Electromuscular incapacitating devices (EMDs), such as Tasers, deliver high current, short duration pulses that cause muscular contractions and temporarily incapacitate the human subject. Some reports suggest that EMDs can kill. To help answer the question, "Can the EMD directly cause ventricular fibrillation (VF)?", ten tests were conducted to measure the dart-to-heart distance that causes VF in anesthetized pigs [mass = 64 kg +/- 6.67 standard deviation (SD)] for the most common X26 Taser. The dart-to-heart distance that caused VF was 17 mm +/- 6.48 (SD) for the first VF event and 13.7 mm +/- 6.79 (SD) for the average of the successive VF events. The result shows that when the stimulation dart is close enough to the heart, X26 Taser current will directly trigger VF in pigs. Echocardiography of erect humans shows skin-to-heart distances from 10 to 57 mm (dart-to-heart distances of 1-48 mm). These results suggest that the probability of a dart on the body landing in 1 cm2 over the ventricle and causing VF is 0.000172.  相似文献

Wwox-deficient human cells show elevated homologous recombination, leading to resistance to killing by double-strand break-inducing agents. Human Wwox binds to the Brca1 981-PPLF-984 Wwox-binding motif, likely blocking the pChk2 phosphorylation site at Brca1-S988. This phosphorylation site is conserved across mammalian species; the PPLF motif is conserved in primates but not in rodents. We now show that murine Wwox does not bind Brca1 near the conserved mouse Brca1 phospho-S971 site, leaving it open for Chk2 phosphorylation and Brca1 activation. Instead, murine Wwox binds to Brca1 through its BRCT domain, where pAbraxas, pBrip1, and pCtIP, of the A, B, and C binding complexes, interact to regulate double-strand break repair pathway response. In Wwox-deficient mouse cells, the Brca1-BRCT domain is thus accessible for immediate binding of these phospho-proteins. We confirm elevated homologous recombination in Wwox-silenced murine cells, as in human cells. Wwox-deficient murine cells showed increased ionizing radiation-induced Abraxas, Brca1, and CtIP foci and long resected single-strand DNA, early after ionizing radiation. Wwox deletion increased the basal level of Brca1-CtIP interaction and the expression level of the MRN-CtIP protein complex, key players in end-resection, and facilitated Brca1 release from foci. Inhibition of phospho-Chk2 phosphorylation of Brca1-S971 delays the end-resection; the delay of premature end-resection by combining Chk2 inhibition with ionizing radiation or carboplatin treatment restored ionizing radiation and platinum sensitivity in Wwox-deficient murine cells, as in human cells, supporting the use of murine in vitro and in vivo models in preclinical cancer treatment research.  相似文献

The efficacy of NovaSil clay (NS) to reduce aflatoxin (AF) biomarkers of exposure was evaluated in 656 blood samples and 624 urine samples collected from study participants during a 3-month phase IIa clinical intervention trial in Ghana. NS was delivered before meals via capsules. Serum AFB (1)-albumin adduct was measured by radioimmunoassay and urinary AFM (1) metabolites were quantified by immunoaffinity-high-performance liquid chromatography (HPLC)-fluorescence methods. Levels of AFB (1) -albumin adduct in serum samples collected at baseline and at 1 month were similar (p = 0.2354 and p = 0.3645, respectively) among the placebo (PL), low dose (LD, 1.5 g NS day (-1)), and high dose (HD, 3.0 g NS day (-1)) groups. However, the levels of AFB (1)-albumin adduct at 3 months were significantly decreased in both the LD group (p < 0.0001) and the HD group (p < 0.0001) compared with levels in the PL group. Levels of AFM(1) in urine samples collected at baseline and at 1 month were not statistically different among the three study groups. However, a significant decrease (up to 58%) in the median level of AFM (1) in samples collected at 3 months was found in the HD group when compared with the median level in the PL group (p < 0.0391). In addition, significant effects were found for dose, time, and dose-time interaction with serum AFB(1)-albumin adduct and dose-time interaction with urinary AFM (1) metabolites. The results suggest that capsules containing NS clay can be used to reduce effectively the bioavailability of dietary AF based on a reduction of AF-specific biomarkers.  相似文献

