Gelatinized wheat, potato and waxy maize starches were treated enzymatically in order to increase the degree of branching of the amylopectin fraction and thereby change the starch degradation profile towards a higher proportion of slowly digestible starch (SDS). The materials were characterized by single-pulse (1)H HR-MAS NMR spectroscopy and in vitro digestion profile according to the Englyst procedure. Using various concentrations and incubation times with branching enzyme (EC 2.4.1.18) without or with additional treatment with the hydrolytic enzymes; β-amylase (EC 3.2.1.2), α-glucosidase (EC 3.2.1.20), or amyloglucosidase (EC 3.2.1.3) the proportion of α-(1-6) linkages was increased by up to a factor of 4.1, 5 and 5.8 in waxy maize, wheat and potato starches, respectively. The proportion of SDS was significantly increased when using hydrolytic enzymes after treatment with branching enzyme but it was only for waxy maize that the proportion of α-(1-6) bonds and the in vitro digestion profile was significantly correlated.  相似文献

Premilking udder preparation (including forestripping and duration of lag time-the time between first tactile stimulation and attachment of milking unit) might influence milking measures such as milking unit on-time, incidence of bimodality, and milk flow rates in Holstein cows milked 3 times daily. Holstein cows (n=786) from an 1,800-cow commercial dairy herd were enrolled under a restricted randomized design to determine the effect of 9 different premilking routines. Lag times were 0, 60, 90, 120, and 240s and included forestripping or no forestripping for a total of 9 treatments (no forestripping for 0 lag time); the study was conducted from February to November 2008. All cow-treatment combinations were compared with the control: predipping plus forestripping and drying with 90s of lag time. Cows were initially assigned to 1 of 3 treatments for a period of 7d and upon completion of the first 7-d period were reassigned to a different treatment until all treatments had been completed. From one treatment period to the next, cows had to switch stimulation method with no restriction on lag time. Cows did not receive all treatments during the duration of the trial. Early- to mid-lactation cows (EML; 17-167 DIM) and late-lactation cows (LL; 174-428 DIM) were housed in 2 different pens. Milk yield was significantly different between dip + forestrip and dip+dry for 2 of the treatments for EML cows compared with dip + forestrip and 90 s of lag-time (DF90); however, this was not thought to be due to treatment because the significant lag times were very different (60 and 240 s) and neither was an extreme value. Milk yield did not differ with treatment for the LL cows. Milking unit on-time did not differ when comparing all treatments for EML with treatment DF90; however, an increase in milking unit on-time occurred when lag time was 60s or less for LL cows. The highest incidence of bimodal milk curves was when lag time = 0 and this was independent of stage of lactation; a lag time of 240 s had the second-highest incidence of bimodal milk curves for EML and LL cows. Milk harvested in the first 2 min was lower for lag times of 0 and 240 s when compared with DF90. Increasing the lag time for all cows appeared to improve overall milking time efficiency (although lag time had no effect on EML cows).  相似文献

Asthma and many autoimmune diseases, such as systemic lupus erythematosus, have been reported to associate with vitamin D deficiency recently. Growth-related oncogene-α (GRO-α)/CXCL1, a neutrophil-related chemokine, have an important influence on the chronic inflammation of these diseases. It is unknown whether vitamin D has regulatory effects on GRO-α expression in human monocytes. To this end, the human monocytic leukemia cell line, THP-1, and human primary monocytes were pretreated with 1α, 25-(OH)(2)D(3), and was stimulated with lipopolysaccharide (LPS). Supernatants were collected to determine GRO-α level by ELISA. The intracellular signaling was investigated by nuclear factor (NF)-κB inhibitor, the mitogen-activated protein kinase (MAPK) inhibitors, and Western blot. In our studies, LPS-induced GRO-α was significantly enhanced in THP-1 cells, but suppressed in human primary monocytes by 1α, 25-(OH)(2)D(3). Western blotting revealed that 1α, 25-(OH)(2)D(3) increased LPS-stimulated pp38 expression in THP-1 cells, but suppressed LPS-stimulated pMEK1/2-pERK and pJNK in human primary monocytes. In conclusion, the opposite effects of 1α, 25-(OH)(2)D(3) on GRO-α expression in THP-1 cells and human primary monocytes indicated that the data from THP-1 cells should be further confirmed by human primary monocytes. Moreover, vitamin D3 may have potentiality in treating GRO-α-related chronic inflammatory diseases, like asthma and autoimmune diseases.  相似文献

