Male Wistar rats received a combination of 25 ppm PbCl2 and 5 ppm CdCl2 in drinking water at different developmental stages. Pb and Cd levels of incisors, molars, epiphyses, diaphyses and kidney cortex were recorded by atomic absorption spectrophotometry. The results confirmed that Pb has an affinity for hard tissue and especially to teeth, but it also accumulated in soft tissues under the above conditions. In addition, the results indicated none or only a moderate placental or mammary barrier for Pb, and suggested a high absorption of lead from the intestine of sucklings. The results also confirmed that kidney cortex is a primary target for Cd, and indicated that Cd has crossed the placental and mammary barriers to some degree. The positive significant correlation between Cd levels in molars and kidney cortex suggested that rodent molars indicate a degree of Cd absorption, whereas incisors do not. It is supposed that rodent molars are comparable to human deciduous teeth. It is thus confirmed that human deciduous teeth indicate previous lead exposure. It is further inferred that they also can be used as indicators of previous Cd‐exposures.  相似文献

Ischemic conditioning and exercise have been suggested for protecting against brain ischemia-reperfusion injury. However, the endogenous protective mechanisms stimulated by these interventions remain unclear. Here, in a comprehensive translational study, we investigated the protective role of extracellular vesicles (EVs) released after remote ischemic conditioning (RIC), blood flow restricted resistance exercise (BFRRE), or high-load resistance exercise (HLRE). Blood samples were collected from human participants before and at serial time points after intervention. RIC and BFRRE plasma EVs released early after stimulation improved viability of endothelial cells subjected to oxygen-glucose deprivation. Furthermore, post-RIC EVs accumulated in the ischemic area of a stroke mouse model, and a mean decrease in infarct volume was observed for post-RIC EVs, although not reaching statistical significance. Thus, circulating EVs induced by RIC and BFRRE can mediate protection, but the in vivo and translational effects of conditioned EVs require further experimental verification.  相似文献

Amyloidosis is a rare disease caused by the misfolding and extracellular aggregation of proteins as insoluble fibrillary deposits localized either in specific organs or systemically throughout the body. The organ targeted and the disease progression and outcome is highly dependent on the specific fibril-forming protein, and its accurate identification is essential to the choice of treatment. Mass spectrometry-based proteomics has become the method of choice for the identification of the amyloidogenic protein. Regrettably, this identification relies on manual and subjective interpretation of mass spectrometry data by an expert, which is undesirable and may bias diagnosis. To circumvent this, we developed a statistical model-assisted method for the unbiased identification of amyloid-containing biopsies and amyloidosis subtyping. Based on data from mass spectrometric analysis of amyloid-containing biopsies and corresponding controls. A Boruta method applied on a random forest classifier was applied to proteomics data obtained from the mass spectrometric analysis of 75 laser dissected Congo Red positive amyloid-containing biopsies and 78 Congo Red negative biopsies to identify novel “amyloid signature” proteins that included clusterin, fibulin-1, vitronectin complement component C9 and also three collagen proteins, as well as the well-known amyloid signature proteins apolipoprotein E, apolipoprotein A4, and serum amyloid P. A SVM learning algorithm were trained on the mass spectrometry data from the analysis of the 75 amyloid-containing biopsies and 78 amyloid-negative control biopsies. The trained algorithm performed superior in the discrimination of amyloid-containing biopsies from controls, with an accuracy of 1.0 when applied to a blinded mass spectrometry validation data set of 103 prospectively collected amyloid-containing biopsies. Moreover, our method successfully classified amyloidosis patients according to the subtype in 102 out of 103 blinded cases. Collectively, our model-assisted approach identified novel amyloid-associated proteins and demonstrated the use of mass spectrometry-based data in clinical diagnostics of disease by the unbiased and reliable model-assisted classification of amyloid deposits and of the specific amyloid subtype.  相似文献

