Two purified animal venom toxins, crotoxin and cardiotoxin, have been combined to produce a unique natural product (VRCTC-310) currently under investigation as an antitumor agent by the National Cancer Institute. In vitro, it has demonstrated cytotoxic disease specificity and a unique mechanism of action when submitted to COMPARE analysis. In vivo, tolerance was developed to the neurotoxic properties of crotoxin which allowed comparison of several schedules of fixed and escalating daily i.m. doses to mice bearing s.c. Lewis Lung carcinoma. An 83% inhibition of tumor growth was achieved using an escalating dose schedule starting at 1.8 mg/kg and reaching 6.3 mg/kg/day on day 20. Although some irritation around the sites of i.m. injection was noted, animal weight loss was negligible and there were no other signs of adverse toxicity. This natural product represents a new, membrane interactive anticancer agent which produces a unique spectrum of cytotoxicity in vitro and which has demonstrated interesting in vivo antitumor efficacy. 相似文献

The G protein beta5 subunit differs substantially in amino acid sequence from the other known beta subunits suggesting that beta gamma dimers containing this protein may play specialized roles in cell signaling. To examine the functional properties of the beta5 subunit, recombinant beta5 gamma2 dimers were purified from baculovirus-infected Sf9 insect cells using a strategy based on two affinity tags (hexahistidine and FLAG) engineered into the N terminus of the gamma2 subunit (gamma2HF). The function of the pure beta5 gamma2HF dimers was examined in three assays: activation of pure phospholipase C-beta in lipid vesicles; activation of recombinant, type II adenylyl cyclase expressed in Sf9 cell membranes; and coupling of alpha subunits to the endothelin B (ETB) and M1 muscarinic receptors. In each case, the efficacy of the beta5 gamma2HF dimer was compared with that of the beta1 gamma2HF dimer, which has demonstrated activity in these assays. The beta5 gamma2HF dimer activated phospholipase C-beta with a potency and efficacy similar to that of beta1 gamma2 or beta1 gamma2HF; however, it was markedly less effective than the beta1 gamma2HF or beta1 gamma2 dimer in its ability to activate type II adenylyl cyclase (EC50 of approximately 700 nM versus 25 nM). Both the beta5 gamma2HF and the beta1 gamma2HF dimers supported coupling of M1 muscarinic receptors to the Gq alpha subunit. The ETB receptor coupled effectively to both the Gi and Gq alpha subunits in the presence of the beta1 gamma2HF dimer. In contrast, the beta5 gamma2HF dimer only supported coupling of the Gq alpha subunits to the ETB receptor and did not support coupling of the Gi alpha subunit. These results suggest that the beta5 gamma2HF dimer binds selectively to Gq alpha subunits and does not activate the same set of effectors as dimers containing the beta1 subunit. Describes advanced protocols for the discrimination and classification of neuronal spike waveforms within multichannel electrophysiological recordings. The programs are capable of detecting and classifying the spikes from multiple, simultaneously active neurons, even in situations where there is a high degree of spike waveform superposition on the recording channels. The protocols are based on the derivation of an optimal linear filter for each individual neuron. Each filter is tuned to selectively respond to the spike waveform generated by the corresponding neuron, and to attenuate noise and the spike waveforms from all other neurons. The protocol is essentially an extension of earlier work (S. Andreassen et al., 1979; W.M. Roberts and D.K. Hartline, 1975; R.B. Stein et al., 1979). However, the protocols extend the power and utility of the original implementations in two significant respects. First, a general single-pass automatic template estimation algorithm was derived and implemented. Second, the filters were implemented within a software environment providing a greatly enhanced functional organization and user interface. We show the massive reduction achievable, in both emissions and climate change impact, from enhanced nuclear energy use on the forecasts of future world energy use and its associated environmental impacts. A range encompassing the major scenarios for the World's energy demand have been analyzed using the latest version of the climate-modeling MAGICC/SCENGEN software (Version 4.1). We have updated and predicted the impacts of 80% substitution with CO2-free sources (likely predominantly nuclear) for coal-fired electricity (by 2030) and for transportation fuel (by 2040). For transportation, hydrogen produced by CO2-free sources would replace gasoline and diesel fuels. In this paper, to bracket the range of futures, we simply focus on two scenarios from the Intergovernmental Panel on Climate Change's (IPCC), one (A1FI) that is energy-profligate and one (B2) that is energy-conserving.The results show that, interestingly, projected average