A total of 35 coliforms isolated from bovine intramammary infections were tested for serum susceptibility. Growth responses of Klebsiella pneumoniae isolated from intramammary infections originating during the dry period were inhibited more than were growth responses of Klebsiella pneumoniae isolated from infections originating during lactation. Escherichia coli growth responses did not differ between isolates originating from dry period and lactational intramammary infections. Mean growth responses to serum did not differ between Klebsiella pneumoniae and Escherichia coli. Bactericidal properties of serum were due to complement activity. The classical pathway of complement appeared to be the principal source of bactericidal activity of serum toward Klebsiella pneumoniae. Alternative and classical pathways of complement were both important for bactericidal effects of serum on Escherichia coli. Iron saturation of serum did not affect growth responses of either Klebsiella pneumoniae or Escherichia coli. These data illustrate that coliforms isolated from intramammary infection originating during the dry period were not more resistant to the bactericidal activity of bovine serum than were coliforms isolated from intramammary infection occurring during lactation. 相似文献

Intramammary immunization was investigated as a procedure to reduce the clinical signs of coliform mastitis. Twenty-four cows were equally distributed to the following Escherichia coli J5 immunization schedules: 1) Subcutaneous injection 14 d prior to the end of lactation, intramammary immunization 7 d after drying off, and subcutaneous injection 30 d into the dry period; 2) subcutaneous injections at drying off, at 30 d into the dry period, and within 12 h after calving; and 3) unimmunized controls. Intramammary immunizations were the infusion of vaccine via the teat canal into each of the four mammary glands. Cows were challenged by infusion of E. coli 727 into one uninfected mammary quarter at approximately 30 d after calving. Intramammary immunization enhanced antibody titers against E. coli J5 and E. coli 727 compared with subcutaneous immunization. Immunoglobulin G titers against E. coli J5 and E. coli 727 in whey were greater at the time of challenge and 7 d after challenge for cows that received the intramammary immunization than for cows immunized by only subcutaneous injections. Serum IgG titers against E. coli 727 were enhanced at 7 d after challenge for cows receiving intramammary immunizations compared with conventionally immunized cows. Serum IgM titers against E. coli 727 were higher at calving for cows receiving intramammary immunization compared with conventionally immunized cows. Immunization schedule had minimal effect on systemic and local signs of clinical mastitis following challenge. 相似文献

