The utility of myocardial imaging and assessment of regional myocardial metabolism of omega-(123I-paraphenyl-)pentadecanoic acid (I-PPA) by means of serial single-photon tomography is demonstrated in animal experiments. High quality cross sectional images of dog hearts with clear delineation of left ventricular walls are obtained. Myocardial infarcts are visualized as areas of deficient radioactivity uptake. I-PPA elimination from non-infarcted myocardial regions is significantly (p less than 0.001) prolonged when compared with unaffected controls. Hence, not only localized absence of uptake of free fatty acid by infarcted myocardium can be demonstrated with serial single-photon tomography but also general impairment of cardiac FFA-metabolism.  相似文献

A method of positive identification of the specificity of polymerase chain reaction (PCR) product using internal oligonucleotide probe is introduced. The hybridization was done on the agarose gel which was dried after electrophoresis. Detection of the expression of T cell receptor a chain variable (TCR V alpha) genes on mRNA level was used as the experimental model. Twenty nine TCR V alpha gene subfamilies could be distinguished clearly in healthy human peripheral blood lymphocytes by this method. Positive identification of PCR product on dried agarose gel by internal oligonucleotide probe is relatively simple and less time consuming.  相似文献

Muscle fibres isolated from pork were mounted in a microscope chamber and pH was controlled with 0.2 M phosphate buffer. Optical path differences between ordinary and extraordinary rays were measured by ellipsometry while a scanning stage moved the fibre across the optical axis of the microscope. The depth of fibre in the optical axis was estimated from the lateral diameter of fibres. Path differences increased as fibre depth increased. Static ellipsometry of unmounted fibre fragments showed maximum path differences at pH 5.5 were higher than at pH 7.0, but variance was high and the difference was not significant (64.4±15.1 nm at pH 5.5 versus 58.9±15.2 nm at pH 7.0, P>0.05, n=40). However, the mean depth of fibre fragments at pH 5.5 was less than at pH 7.0 (53.9±15.1 μm at pH 5.5 versus 70.6±16.1 μm at pH 7.0, P<0.005, n=40). Thus, path differences per micrometre of fibre depth were greater at pH 5.5 than at pH 7.0 (1.25±0.37 nm−1 μm at pH 5.5 versus 0.87±0.31 nm−1 μm at pH 7.0, P<0.0005, n=40). As pH decreases, therefore, muscle fibre diameter decreases while birefringence increases. Refraction through muscle fibres may contribute to pork paleness.  相似文献

Breast-feeding may expose infants to high levels of toxic chlorinated dioxins. To diminish intake of these lipophilic compounds by the baby, two diets were tested for their ability to reduce concentrations of dioxins in human milk. The diets were a low-fat/high- carbohydrate/low-dioxin diet. (about 20% of energy intake derived from fat) and a high fat /low-carbohydrate/low-dioxin diet. These diets were tested in 16 and 18 breast-feeding women, respectively. The test diets were followed for 5 consecutive days in the fourth week after delivery. Milk was sampled before and at the end of the dietary regimen, and dioxin concentrations and fatty acid concentrations were determined. Despite significant influences of these diets on the fatty acid profiles, no significant influence on the dioxin concentrations in breast milk could be found. We conclude that short-term dietary measures will not reduce dioxin concentration in human milk.  相似文献
