Short-term exposure to high concentrations of ozone has been shown to increase airway responsiveness in normal humans and in all laboratory animal species studied to date. While our knowledge concerning the pulmonary effects of single exposures to ozone has increased rapidly over recent years, the effects of repeated exposures are less understood. The goal of the present study was to determine whether airway responsiveness is increased after near-lifetime exposure to ozone. Airway segments representing approximately eighth generation airways were isolated from Fischer 344 rats of both genders that had been exposed for 6 hr per day, 5 days per week for 20 months to 0, 0.12, 0.5, or 1.0 parts per million (ppm) ozone. Circumferential tension development was measured in isolated airways in response to bethanechol, acetylcholine, and electrical field stimulation. Responsiveness of the airways to the contractile stimuli was described by the effective dose or frequency that elicited half-maximum contraction (ED50) and the maximum response. Since ozone exposure is associated with remodeling of peripheral airways, smooth muscle area was determined and tension responses were normalized to the area measurements. Before normalization of tension data to smooth muscle area, neither the ED50 nor maximum response of small bronchi to the contractile stimuli was altered after chronic ozone exposure. Smooth muscle area was greater in airways isolated from animals that had been exposed to 0.5 ppm ozone. After accounting for smooth muscle area, maximum responses of the small bronchi isolated from male rats were significantly reduced after 0.12 and 0.5 ppm ozone. Although not significant statistically, a similar trend was observed in airways isolated from female rats. These results suggest that the increase in airway responsiveness associated with acute ozone exposure does not persist during near-lifetime exposure. Although the mechanism responsible for the adaptation to the effects of O3 on airway responsiveness is unknown, the results indicate that smooth muscle cell function was compromised by the chronic exposure. The mechanism(s) responsible for mediating this effect and the relevance of these results to humans remains to be determined.  相似文献

In situ hybridization of a biotin-labeled specific dopamine1A (D1A) receptor gene oligonucleotide probe combined with computer-assisted image analyzer was used to directly visualize D1A receptor mRNA and quantify the relative mRNA levels in sections of rat aorta and pulmonary and caudal arteries. Positive D1A receptor mRNA signals were found in rat aorta and pulmonary arteries, while no specific signals could be detected in the caudal artery. D1A receptor mRNA was located mainly within the medial layer of aorta, with intimal distribution in the pulmonary artery. The density of D1A receptor mRNA in different vascular beds demonstrated heterogeneity. D1A receptor mRNA levels in the aorta were much higher than those in the pulmonary artery (p < 0.01). These results demonstrate the existence of D1A receptor mRNA in both aorta and pulmonary beds, although with different distribution and density. The results further support the heterogeneity of the D1A receptor in different vascular beds.  相似文献

An antibody ELISA test and a PCR method for identifying the risk of Salmonella contamination were compared in a field study on the same lots of animals in a slaughterhouse. The results were compared to investigations carried out on two farms with different prevalences of Salmonella antibody-positive animals. Salmonella antibody ELISA testing was carried out on all 383 meat juice samples derived from the diaphragm pillar muscle of each pig. Salmonella DNA analysis was performed by PCR technique on small intestine samples with lymph nodes from all 383 pigs, and on tonsils from the last 129 pigs. The 383 animals tested came from 32 different pig farms. Furthermore, the herd antibody blood serum status against Salmonella spp. of weaners was determined on two selected pig fattening farms, one with low and one with high seroprevalence in meat juice. A total of 7.0% (ELISA cut-off OD% > or =40) of the slaughtered pigs from 6 of 32 fattening farms were seropositive. Salmonella DNA was found in 16.4% of the jejunum/lymph nodes (383 animals) and in 15.5% of the tonsils (129 animals). Salmonella DNA was found in the jejunum/lymph nodes of 41% of the seropositive pigs. However, serotitres were also positive in only 17.5% of all pigs positive in the jejunum DNA test. Two farms were selected for further investigation: farm 13 (F13), with a high prevalence of seropositive pigs, 29.0%, Category II; and F11, with 9.4%, Category I. However, categorization according to the blood serum tests of the fattening pigs after on-farm testing was very different: F13 had 5% positive animals (Category I); and F11, 23.3% (Category II). The study led to the following results and recommendations: First, ELISA tests are useful for the detection of farms that are regularly contaminated with Salmonella, but such tests cannot give information on the infectious status of a single animal (or a group) at the point of slaughter. Second, it is crucial that management measures are taken to prevent the spread of infections by trade and transport: piglets should be supplied exclusively by a single, well-known producer, and finishers should be tested serologically on farm before going to slaughter. Third, ELISA tests and the PCR method are suitable for the detection of Salmonella and are recommended as analytical tools for all pork quality control programmes. Fourth, animals from suspicious farms should always be slaughtered at the end of the slaughter day, followed by thorough cleaning and disinfection.  相似文献