A series of compounds based on the mGluR5-selective ligand 2-methyl-6-(phenylethynyl)pyridine (MPEP) were designed and synthesized. The compounds were found to be either structural analogues of MPEP, substituted monomers, or dimeric analogues. All compounds retained mGluR5 selectivity with only weak or no activity at other mGluRs or iGluRs. The substituted analogue, 1,3-bis(pyridin-2-ylethynyl)benzene (19), is a potent negative modulator at mGluR5, whereas all other compounds lost potency relative to MPEP and showed that activity is highly dependent on the position of the nitrogen atom in the pyridine moieties. A homology modeling and ligand docking study was used to understand the binding mode and the observed selectivity of compound 19.  相似文献

The objective of the research was to estimate genetic parameters, such as heritabilities and genetic correlations, using daily test day data for milk yield (MY), milking speed (MS), dry matter intake (DMI), and body weight (BW) using random regression methodology. Data were from first lactation dairy cows (n = 320) from the Chamau research farm of the Swiss Federal Institute of Technology, Switzerland over the period from April 1994 to 2004. All traits were recorded daily using automated machines. Estimated heritabilities (h2) varied from 0.18 to 0.30 (mean h2 = 0.24) for MY, 0.003 to 0.098 (mean h2 = 0.03) for MS, 0.22 to 0.53 (mean h2 = 0.43) for BW, and 0.12 to 0.34 (mean h2 = 0.23) for DMI. A permanent environmental effect was included in both the univariate and bivariate models, but was assumed constant in estimating some genetic correlations because of convergence problems. Estimated genetic correlations varied from 0.31 to 0.41 between MY and MS, from −0.47 to 0.29 between MY and DMI, from −0.60 to 0.54 between MY and BW, from 0.17 to 0.26 between MS and DMI, from −0.18 to 0.25 between MS and BW, and from −0.89 to 0.29 between DMI and BW. Genetic correlations for MY, MS, DMI, and BW from calving to midlactation decreased similarly to 0.40, 0.36, 0.14, and 0.36 and, at the end of the lactation, decreased to −0.06, 0.23, −0.07, and 0.09, respectively. Daily genetic variance-covariance of many functional traits are reported for the first time and will be useful when constructing selection indexes for more than one trait based on longitudinal genetic parameters.  相似文献

Drug release characteristics of freeze-dried wafers and solvent-cast films prepared from sodium carboxymethylcellulose have been investigated and compared. In vitro drug dissolution studies were performed using an exchange cell and drug release was measured by UV spectroscopy at 272?nm using distilled water. The dissolution profiles of hydrochlorothiazide from the wafers and films were compared by determining the rates of drug release, estimated from the % release versus time profiles and calculating their difference (f(1)) and similarity (f(2)) factors. The effects of drug loading, polymer content and amount of glycerol (GLY) (films) on the drug release characteristics of both formulations were investigated. Both the wafers and films showed sustained type release profiles that were best explained by the Korsmeyer-Peppas equation. Changes in the concentration of drug and GLY (films) did not significantly alter the release profiles whilst increasing polymer content significantly decreased the rate of drug release from both formulations. The rate of release was faster from the wafers than the corresponding films which could be attributed to differences in the physical microstructure. The results show the potential of employing both formulations in various mucosal drug delivery applications.  相似文献

We present the fabrication and analysis of efficient and highly dispersive gratings for the x-ray and extreme ultraviolet (EUV) regime. We show that an asymmetric-cut multilayer structure can act as a near-perfect blazed grating. The precision and high line density are achieved by layer deposition of materials, which can be controlled to the angstrom level. We demonstrate this in the EUV regime with two structures made by cutting and polishing magnetron-sputtered multilayer mirrors of over 2000 bilayers thick, each with a period of 6.88 nm. These were cut at angles of 2.9° and 7.8° to the surface. Within the 3% bandwidth rocking curve of the multilayer, the angular dispersion of the diffracted wave was in agreement with the grating equation for elements with 7250 and 19,700 line pairs/mm, respectively. The dependence of the measured efficiency was in excellent agreement with a formulation of dynamical diffraction theory for multilayered structures. At a wavelength of 13.2 nm, the efficiency of the first-order diffraction was over 95% of the reflectivity of the uncut multilayer. We predict that such structures should also be effective at shorter x-ray wavelengths. Both the Laue (transmitting) and Bragg (reflecting) geometries are incorporated in our formalism, which is applied to the analysis of multilayer Laue lenses and focusing and dispersing Bragg optics.  相似文献

Methyltrioxorhenium (MTO) and its derivatives were utilized as catalysts for a broad variety of organic reactions after the discovery of an easy synthetic access way to this class of compounds. Among the numerous characterization techniques, 17O-NMR spectroscopy and vibrational spectroscopy were exemplary discussed.
