We show that generic viewpoint and lighting assumptions resolve standard visual ambiguities by biasing toward planar surfaces. Our model uses orthographic projection with a two-dimensional affine warp and Lambertian reflectance functions, including cast and attached shadows. We use uniform priors on nuisance variables such as viewpoint direction and the light source. Limitations of using uniform priors on nuisance variables are discussed. 相似文献

Using the pH-jump (neutral to above 11) for Tyr-OH ionization as a probe, two glucoamylases from Rhizopus niveus and Rhizopus delemar were found to be caused the change in conformation, which is in a kinetically single step. The pH titration at pHs below 12 was carried out on a multidimensional correlation spectrophotometer. Correlations among the spectrophotometric properties: fluorescence, absorption and circular dichroism are almost identical for the two enzymes. These results suggest that conformational characteristics of the niveus enzyme is almost the same as that of the delemar one. 相似文献

Based on the subsite structure of an α-amylase from Bacillus subtilis, production of glucose from a substrate maltohexaose in the amylase-catalyzed reaction was theoretically predicted as a function of time. The theoretical time course was found consistent with the experimental results, which were obtained by the determination of a product glucose with a commercial assay-kit. Hence, glucose, product from a substrate maltohexaose, will be a useful probe for the amylase-catalyzed reaction. 相似文献

In silico comparison of 34 putative pks genes in Aspergillus niger strain CBS 513.88 versus A. niger strain ATCC 1015 genome revealed significant nucleotide identity (>95% covering a minimum of 99% of the gene sequence) for 31 of these genes (approximately 91%). A. niger CBS 513.88 harbors three putative pks genes (An01g01130, An11g05940, and An15g07920), for which nucleotide identity was not found in A. niger ATCC 1015. To compare the results of the in silico analysis with the in vivo situation, experimental data were obtained for a large number of A. niger strains obtained from different substrates and geographical regions. Three putative pks genes that were found to be variable between the two A. niger strains using bioinformatics tools were in fact strain-specific genes based on experimental data. The PCR amplification signals for the An01g01130, An11g05940, and An15g07920 pks genes were detected in only 97%, 71%, and 26% of the strains, respectively. Southern blot analyses confirmed the PCR data. Because one of the strain-specific pks genes (An15g07920) is located in a putative ochratoxin cluster, we focused our investigation on that region. We assessed the ochratoxin production capability of the 119 A. niger strains and found a positive association between the presence of this pks gene and the capability of the respective strain to produce ochratoxin. 相似文献

A simple method was developed for the analysis of hyoscyamine and scopolamine in human serum and urine using liquid chromatography with tandem mass spectrometry (LC/MS/MS). Hyoscyamine and scopolamine in serum and urine were cleaned up with an Oasis HLB cartridge and a PSA cartridge. The LC separation was carried out on an ODS column, using linear gradient elution with 5 mmol/L IPCC-MS3-methanol as the mobile phase. The mass spectral acquisition was done in the positive ion mode by applying selected reaction monitoring (SRM). The recoveries of hyoscyamine and scopolamine were 86.0-105% from human serum and urine fortified at 0.2 ng/mL and 10 ng/mL. The detection limits of hyoscyamine and scopolamine were 0.02 ng/mL. Four serum and three urine samples of humans poisoned by eating Datura innoxia Mill. were analyzed by this method. Hyoscyamine and scopolamine were detected at the levels of 0.45-3.5 ng/mL in all serum samples and 170-670 ng/mL in all urine samples. 相似文献

