The cell-free rumen fluid from cattle fed hay or grain exhibited the following biological characteristics which strongly suggest the presence of endotoxin or a toxic principle similar to endotoxin of gram-negative bacteria: proved lethal to mice when injected with actinomycin D; proved extremely lethal to chick embryo; induced biphasic pyogenic response in rabbits; enhanced susceptibility to bacterial infection in mice; evoked positive epinephrine skin reaction in rabbits and phenol-water or aqueous ether proved lethal to mice and chick embryos. A quantitative difference in concentrations of endotoxin was observed on LD50 in mice and chick embryos and response to the epinephrine skin test in rabbits. Cell-free rumen fluid of grain-fed cattle contained at least twice as much endotoxin as that of hay-fed cattle. Endotoxin in cell-free rumen fluid and in higher concentration in cattle fed grain than in those fed hay support the hypothesis that rumen bacterial endotoxins may participate in the pathogenesis of diseases associated with high grain feeding such as lactic acidosis and the sudden-death syndrome.  相似文献

Experimental measurements support a theoretical argument suggesting that if the collector current of a b.j.t. is plotted against collector-base voltage, for set values of base-emitter drive voltage, then the tangents to the characteristics at a given collector-base voltage, in the forward-active region, have a common intersection point on the collector-base voltage axis. Primary cultures of fetal mouse brain and spinal cord were examined for the presence of binding sites for [3H]diazepam. Both brain and spinal cord cultures contain high affinity binding sites which resemble benzodiazepine receptors found in mammalian CNS with respect to both pharmacologic profile and response to exogenously applied GABA. These observations, coupled with the electrophysiologic properties of these cells suggest that primary cultures of fetal mouse brain and spinal cord may be valid models for studying the role and regulation of the benzodiazepine receptor.  相似文献

Numerous studies have identified depressive symptoms as antecedent to stressful experiences. However, the interrelations of depressive symptoms, personal resources, and coping as antecedents of stressful experiences are largely unknown. For this investigation, a prospective design was used to test a model estimating two categories of daily hassles at Time 2 in relation to depressive symptoms, self-esteem, gender, coping, and daily hassles at Time 1. Utilizing simultaneous equations, direct effects were observed for depressive symptoms, Time 1 hassles, gender, and avoidance coping in relation to category of hassles at Time 2. Several N-(phosphonomethyl) amides have previously been reported to act as particularly effective flame retardants for cellulose. It has also been demonstrated previously that the efficiency of a flame retardant on cellulose frequently parallels its ability to phosphorylate cellulosic hydroxyls at elevated temperatures. This study of the hydrolysis and alcoholysis reactions of N-(phosphonomethyl) amides establishes their unique reactivity. An explanation for this high reactivity is proposed on the basis of intramolecular assistance of the attack of water or alcohols at phosphorus by the carbonyl oxygen of the amide moiety.  相似文献

The breakdown of the Kohn-Luttinger ansatz for self-averaging of impurity scattering is shown to occur in small finite semiconductor device structures when less than 1000 impurities are in the device volume. As a consequence the standard non-equilibrium Green function formalism for quantum transport is invalid. Instead, a non-self-averaged propagator formalism is proposed based on a T-matrix model for non-asymptotic finite-cluster scattering. The formalism is illustrated by a study of the charge density and current density profiles resulting from coherent flow through a finite atomistic device containing a small number of impurities. The flow typically separates into open streamlines and a set of blocking vortices.  相似文献

Amounts of hepatotoxic microcystin and neurotoxic anatoxin-a were estimated in natural blooms and strains of cyanobacteria from freshwaters in Japan. A simultaneous analysis method of anatoxin-a and microcystin was applied to natural bloom samples, which has been dominated by several species and the strains of cyanobacteria which produced simultaneously both toxins. The natural blooms examined in the present study were mainly composed of Anabaena and Oscillatoria, but most also contained Microcystis and other cyanobacteria. Only one sample was almost unialgal, Anabaena spiroides, collected from Lake Sagami. The toxins in 14 samples collected from nine different natural blooms during 1988-1992 were identified as microcystins-RR, -YR, and -LR; desmethyl-7-microcystin-LR (7-DMLR); and anatoxin-a. Microcystins were the main toxins contained in these natural blooms, with anatoxin-a not being detected or of very little quantity. 7-DMLR was detected in samples only from Lake Kasumigaura. Five strains of Anabaena isolated from waters in Japan produced a small amount of anatoxin-a, but no microcystins. One half of the strains of Microcystis produced microcystins and/or anatoxin-a. This is the first study showing Microcystis producing both anatoxin-a and microcystins.  相似文献

Staphylococcus aureus is an important pathogen of humans and other animals, causing bacteremia, abscesses, endocarditis, and other infectious syndromes. A signature-tagged mutagenesis (STM) system was adapted for use in studying the genes required for in vivo survival of S. aureus. An STM library was ultimately created in S. aureus RN6390, with Tn917 being used to create the transposon mutations. Pools of S. aureus RN6390 mutants were screened in mouse abscess, bacteremia, and wound infection models for growth attenuation after in vivo passage. One of the mutants that was identified displayed marked attenuation following large-pool screening in all three animal models, which was confirmed in bacteremia and endocarditis models of infection with a smaller pool of mutants. Sequence analysis of the entire open reading frame showed a 99% identity to the high-affinity proline permease (putP) gene characterized in another strain of S. aureus. In wound and murine abscess infection models, the putP mutant was approximately 10-fold more attenuated than was wild-type strain RN6390. Another S. aureus strain transduced with the putP mutation also displayed an attenuated phenotype after passage in the wound model. A [3H]proline uptake assay showed that less proline was specifically transported into the putP mutant than into strain RN6390. The reduced viability of the bacteria possessing the mutation in the S. aureus high-affinity proline permease suggests that proline scavenging by the bacteria is important for in vivo growth and proliferation and that analogs of proline may serve as potential antistaphylococcal therapeutic agents.  相似文献