Flashover voltage, lifetimes, and switch performance of insulators utilizing square thin and thick film electrodes were examined to determine the viability of using thin electrodes for reliable surface discharge switching. Gold, silver, and platinum were sputtered (0.25 /spl mu/m) and screen printed (15 /spl mu/m) onto Al/sub 2/O/sub 3/, TiO/sub 2/, and modified BaTiO/sub 3/ (MBT), then tested in air at 10/sup 5/ Pa, under vacuum (10/sup -3/ torr), and while immersed in an insulating fluid, SF-2 (manufactured by 3M). For the measured range of 0.5 to 3 mm in air, the flashover voltage for all three insulators was found to have a linear dependence on the electrode separation distance with 15 /spl mu/m thick screen printed electrodes and a square root dependence with 0.25 /spl mu/m thick sputtered electrodes. Delay times of approximately 20 ns with a corresponding jitter of 6 ns were observed across all three insulators under triggered flashover. Insulators in air with sputtered electrodes had lifetimes of approximately 5 flashovers for dc flashover and 40 for triggered flashover. Screen printed TiO/sub 2/ and MBT had dc lifetimes of approximately 10 flashovers in air, and 3 flashovers in vacuum and SF-2. Screen printed TiO/sub 2/ and MBT had triggered lifetimes of greater than 200 flashovers in air, and <3 flashovers in vacuum and SF-2. Screen printed Al/sub 2/O/sub 3/ had dc and triggered lifetimes of greater than 200 flashovers in air, vacuum and SF-2. Insulator failure during dc flashover was determined to be due to the formation of a conductive channel between the anode and cathode. Formation of the channel was attributed to insulator thermal and dielectric properties and the presence of vaporized electrode species in the gap region during flashover.  相似文献

Before the developmental trajectory, outcomes, and related interventions of gratitude can be accurately and confidently studied among the youth, researchers must ensure that they have psychometrically sound measures of gratitude that are suitable for this population. Thus, considering that no known scales were specifically designed to measure gratitude in youth, this study aimed to answer an important question: Are the existing gratitude scales used with adults valid for use with youth? The present study is an empirical investigation, based on a large youth sample (N = 1,405) with ages ranging from 10 to 19 years old, of the psychometric properties of scores of the Gratitude Questionnaire-6 (GQ-6; M. E. McCullough, R. A. Emmons, & J.-A. Tsang, 2002), the Gratitude Adjective Checklist (GAC; M. E. McCullough, R. A. Emmons, & J.-A. Tsang, 2002), and the Gratitude Resentment and Appreciation Test (GRAT)-short form (M. Thomas & P. Watkins, 2003). Single-group and multiple-group confirmatory factor analyses indicated that the factor structures of these gratitude scales resemble those found with adults and were invariant across age groups. Scores of all three gratitude scales revealed acceptable internal consistency estimates (i.e., >.70) across age groups. Results showed that whereas scores of all three gratitude scales were positively correlated with each other for 14- to 19-year-olds, GRAT-short form scores tended to display relatively low correlations with scores of the other two measures for younger children (10–13 years old). Furthermore, the nomological network analysis showed that scores of all three gratitude scales were positively correlated with positive affect and life satisfaction scores across the age groups. The relationships with negative affect and depression scores, however, seemed dependent on the child's age. As regulated energy consumption in buildings is reduced, the proportional importance of unregulated energy consumption increases. Reducing unregulated energy use in the commercial office requires an understanding of the factors that influence workplace behaviour. To date these factors have been assumed to be similar to those that influence behaviour in the home. However, the social dynamics of the workplace are different to those in the home. This study examines the degree to which theories of behaviour change generated largely in a domestic building setting could be used as the basis for designing interventions to reduce unregulated energy consumption in the workplace. It studies the unregulated energy consumption of 39 workers engaged in office-type activities in two separate locations. Following a 100-day monitoring period, three behaviour change interventions were developed and their impact measured over a 100-day period. Results from the study found, on average, an 18.8% reduction in energy use was achieved. Furthermore, by comparing pre- and post-intervention responses to an environmental questionnaire, it was evident that savings were realized without significant changes to pro-environmental attitude or perceived social norms, which may have implications for energy-saving interventions in the commercial sector.  相似文献