Based on fungal and fumonisin contamination of 870 freshly harvested samples, the quality of corn used by processing industries in the Northern region of Paraná State, Brazil (2003 and 2004 crop-year) was evaluated. Sampling was carried out for each crop at two points in the production chain, i.e. at reception by the processors and at the pre-drying step. Corn samples were more frequently contaminated with Fusarium sp. (100%) and Penicillium sp. (84.1–95.3%) than Aspergillus sp. (5.6–19.8%). Fumonisin B1 (FB1) was detected in all samples from the two points in both crop-years. FB1 levels ranged 0.02–11.83 µg g?1 in the reception and 0.02–10.98 µg g?1 in the pre-drying samples of the 2003 crop. Samples from the 2004 crop showed FB1 levels ranging 0.03–12.04 µg g?1 in the reception and 0.06–7.74 µg g?1 in the pre-drying samples. FB2 levels ranged 0.02–5.25 µg g?1 in the reception and 0.01–7.89 µg g?1 in the pre-drying samples (2003 crop-year). In samples from the 2004 crop, FB2 levels ranged 0.02–6.12 µg g?1 in the reception and 0.05–3.47 µg g?1 in the pre-drying samples. Low fumonisin levels were detected in most corn samples used by processors in the Northern region of Paraná State, showing a decreasing trend in fumonisin contamination over the years. 相似文献

The influence of platelet-type clay nanoparticle (nanoplatelet) on the structural evolution in injection-molded nylon 6)/carbon composites was investigated. In the absence of nanoplatelets, the nylon 6/CB systems were found to exhibit unoriented structure with nylon 6 crystalline regions exhibiting exclusively α crystal form throughout the thickness of the samples. However, inclusion of nanoplatelets induces substantial local orientation of the nylon 6 chains in the molded parts in all processing conditions and compositions. In these clay/nylon 6/CB ternary nano systems, nylon 6 matrix was found to be exclusively in γ crystal form at the skin regions and α crystal form fraction increases towards the core of the molded parts as a result of decrease in cooling rate with depth during the solidification stage. Two nanoplatelet orientation behaviors were identified: (i) in the absence of irregular shaped CB, the nanoplatelets align parallel with one another following the local flow patterns. The latter behavior also causes enhanced orientation of the nylon 6 chains undergoing substantial shear amplification trapped in between the nanoplatelets (ii) in the CB-enriched regions, nanoplatelets though still remaining parallel to one another, are randomized by following the local contours of irregular shaped CB aggregates. The WF (wall failure) test of the EAGLE program, in which 2 kg of uranium dioxide fuel-pins were melted by nuclear heating, was successfully conducted in the IGR (Impulse Graphite Reactor) of NNC/Kazakhstan. In this test, a 3 mm-thick stainless steel (SS) wall structure was placed between fuel pins and a 10 mm-thick sodium-filled channel (sodium gap). During the transient, fuel pins were heated, which led to the formation of a fuel-steel mixture pool. Under the transient nuclear heating condition, the SS wall was strongly heated by the molten pool, leading to wall failure. The time needed for fuel penetration into the sodium-filled gap was very short (less than 1 s after the pool formation). The result suggests that molten core materials formed in hypothetical LMFBR core disruptive accidents have a certain potential to destroy SS-wall boundaries early in the accident phase, thereby providing fuel escape paths from the core region. The early establishment of such fuel escape paths is regarded as a favorable characteristic in eliminating the possibility of severe re-criticality events. Ammonia generation was studied in the reaction between water and nitrogen-containing iron at 323 K and atmospheric pressure. Similar to metallic Fe, the interstitial compound Fe3N reduced water through Fe oxidation to produce hydrogen gas, while the N combined with atomic hydrogen to produce ammonia as a byproduct. The addition of carbon dioxide to this system accelerated the reaction with concomitant consumption of carbon dioxide. The promoted ammonia production upon addition of carbon dioxide can be attributed to the generation of atomic hydrogen from the redox reaction of carbonic acid and Fe, as well as removal of used Fe from the reaction system through the formation of a soluble carbonato complex. When carbonate was added to the reaction system, the production rates of ammonia and hydrogen increased further. The results here confirmed that ammonia can be synthesized from iron nitride under mild conditions by utilizing carbon dioxide. 相似文献
