We tested the hypothesis that long-duration exercise (LDE) of moderate intensity, but not LDE of low intensity, during the daytime changes the typical temporal patterns of hormone release during subsequent nocturnal sleep. Ten trained healthy men participated in a balanced crossover study including three conditions: 1) no exercise, 2) LDE of low intensity (biking 40 km; 1800-2030), and 3) LDE of moderate intensity (biking 120-150 km; 1600-2030). During the subsequent night (2300-0700), somnopolygraphic sleep recordings were obtained, and concentrations of cortisol, growth hormone (GH), and testosterone were measured every 15 min. During the no exercise nights, the typical secretory patterns were present with peak concentrations of GH but nadir concentrations of cortisol during the first half of sleep but increased cortisol levels and minimum GH levels during the second part of sleep. Testosterone concentrations increased during the second half of sleep. LDE of moderate intensity reduced rapid-eye-movement sleep [13.9 vs. 16.9% (no exercise); P < 0.01]. Levels of testosterone decreased with increasing intensity of daytime exercise (P < 0.05). Moderate-, but not low-intensity, LDE decreased GH levels in the first half (P < 0.05) and increased GH levels in the second half (P < 0.005) of sleep. Also, LDE of moderate intensity but not LDE of low intensity increased cortisol levels during the first half (P < 0.005) and decreased cortisol secretion during the second half (P < 0.05) of sleep. Results suggest that nocturnal profiles of GH and cortisol concentrations may serve to indicate the disturbance of normal anabolic functions of sleep due to daytime exercise. 相似文献

DNA vaccination is an effective means of eliciting strong antibody responses to a number of viral antigens. However, DNA immunization alone has not generated persistent, high-titer antibody and neutralizing antibody responses to human immunodeficiency virus type 1 (HIV-1) envelope glycoprotein (Env). We have previously reported that DNA-primed anti-Env antibody responses can be augmented by boosting with Env-expressing recombinant vaccinia viruses. We report here that recombinant Env protein provides a more effective boost of DNA-initiated antibody responses. In rabbits primed with Env-expressing plasmids, protein boosting increased titer, persistence, neutralizing activity, and avidity of anti-Env responses. While titers increased rapidly after boosting, avidity and neutralizing activity matured more slowly over a 6-month period following protein boosting. DNA priming and protein immunization with HIV-1 HXB-2 Env elicited neutralizing antibody for T cell line-adapted, but not primary isolate, viruses. The most effective neutralizing antibody responses were observed after priming with plasmids which expressed noninfectious virus-like particles. In the budding yeast, Saccharomyces cerevisiae, four separate but structurally related mitogen-activated protein kinase (MAPK) activation pathways are known. The best understood of these regulates mating. Pheromone binding to receptor informs cells of the proximity of a mating partner and induces differentiation to a mating competent state. The MAPK activation cascade mediating this signal is made up of Ste11 (a MEK kinase [MEKK]), Ste7 (a MAPK/ERK kinase [MEK]), and the redundant MAPK-related Fus3 and Kss1 enzymes. Another MAPK activation pathway is important for cell integrity and regulates cell wall construction. This cascade consists of Bck1 (a MEKK), the redundant Mkk1 and Mkk2 enzymes (MEKs), and Mpk1 (a MAPK). We exploited these two pathways to learn about the coordination and signal transmission fidelity of MAPK activation cascades. Two lines of evidence suggest that the activities of the mating and cell integrity pathways are coordinated during mating differentiation. First, cells deficient in Mpk1 are susceptible to lysis when they make a mating projection in response to pheromone. Second, Mpk1 activation during pheromone induction coincides with projection formation. The mechanism underlying this coordination is still unknown to us. Our working model is that projection formation generates a mobile second messenger for activation of the cell integrity pathway. Analysis of a STE7 mutation gave us some unanticipated but important insights into parameters important for fidelity of signal transmission. The Ste7 variant has a serine to proline substitution at position 368. Ste7-P368 has higher basal activity than the wild-type enzyme but still requires Ste11 for its function. Additionally, the proline substitution enables the variant to transmit the signal from mammalian Raf expressed in yeast. This novel activity suggests that Ste7-P368 is inherently more permissive than Ste7 in its interactions with MEKKs. Yet, Ste7-P368 cross function in the cell integrity pathway occurs only when it is highly overproduced or when Ste5 is missing. This behavior suggests that Ste5, which has been proposed to be a tether for the kinases in the mating pathway, contributes to Ste7 specificity and fidelity of signal transmission. 相似文献

