposit in all species of the Brassicaceae. The relationship between M. aeneus and candytuft, Iberis amara (Brassicacae), was investigated as part of chemical ecological studies into the development of control methods employing non-host-derived repellents. In choice and nonchoice feeding tests, M. aeneus completely rejected I. amara. However, in a field experiment using traps baited with flowering racemes of I. amara and B. napus, M. aeneus was attracted to both species. Gas chromatographic (GC) and GC-electroantennogram (GC-EAG) analyses indicated that the profiles of the floral volatiles of the two species are different. At least 12 compounds among the I. amara floral volatiles were detected by the M. aeneus antenna, and, of these, hexanoic acid, (E)-4,8-dimethyl-1,3,7-nonatriene and alpha-cedrene were not found among B. napus flower volatiles. Since M. aeneus is stimulated by floral volatiles to approach I. amara, but rejects it near, or at, the plant surface, I. amara does not produce repellents that could be used to manipulate M. aeneus. However, it may contain feeding deterrent(s) that could be used in "push-pull" control techniques or in the development of resistant brassicaceous crops.  相似文献

Polybrominated diphenyl ethers (PBDEs), polybrominated biphenyls (PBBs), and polychlorinated naphthalenes (PCNs) were quantified in blubber biopsy samples collected from free-ranging male and female killer whales (Orcinus orca) belonging to three distinct communities (southern residents, northern residents, and transients) from the northeastern Pacific Ocean. High concentrations of sigmaPBDE were observed in male southern residents (942+/-582 ng/g Iw), male and female transients (1015+/-605 and 885+/-706 ng/g Iw, respectively), and male and female northern residents (203+/-116 and 415+/-676 ng/g Iw, respectively). Because of large variation within sample groups, sigmaPBDE levels generally did not differ statistically with the exception of male northern residents, which had lower sigmaPBDE concentrations than male southern residents, male transients, and female transients, perhaps reflecting the consumption of less contaminated prey items. Male transient killer whales, which consume high trophic level prey including other cetaceans and occasionally spend time near populated areas, had sigmaPBDE concentrations approximately equal to southern residents. No significant age-related relationships were observed for sigmaPBDE concentrations. sigmaPBDE concentrations were approximately 1-3 orders of magnitude greater than those of sigmaPBB (3.0-31 ng/g Iw) and sigmaPCN (20-167 ng/g Iw) measured in a subset of samples, suggesting that PBDEs may represent a contaminant class of concern in these marine mammals.  相似文献

Volatile species of the conventional hydride-forming elements (As, Bi, Sb, Se, Sn, Pb, Cd, Te), Hg, transition metals (Ni, Co, Cu, Fe), noble metals (Ag, Au, Rh, Pd, Pt), and nonmetals (I, S) were generated following UV irradiation of their aqueous solutions to which low molecular weight carboxylic acids (formic, acetic, propionic) had been added. Free radicals arising from photodissociation of the latter provide a new and useful alternative to the common methods of chemical/electrochemical vapor generation techniques for the determination of these analytes by atomic spectrometry. Quantitative estimates of the efficiencies of these generation processes were not undertaken, although calculated signal-to-background ratios (>1500 for 5 ng/mL As, Sb, Bi, Se, and Te; 20 for 10 ng/mL Sn, Cu, Rh, Au, Pd, Pt, and Cd; 2400 for 1 ng/mL Hg; and 1000 for Co using ICP-TOF-MS detection) do provide clear evidence of the efficacy of this approach for sample introduction. In the case of Ni and Se, the tetracarbonyl and alkylated selenium compounds have been identified, respectively.  相似文献

Many studies have demonstrated that the exposure of nuclear medicine technologists arises primarily from radioactive patients rather than from preparation of radiopharmaceuticals. However, in order to devise strategies to reduce staff exposure, it is necessary to identify the specific tasks within each procedure that result in the highest radiation doses. An ESM Eberline FH41B-10 radiation dosemeter, which records the ambient dose equivalent rate, was used to monitor the radiation exposure of a technologist and to record the dose rate in microSv per hour every 32 s throughout a working day. The technologist recorded the procedures that were being performed so that the procedures that resulted in higher doses could be identified clearly. The measured doses clearly showed that the major contributions to the technologist's dose were the following: (1) transferring incapacitated patients from the imaging table to a hospital trolley; (2) difficult injections without syringe shields; and (3) setting up patients for gated myocardial scans. The average dose to the technologist from transferring patients after a bone scan was 0.54 microSv, 40% of the total dose of 1.3 microSv for the complete bone scan procedure. The average dose received injecting 900 MBq of 99Tcm-HDP using a tungsten syringe shield was 0.57microSv, but the highest dose was 1.6 microSv, in a patient in whom the injection was difficult. A 0.5 mm lead apron was found to reduce the dose when setting up a patient for a gated stress 99Tcm-sestamibi myocardial scan by approximately a factor of 2. The average dose per patient for this task was reduced from 1.1 to 0.6 microSv. It is recommended that staff waiting for assistance with patient transfers stand away from the patient, that tungsten syringe shields be used for all radiopharmaceutical injections and that a 0.5 mm lead apron be worn when attending patients containing high activities of 99Tcm radiopharmaceuticals, such as those having myocardial imaging.  相似文献

Argues that the characteristics of the obese behavior syndrome identified by S. Schachter (see record 1971-24450-001) are suited to the survival of the predator. Two types of hypothalamic ventral nuclei (HVN) are suggested: the predator-facilitating HVN and the vegetarian-facilitating HVN. A new vapor generation technique utilizing UV irradiation coupled with atomic absorption for the determination of selenium in aqueous solutions is described. In the presence of low molecular weight organic acid solutions, inorganic selenium(IV) is converted by UV irradiation to volatile selenium species, which are then rapidly transported to a heated quartz tube atomizer for detection by atomic absorption spectrometry. Optimum conditions for photochemical vapor generation and interferences from concomitant elements were investigated. Identification of the volatile products using cryotrapping GC/MS analysis revealed that inorganic selenium(IV) is converted to volatile selenium hydride, selenium carbonyl, dimethyl selenide, and diethyl selenide in the presence of formic, acetic, propionic, and malonic acids, respectively. In acetic acid solution, the efficiency of generation was estimated to be 50 +/- 10%. No interference from Ni(2+) and Co(2+) at concentrations of 500 and 100 mg L(-)(1), respectively, was evident. A detection limit of 2.5 microg L(-)(1) and a relative sensitivity of 1.2 microg L(-)(1) (1% absorption) with a precision of 1.2% (RSD, n = 11) at 50 microg L(-)(1) were obtained.  相似文献