OBJECTIVES: To determine the impact of HIV infection on Streptococcus pneumoniae bacteraemia in adults and children by analysing the prevalence and clinical features of such diseases and determining the prevalent serotypes/serogroups and susceptibility patterns of isolates. DESIGN: Patients were identified prospectively from January to October 1996. SETTING: Chris Hani Baragwanath Hospital, Soweto, a tertiary referral hospital treating adults and children, in an urban district near Johannesburg, South Africa. PATIENTS AND METHODS: All patients with S. pneumoniae isolated from blood culture by the Microbiology Department, Chris Hani Baragwanath Hospital were studied. Clinical and microbiological features were recorded. RESULTS: A total of 178 patients with S. pneumoniae were investigated as part of the study; 49 were aged < 13 years. HIV seroinfection was present in 25 (51%) children and 58 (45%) adults. The incidence of S. pneumoniae bacteraemia was 36.9-fold increased in HIV-seropositive children and 8.2-fold increased in HIV-seropositive adults compared with HIV-seronegative individuals. Both adult and paediatric HIV-seropositive patients with S. pneumoniae bacteraemia were significantly younger than HIV-seronegative patients. Pneumonia was a significantly more common presentation in HIV-seropositive children, otherwise the spectrum of disease and outcome were similar in HIV-seronegative and positive groups. Serotype 1 S. pneumoniae isolates were significantly less common in HIV-infected individuals (both adults and children). Resistance to penicillin was increased in S. pneumoniae isolates from HIV-infected patients (significant in adults). Patients with penicillin-resistant isolates did not have a poorer outcome. The potential coverage of serotypes/serogroups included in the proposed nine-valent conjugate pneumococcal vaccine was 88% in HIV-seronegative children and 83% in HIV-seropositive children. The potential coverage of the currently available 23-valent pneumococcal vaccine for adults was 98.2 and 100)% for HIV-infected and HIV-uninfected adults, respectively. CONCLUSION: The burden of bacteraemia due to S. pneumoniae in HIV-seropositive individuals admitted to our hospital is considerable. Differences in the S. pneumoniae serotypes/serogroups in HIV-infected patients have been demonstrated with resultant differences in antibiotic susceptibility patterns. Excellent potential for vaccine coverage was demonstrated for both HIV-seronegative and HIV-seropositive individuals. Further studies are necessary to test the clinical efficacy of pneumococcal vaccination of HIV-seropositive adults and children as a potential preventative measure against this prevalent disease.  相似文献

Novel attenuation effects and the generation of the third harmonic frequency in nonlinear zero and first sound in 3 He were found in pulsed sound-transmission experiments at various pressures (0, 10 bar), frequencies (10-50 MHz), sound-pulse energies (0.01-110 nJ) and temperatures ranging from 0.8 mK to 200 mK. The nonlinear damping of zero sound increases with the square of the frequency and demonstrates that quantum-damping exists in a Fermi liquid even for low frequency phonons at temperatures as high as 1 mK due to multiple phonon absorption processes. The origin of the additional damping found in first sound at high temperatures and sound pulse powers is unclear, but part of the apparent attenuation is caused by third harmonic generation which was also found in superfluid 3 He-B. In zero- as well as in first sound the third harmonic generation takes place in the leading edge of the propagating sound pulse at times when the fundamental frequency has not even developed to its full amplitude.  相似文献