Block copolymers remain one of the most extensively investigated classes of polymers due to their abilities to self-organize into various nanostructures and modify polymer/polymer interfaces. Despite fundamental and technological interest in these materials, only a handful of experimental phase diagrams exist due to the laborious task of preparing such diagrams. In this work, two copolymer series are each synthesized from a single macromolecule via sequential living anionic polymerization to yield molecularly asymmetric diblock and triblock copolymers systematically varying in composition. The phase behavior and morphology of these copolymers are experimentally interrogated and quantitatively compared with predictions from mean-field theories, which probe copolymer phase behavior beyond current experimental conditions.  相似文献

Efficient fertilizer use is a prerequisite for achieving optimum crop yield while avoiding environmental contamination. Cereal response to nitrogen (N), sulfur (S), and phosphorus (P) were determined for 6 years under differing tillage [conventional-till (CT) vs. no-till (NT)] and intensity of cropping (cereal/fallow vs. cereal/cereal). Semidwarf white winter wheat (Triticum aestivum L.) alternated yearly with either fallow or spring cereal [barley (Hordeum vulgare L.) or spring wheat] on a Typic Haploxeroll soil in a 415 mm rainfall zone. Fertilizer treatments were no fertilizer (None), N only (N), N plus S (NS), and N plus S plus P (NSP). Average application rate, when applied, was 109 kg N, 18 kg S, and 11 kg P ha–1. Average cereal yield without fertilizer was 1.82 t ha–1. Nitrogen increased grain yield in 6 of 6, S in 4 of 6, and P in 3 of 6 years, with P and S response significant the remaining years at the 10% probability level. Average yield increases were 1.11 t ha–1 for N, 0.93 t ha–1 for S, and 0.47 t ha–1 for P. The NT/CT yield ratio was 0.60, 0.75, 0.93, and 0.95 with None, N, NS, and NSP addition, respectively, indicating that N and S deficiency were more severe in no-till. Limited increase in the NT/CT ratio with P addition indicated that P deficiency was less affected by tillage. Winter wheat always yielded less under NT than CT regardless of fertility, whereas spring cereals reached equality when fertilized with NSP. Annually-cropped wheat yielded 52, 67, 89, and 90% of wheat after fallow with None, N, NS, and NSP, respectively. Thus N and S, but not P, deficiency was more intense with increased frequency of cropping. Adequate fertility was a prime prerequisite for efficient yield in all systems.  相似文献

The milk fat globule membrane-associated proteins adipophilin (alias adipocyte differentiation-related protein) and butyrophilin were purified from bovine milk by reverse-phase chromatography. The nucleotide sequence of bovine adipophilin was obtained via peptide mapping and sequencing of a mammary gland cDNA clone, which comprises 1841 nucleotides and has an open reading frame of 450 amino acids. By peptide mapping, 19% of the amino acid sequence was confirmed. The obtained amino acid sequence has 87 and 80% identical residues with human and mouse adipophilin, respectively. Alignment with the proteins perilipin and TIP47 revealed two highly conserved segments, which may assemble into amphipathic alpha-helices.  相似文献

This study explored the relation between preschoolers' television exposure and one important indicator of cognitive processing called theory of mind (ToM). A total of 107 preschoolers and their parents provided data on the preschoolers' television exposure (including both intentional viewing and exposure via background television), parent–child discussion of television, and preschoolers' ToM. The results indicated that preschoolers who were exposed to more background television and who had a television in their bedroom performed more poorly on ToM assessments compared with other children. Parent–child discussion of television was positively related to ToM performance, however. These results have implications for how we understand the effects of television on preschoolers.  相似文献

A proof of concept study of the feasibility of fully three-dimensional (3D) time-dependent simulation of nano-imprint lithography of polymer melt, where the polymer is treated as a structured liquid, has been presented. Considering the flow physics of the polymer as a structured liquid, we have followed the line initiated by de Gennes, using a Molecular Stress Function model of the Doi and Edwards type. We have used a 3D Lagrangian Galerkin finite element methods implemented on a parallel computer architecture. In a Lagrangian techniques, the node point follows the particle movement, allowing for the movement of free surfaces or interfaces. We have extended the method to handle the dynamic movement of the contact line between the polymer melt and stamp during mold filling.  相似文献