Rates of environmental streptococcal and coliform intramammary infections were compared among quarters uninfected and infected with either Corynebacterium bovis or Staphylococcus species. Rate of environmental streptococcal intramammary infections was 3.9 times greater in C. bovis-infected quarters than in uninfected quarters. Rate of environmental streptococcal infections was 2.6 times greater in quarters infected with Staphylococcus species than in uninfected quarters. Rate of coliform intramammary infections did not differ among quarters with differing bacteriological infection statuses. Quarters infected with either C. bovis or Staphylococcus species had higher milk SCC than did uninfected quarters. Intramammary infection with either Corynebacterium bovis or Staphylococcus species did not protect quarters against coliform infection. Examined the peer relations and self-concepts of students prior to and following their identification by the school district as learning disabled (LD) in a 4- to 5-yr prospective study. Self-concept ratings (kindergarten–4th grade) and peer acceptance ratings (kindergarten–3rd grade), as well as academic achievement scores, were compared across 3 groups: LD students who were placed in resource special education programs during 2nd grade, low-achieving (LA) students, and average-achieving/high-achieving (AA/HA) students. For peer acceptance, AA/HA students' scores were higher than LA students' scores only. No between-groups differences were obtained during any school year on the self-concept measure. Findings suggest that LD students' self-perceptions are not negatively affected by academic and social difficulties in the early grades or by the identification and labeling process. Though generalization is limited by the small sample size, few studies have examined students with learning disabilities longitudinally or prior to and following their identification. The behavioral approach system (BAS) reflects the propensity to respond to signals of reward, including stimuli associated with safety and goal-oriented attack (e.g., anger). Hypomania/mania has been posited to involve increased BAS activity. In contrast, depression has been posited to involve decreased BAS activity. Building on past research, which suggests that increased left frontal cortical activity is a neurophysiological index of BAS activity, the present research tested the hypotheses that proneness toward hypomania/mania symptoms would be related to increased relative left frontal activity and that proneness toward depression symptoms would be related to decreased relative left frontal activity in response to an anger-evoking event. Results from 67 individuals who had completed a measure of proneness toward these affective symptoms and were exposed to an anger-evoking event supported the hypotheses. The association of abnormal uterine discharge with the development of intramammary infection (IMI) was studied in 62 multiparous Holstein cows during the nonlactating period and from lactation days 3 through 30. Duplicate milk samples were obtained from each mammary gland at approximately day 30 of the nonlactating period. Milk samples for bacteriologic culture also were obtained from each gland from all cows at the end of the previous lactation, at parturition, and on a minimum of 7 additional dates during the first 30 days of lactation. Beginning after parturition and continuing once weekly for 4 weeks, each cow was examined, using a vaginal speculum to visually estimate the quantity of abnormal uterine discharge in the vagina. Additionally, uterine swab specimens were obtained for aerobic bacteriologic culture. Cows were allotted to groups on the basis of the maximal amount of abnormal uterine discharge observed at any 1 of the 4 examinations. Cows in group 1 had normal discharge or < 30 ml of abnormal discharge; in group 2, > or = to 30 ml of abnormal discharge, observed only on examination by vaginal speculum; and in group 3, > or = to 30 ml of abnormal discharge visible externally. A difference was not detected in the development of new IMI in the nonlactating period between cows that subsequently developed uterine discharge and those that did not. The effects of intracellular levels of polyamines on both the in vivo inhibition of protein synthesis and the decrease of translation accuracy induced by streptomycin have been studied in polyamine-auxotrophic strains of Escherichia coli infected with the MS2 bacteriophage. The amount of viral coat protein formed was strongly reduced upon addition of increasing concentrations of streptomycin to polyamine-supplemented bacteria. In contrast, the antibiotic almost did not inhibit coat protein synthesis in polyamine-starved cells. The increase of mistranslation frequency elicited by streptomycin was only observed in bacteria grown with putrescine. In these cells several coat protein-satellites were detected after two-dimensional gel electrophoresis. These proteins, more basic than the normal MS2 coat protein, contain multiple substitutions of lysine for asparagine. 相似文献

DNA branch migration, a process whereby two homologous DNA duplexes exchange strands, is an essential component of genetic recombination. Models for homologous recombination have invoked spontaneous branch migration as one mechanism for the generation of large regions of heteroduplex DNA. During recombination, two homologous parental duplexes that contain similar, but not identical, sequences are paired and undergo strand exchange. An important issue is whether spontaneous branch migration is capable of traversing sequence heterology such as mismatches, insertions and deletions. We use a model four-strand system to examine the effect of mispaired or unpaired bases on branch migration. The assay consists of annealing two short duplexes having defined sequence heterologies. Following annealing, a Holliday junction is formed that is free to branch migrate. Our results demonstrate that a single base mismatch, insertion or deletion is sufficient to pose a substantial barrier to spontaneous branch migration. In the presence of magnesium, branch migration through such sequence heterologies is almost completely blocked. Others have shown that non-mobile four-way junctions undergo a dramatic shift in conformation in the presence of magnesium. Our data suggest that a similar transition occurs for the mobile Holliday junction. We also discuss how proteins may facilitate branch migration through sequence heterologies in vivo. 相似文献

The broad array of K- channels in epithelial cells includes voltage-dependent (mainly outward) and Ca(2+)-activated channels, and K+ channels modulated by adenosine triphosphate (ATP). Voltage-dependent K+ channels mediate Na+/K+ absorption and secretion; typically, they are found in the basolateral membrane and exhibit burst activity. Ca(2+)-activated K+ (Ca2+/K+) channels regulate activity by decreasing Ca2+ influx via voltage-gated Ca2+ channels. Ca2+/K+ channels exhibit conductances of 4-300 pS, and have a low open probability (< 10(-7)) at the level of the resting membrane potential. ATP-sensitive channels have been observed mainly in insulin-secreting pancreatic beta-cells and in the urinary tract, where the open state is rapidly closed by ATP. The channels are voltage-dependent, exhibit burst activity, and, in the case of the urinary-tract cells, are Ca2+ dependent. Chemical compounds that selectively open or block K+ channels have been exploited to characterize channels found in different cells, but no opener or blocker has been found that specifically affects only one type of K+ channel. Specialized model systems and recombinant techniques have led to a general understanding of the structure of K+ channels, but many important details remain to be determined. 相似文献