The use of multiple sequence alignments for secondary structurepredictions is analysed. Seven different protein families, containingonly sequences of known structure, were considered to providea range of alignment and prediction conditions. Using alignmentsobtained by spatial superposition of main chain atoms in knowntertiary protein structures allowed a mean of 8% in secondarystructure prediction accuracy, when compared to those obtainedfrom the individual sequences. Substitution of these alignmentsby those determined directly from an automated sequence alignmentalgorithm showed variations in the prediction accuracy whichcorrelated with the quality of the multiple alignments and distanceof the primary sequence. Secondary structure predictions canbe reliably improved using alignments from an automatic alignmentprocedure with a mean increase of 6.87percnt;, giving an overallprediction accuracy of 68.5%, if there is a minimum of 25% sequenceidentity between all sequences in a family. 相似文献

Preemptive scheduling problems on parallel machines are classic problems. Given the goal of minimizing the makespan, they are polynomially solvable even for the most general model of unrelated machines. In these problems, a set of jobs is to be assigned to run on a set of m machines. A job can be split into parts arbitrarily and these parts are to be assigned to time slots on the machines without parallelism, that is, for every job, at most one of its parts can be processed at each time. Motivated by sensitivity analysis and online algorithms, we investigate the problem of designing robust algorithms for constructing preemptive schedules. Robust algorithms receive one piece of input at a time. They may change a small portion of the solution as an additional part of the input is revealed. The capacity of change is based on the size of the new piece of input. For scheduling problems, the supremum ratio between the total size of the jobs (or parts of jobs) which may be re-scheduled upon the arrival of a new job j, and the size of j, is called migration factor. We design a strongly optimal algorithm with the migration factor $1-\frac{1}{m}$ for identical machines. Strongly optimal algorithms avoid idle time and create solutions where the (non-increasingly) sorted vector of completion times of the machines is lexicographically minimal. In the case of identical machines this results not only in makespan minimization, but the created solution is also optimal with respect to any ? p norm (for p>1). We show that an algorithm of a smaller migration factor cannot be optimal with respect to makespan or any other ? p norm, thus the result is best possible in this sense as well. We further show that neither uniformly related machines nor identical machines with restricted assignment admit an optimal algorithm with a constant migration factor. This lower bound holds both for makespan minimization and for any ? p norm. Finally, we analyze the case of two machines and show that in this case it is still possible to maintain an optimal schedule with a small migration factor in the cases of two uniformly related machines and two identical machines with restricted assignment. 相似文献

We consider detonation wave propagation in an inhomogeneous fuel gas mixture at rest, whose initial state is characterized by values of pressure and density that are in the general case, functions of the spatial and time coordinates. The distance form the plane, axis or center of symmetry can be used as the spatial coordinate. The sources of mass, pulse, and energy are assumed to be present behind the detonation front. The appropriate necessary conditions for the detonation wave to propagate in the Chapman-Jouguet regime are determined. A novel membrane effective in the separation of ethanol-water mixtures by pervaporation was made by combining zeolite NaA and poly(vinyl chloride) modified by 2-(2′-butoxyethoxy)ethyl thiolate. Under ambient conditions, a separation factor (α) of 29 and pervaporability (P) of 4 × 10?4 g m?1 h?1 were obtained for the azeotropic mixture whereas, in the absence of zeolite, the respective values were 7 and 7 × 10?5. Direct initiation of detonation by explosion of a TNT charge in a hydrogen—air mixture is considered. The critical mass of initiating charge is determined numerically using a finite-difference method based on the Godunov scheme, taking into account the real chemical kinetics of combustion of hydrogen in air and the real equation of state for gaseous detonation products of TNT. We investigated the explosion of a cylindrical charge, which lies on a rigid surface or is positioned at some height above it. The explosion is modeled by the jump between the background and the region occupied by gas at a high pressure, density, and temperature. In an analogous formulation, the explosion from the Tunguska meteorite impact [1–4] was modeled by spherical and cylindrical explosive waves which consider the gravitational back pressure.
