We have developed a computer program for the rapid assessment of the primary structure differences between a protein of unknown sequence and a homologous known protein. Both proteins are reduced, alkylated, and digested with the same hydrolytic agent. The unfractionated peptide mixtures are submitted to automatic sequence analysis. Based on the knowledge of the reference sequence, the program utilizes the analysis data to identify all the potential peptides present in the two mixtures, determining their primary structure, homology degree, and molecular weight calculated both as integer MH+ and average mass variables. These fingerprints allow the user to easily identify the structural differences between the two proteins and clarify possible doubts by a mass spectrometric analysis of the two mixtures. OBJECTIVES: To assess retinal complications and to identify risk factors for retinal complications following aqueous shunt procedures. MATERIALS AND METHODS: Records of 38 consecutive aqueous shunt procedures that were performed on 36 patients at the Eye Institute of the Medical College of Wisconsin, Milwaukee, from June 1993 to March 1995 (minimum follow-up, 6 months) were reviewed. The mean +/- SD follow-up was 11.4 +/- 5.2 months (median, 10.5 months). RESULTS: Twelve patients (32%) had the following retinal complications: 4 serous choroidal effusions (10%) that required drainage, 3 suprachoroidal hemorrhages (8%), 2 vitreous hemorrhages (5%), 1 rhegmatogenous retinal detachment (3%), 1 endophthalmitis (3%), and 1 scleral buckling extrusion (3%). Surgical procedures for retinal complications were required in 8 (67%) of these 12 patients. Visual acuity decreased 2 lines or more in 9 (75%) of these 12 patients. The median onset of a postoperative retinal complication was 12.5 days, with 10 patients (83%) experiencing complications within 35 days. Serous choroidal effusions developed in 10 other patients (26%), and these effusions resolved spontaneously. Visual acuity decreased 2 lines or more in 2 (20%) of these additional 10 patients. Patients who experienced serious retinal complications were significantly older, had a higher rate of hypertension, and postoperative ocular hypotony. Serious retinal complications were distributed evenly among patients with Krupin valves with discs and Molteno and Baerveldt devices. Experience with the Ahmed glaucoma valve implant was limited. Knowledge of the viscosity of virgin olive oils (VOOs) is of great importance for the design of pilot plants, to determine the time required for the settling of particles at the end of the production chain and from a sensory view point. The dynamic viscosities of French VOOs from four different cultivars (‘Aglandau’, ‘Bouteillan’, ‘Salonenque’ and ‘Tanche’) were studied as a function of their fatty acid and TAG compositions and of the temperature [10–50°C]. These four VOOs had different TAG and fatty acid compositions representative of the range of compositional variations in the main French oils. Their viscosities were similar, although small but measurable differences that depended on their compositions were apparent. All the VOO samples exhibited the same dynamic viscosity pattern over temperature. For a given temperature, the viscosity difference was the greatest between Aglandau and Salonenque oils, Aglandau being the oil with the highest viscosity. The correlation between temperature and viscosity was highlighted by an Arrhenius model for this Newtonian fluid. The Arrhenius activation energy was correlated (R2 = 0.993) with the percentage of triolein, the main TAG in olive oil. 相似文献

The aim of the present work is to extend the concept of interphase and equivalent imperfect interface in the context of viscoplasticity. The interphase takes the form of a thin curved layer of constant thickness, made up of a rigid viscoplastic material located between two other surrounding materials. We aim at representing this interphase by an interface with appropriately devised interface conditions. To reach this objective, a Taylor expansion of the relevant physical fields in the thin region is used. It is shown that, depending of the degree of stiffness of the layer with respect to the neighboring media, this interphase can be replaced by an idealized imperfect interface involving the jump of the velocity field or the traction vector. The first kind of interface model, applicable to soft interphases, is the “spring-type” interface across which the traction are continuous but the velocity field exhibits a discontinuity which is given in term of the traction by a power-law type relation. Moreover, it is shown that the constant of the model can be expressed in terms of the material parameters of the interphase. When the interphase is stiffer than the two surrounding media, it can be replaced by an imperfect interface described by a generalization of the so-called Young Laplace model to viscoplastic solids. 相似文献

We explore a perturbative approach to calculation of binding free energy of multiple ligands, based on a single molecular dynamics simulation of a reference ligand-receptor complex and analysis via a hybrid force field/continuum model potential. The methodology is applied to prediction of relative binding free energies of 10 Influenza neuraminidase inhibitors, using Poisson-Boltzmann and generalised Born models of implicit solvent. These single-step MM-PB/SA and MM-GB/SA approaches predict the experimentally most potent ligand as first- or second-ranked according to total binding free energy. Ranking of inhibitors displays only moderate sensitivity to the choice of reference trajectory and ligand partial charge scheme. When ranked according to total electrostatic binding free energy, correlation with experiment improves (r(2) of 0.72); this may be related to underestimated first solvation shell effects by the implicit water models. Therefore, to increase the generality of this single-step approach as part of a potential computational compound optimisation strategy, further development of the treatment of short-range solvent interactions is warranted. 相似文献

A slip model for gas flows in micro/nano-channels induced by external body forces is derived based on Maxwell’s collision theory between gas molecules and the wall. The model modifies the relationship between slip velocity and velocity gradient at the walls by introducing a new parameter in addition to the classic Tangential Momentum Accommodation Coefficient. In PSA Peugeot Citroen factories, high precision requirements of workstations make them being manual. One of the main goal of the car manufacturer is to minimize the pain of workers while maintaining high efficiency of production lines. Consequently, assisting operators with an exoskeleton is a potential solution for improving ergonomics of painful workstations while respecting industrial constraints. To determine ergonomic performances of an exoskeleton, human joint angles and torques, ground reaction forces, and duration of operations are analysed for eight subjects performing a representative screwing task. Experiments were performed using ABLE upper-limb exoskeleton, developed by the French Atomic Energy Commission (CEA), which has the functionality to compensate arm and tools loads. Results show a clear reduction of the sum of the joints torques, up to 38.9%, given by ABLE supply and invite to make concrete the use of exoskeletons in car assembly lines. 相似文献

Accurate localization with high availability is a key requirement for autonomous vehicles. It remains a major challenge when using automotive sensors such as single‐frequency Global Navigation Satellite System (GNSS) receivers, a lane detection camera, and proprioceptive sensors. This paper describes a method that enables the estimation of stand‐alone single‐frequency GNSS errors by integrating the measurements from a forward‐looking camera matched with lane markings stored in a digital map. It includes a parameter identification method for a shaping model, which is evaluated using experimental data. An algebraic observability study is then conducted to prove that the proposed state vector is fully observable in a road‐oriented frame. This observability property is the basis to develop a road‐centered Extended Kalman filter (EKF) that can maintain the observability of every component of the state vector on any road, whatever its orientation. To accomplish this, the filter needs to handle road changes, which it does using bijective transformations. The filter was implemented and tested intensely on an experimental vehicle for driverless valet parking services. Field results have shown that the performance of the estimation process is better than solutions based on EKF implemented in a fixed working frame. The proposed filter guarantees that the drift along the road direction remains bounded. This is very important when the vehicle navigates autonomously. Furthermore, the road‐centered modeling improves the accuracy, consistency, and robustness of the localization solver. 相似文献