global temperatures for all scenarios are fairly similar until about 2035 (a further rise beyond the 1990 average temperature of +0.75 ± 0.1 K) regardless of energy usage and its sources. However, by 2050, the different IPCC scenarios diverge markedly. Understandably, A1FI is projected to have noticeably stronger effects than B2 on average global temperatures (about 0.3 K more in 2050) but the effect is much stronger over land at mid and high latitudes (up to almost 1 K more). What is most striking is that the substitution of CO2-free sources gives projected average temperature rises in 2050 over key land areas (North America and China) that are very similar for the two energy-use scenarios—typically 1–1.5 K because A1FI's additional energy is predominantly supplied by nuclear. In contrast, projected rises with the unaltered cases are markedly different being about 2.5 K for A1FI and 1.5–2 K for B2. The projected changes in rainfall distribution show similar patterns, especially for the expected increases in higher latitudes.With the assumption of no additional policies for substitution of energy sources beyond 2040, temperature divergence between the two scenarios of relative energy profligacy or conservation grows in the latter half of the 21st century, even with substitution. However, the early substitution of nuclear energy and hydrogen appears to buy time and is not crucially dependent on severe, near-term curtailment of energy use. Near-term curtailment is too difficult to implement at a time of rapid industrialization of major emerging economies. Of course, proportionately larger deployments of CO2-free energy sources are needed for more energy-intensive scenarios.Nuclear power must dominate as the source of CO2-free energy since it is proven, dependable, available on a large scale, and economic. Social objections to nuclear energy in some countries and quarters are seen as well-meaning but misguided distractions from solving the energy and environmental crises that are now facing world sustainability. Data from 10 sampling sites along the River Njoro are used to examine the contribution of nutrients from upstream land uses draining each of the sampling sites. The data also are used to assess whether both the proportion of land uses and the size of the subwatersheds account for the variability in water quality in the River Njoro watershed. Geographical Information System analysis was used to determine the spatial distribution of land‐cover types and subwatersheds contributing run‐off to the sampling sites in the River Njoro. Standard Digital Elevation Model‐based routines were used to establish the watershed area contributing run‐off to each sampling site. Water and sediment samples were collected for chemical analysis, and the nutrient levels were related to the upstream land‐use types and the size of the subwatersheds. The mid‐stream portion of the River Njoro (near Egerton University) accounts for the highest nutrient contributions. The percentage contribution is magnified by additions from industrial, human settlements and agricultural land uses around the University. There is a significant decrease in nutrient levels downstream, however, indicating natural purification as the river flows through an area of large‐scale farming with intense, well‐preserved riparian and in‐stream vegetation. Steep slopes of the land upstream of Egerton University enhance erosion and nutrient losses from those subwatersheds. Mixed small‐scale agricultural and bare lands contribute over 55% of the phosphorus load to the upper and mid‐reaches of the River Njoro. The size of the subwatershed accounts for about 53% of the variability in the soluble phosphorus in the river. The land‐use subwatershed proportions are important for characterizing and modelling water quality in the River Njoro watershed. Upland land uses are as important as near‐stream land uses. We suggest that conservation of intact riparian corridor along the river and its tributaries contributes significantly to natural purification processes and recovery of the ecological integrity of the River Njoro ecosystem. 相似文献

The present study evaluated the reliability and validity of the Triage Assessment Survey: Organizations (TAS:O), a 27-item, 5-point, Likert summated rating scale. One hundred and seventeen participants responded to the TAS:O after reading mild, moderate, marked and severe organizational crisis scenarios. The overall Cronbach's alpha and split-half reliability were both .93. Confirmatory factor analysis revealed three factors, supporting the hypothesis that the TAS:O is comprised of three distinct factors (i.e., Affect, Behavior, and Cognition). An analysis of variance provided evidence that the TAS:O has the capacity to distinguish among mild, moderate, marked, and severe crises. Because this research is the first to evaluate the TAS:O, further studies are needed to strengthen confidence in the psychometric properties of this scale. 相似文献