Novel algorithms for adaptive approximation and interactive navigation of Pareto sets are applied to various homogeneous distillation processes with recycle streams within an industrial flowsheet simulator. The adaptive approximation scheme shows that the Pareto set consists of both convex and non-convex regions when the product purities are maximised while minimising the total heat duties. It is illustrated how such Pareto sets can be navigated interactively using a ray tracing technique. Our results suggest that the occurrence of non-convex regions in the Pareto sets is due to recycle streams in the flowsheets and is therefore the rule rather than the exception in multi-objective optimisation of flowsheet simulations in chemical engineering.  相似文献

A three‐dimensional steady‐state model to predict the flow and heat transfer in a rotary lime kiln is presented. All important phenomena are considered for the pre‐heat and calcination zones including turbulent gas flow, buoyancy, all modes of heat transfer, evolution and combustion of species and granular bed motion with calcination reaction. The model is based on a global solution of three sub‐models for the hot flow, the bed and the rotating wall/refractories. Information exchange between the models results in a fully coupled 3‐D solution of a rotating lime kiln. The overall model is validated using UBC's pilot kiln trials (5.5 m laboratory kiln). Results for this case are presented and potential implications are discussed.  相似文献

Porous Nylon 6 nanofibers were prepared using silica nanoparticles as the template. Firstly, Nylon 6/silica composite nanofibers were prepared as precursors by electrospinning Nylon 6 solutions containing different contents of silica nanoparticles. Scanning electron microscopy (SEM) and transmission electron microscopy (TEM) were used to examine the surface morphology and the inner structure of composite nanofibers; where it was found that silica nanoparticles were distributed both inside and on the surface of nanofibers. Analytical techniques [Fourier transform infrared (FTIR), differential scanning calorimetry, thermal gravimetric analysis (TGA), and wide‐angle X‐ray diffraction) were used to study the structure and properties of these composite nanofibers. The glass transition, melting, and crystallization processes of the fibers were affected by the addition of silica nanoparticles. Secondly, porous Nylon 6 nanofibers were obtained by removing silica nanoparticles via hydrofluoric acid treatment. The removal of silica nanoparticles was confirmed using FTIR and TGA tests. SEM and TEM observations revealed the formation of the porous structure in these nanofibers. After the formation of the porous structure, Brunauer–Emmett–Teller specific surface areas of nanofibers were increased as compared to solid Nylon 6 and composite nanofibers. © 2010 Wiley Periodicals, Inc. Due to the systematic increase in the production of nanomaterials (NMs) and their applications in many areas of life, issues associated with their toxicity are inevitable. In particular, the performance of heterogeneous NMs, such as nanocomposites (NCs), is unpredictable as they may inherit the properties of their individual components. Therefore, the purpose of this work was to assess the biological activity of newly synthesized Cu/TiO2-NC and the parent nanoparticle substrates Cu-NPs and TiO2-NPs on the bacterial viability, antioxidant potential and fatty acid composition of the reference Escherichia coli and Bacillus subtilis strains. Based on the toxicological parameters, it was found that B. subtilis was more sensitive to NMs than E. coli. Furthermore, Cu/TiO2-NC and Cu-NPs had an opposite effect on both strains, while TiO2-NPs had a comparable mode of action. Simultaneously, the tested strains exhibited varied responses of the antioxidant enzymes after exposure to the NMs, with Cu-NPs having the strongest impact on their activity. The most considerable alternations in the fatty acid profiles were found after the bacteria were exposed to Cu/TiO2-NC and Cu-NPs. Microscopic images indicated distinct interactions of the NMs with the bacterial outer layers, especially in regard to B. subtilis. Cu/TiO2-NC generally proved to have less distinctive antimicrobial properties on B. subtilis than E. coli compared to its parent components. Presumably, the biocidal effects of the tested NMs can be attributed to the induction of oxidative stress, the release of metal ions and specific electrochemical interactions with the bacterial cells.  相似文献

The dielectric behavior of some polar aromatics dissolved in nonpolar unoriented and stretched linear low‐density polyethylene was investigated within the temperature region between 150 and 350 K. The measurements were carried out in the frequency range 1 kHz to 10 MHz. The maximum temperatures and the half widths of the loss tangent peaks depend upon the shape and the polar structure of guest molecules. Stretching the samples induced a shift of the loss tangent to higher temperatures, decreased the height, and increased the width of tan δ peak. Oxidoreductases are attractive biocatalysts that convert achiral substrates into products of higher value, but they are also for the most part dependent on nicotinamide cofactors. Recently, biomimetic nicotinamide derivatives have received attention as less costly alternatives to natural cofactors. However, recycling of biomimetics is still challenging because there are only limited opportunities. Here, we have characterized various biomimetic cofactors with regard to stability and redox potentials to find the best alternative to natural cofactors. Further, the cofactor spectrum of NADH oxidase from Lactobacillus pentosus (LpNox) could be expanded, and the enzymatic activity was also compared to activities with different small‐molecule catalysts. As a result, we succeeded in identifying several strategies for regeneration of oxidized biomimetics.  相似文献
