The serpin plasminogen activator inhibitor 1 (PAI-1) can occur, in vitro, in both an inhibitory and a non-inhibitory but cleavable substrate form. In the present study, we have evaluated the effect of replacing the P13 to P10 region of PAI-1 (Val-Ala-Ser-Ser), with the P13 to P10 region of either the non-inhibitory serpin ovalbumin (Glu-Val-Val-Gly; PAI-1-ovalbumin) or the inhibitory serpin antithrombin III (Glu-Ala-Ala-Ala; PAI-1-antithrombin III). In addition, we have replaced Val at position P13 with Glu (PAI-1-P13 (Val-->Glu)). Wild-type (wt) PAI-1 revealed specific activities of 80+/-9% (mean+/-S.D., n=4) of the theoretical maximum value towards t-PA. PAI-1-ovalbumin, PAI-1-antithrombin III and PAI-1-P13 (Val-->Glu) revealed specific activities of 86+/-15%, 77+/-11%, and 100+/-30% respectively, towards t-PA and similar inhibitory properties towards u-PA. Surprisingly, upon inactivation at 37 degreesC, the active conformation of the PAI-1 mutants converted partly into a substrate conformation (i.e. 52+/-5.2%, 55+/-8.2% and 46+/-4.6% for PAI-1-ovalbumin, PAI-1-antithrombin III and PAI-1-P13 (Val-->Glu), respectively) and partly into a latent conformation. This is in contrast to active wtPAI-1 which, as expected, is converted to the latent conformation (i.e. 86+/-1.0%). In conclusion, even though replacement of the P13 to P10 region of PAI-1 by the corresponding region of a non-inhibitory serpin or of an inhibitory serpin, does not directly affect its inhibitory properties, the nature of the amino acids in this region and of P13 in particular, contributes to its conformational transitions.  相似文献

X-sialon has been produced by hot pressing Si3N4-Al2O3-SiO2 powder mixtures and Si3N4 Powder-mullite gel mixtures at 1650 °C. The formation mechanism ofX-sialon has been studied and is correlated with the processing technique for the two preparation routes. Microprobe analysis of the obtainedX-sialon phase, combined with previous observed compositions and literature formulae suggests that, at 1650 °C,X-sialon exists as a narrow solid solubility region on the Si3N4-mullite line in the Si3N4-SiO2-Al2O3-AIN phase diagram. The physical, mechanical and chemical properties ofX-sialon have been evaluated.X-sialon has a modest hardness of 1280 kg mm–2, a fracture toughness of 1.7 Mpa m1/2, and an elastic modulus of 213 GPa.X-sialon exhibits excellent chemical stability in contact with iron-based alloys at 1200 °C.  相似文献

In many developing countries, integration of ICT is a key component of an educational reform agenda. These countries can draw on a tradition of research in the developed world on factors determining integration of ICT in education. In this tradition, this study investigates the current situation of ICT integration in teacher education in Vietnam, an emerging developing country at the beginning of integrating ICT in education. 783 educators of five Vietnamese teacher education institutions completed a questionnaire. This analysis illuminates teacher educators’ access to ICT, their intensity of use, their related skills, and their confidence in using ICT, as well as their conceptions of learning. Exploratory multiple regression analysis addresses the importance of these different factors at the level of the educator for use of ICT in teaching practice. Even though teacher educators adhere to a constructivist approach to student learning, the use of ICT applications in teaching practice remains limited, mostly replacing traditional teaching practices. The factors currently determining the use of ICT in teaching practice are ICT skills (β = .522) and computer confidence (β = .158). Suggestions are provided for the country to move beyond an access and skills based approach of integration of ICT in education and for emerging developing countries to cease the promise of ICT for education.  相似文献

Abstract. The traditional worst-case analysis often fails to predict the actual behavior of the running time of geometric algorithms in practical situations. One reason is that worst-case scenarios are often very contrived and do not occur in practice. To avoid this, models are needed that describe the properties that realistic inputs have, so that the analysis can take these properties into account. We try to bring some structure to this emerging research direction. In particular, we present the following results: • We show the relations between various models that have been proposed in the literature. • For several of these models, we give algorithms to compute the model parameter(s) for a given (planar) scene; these algorithms can be used to verify whether a model is appropriate for typical scenes in some application area. • As a case study, we give some experimental results on the appropriateness of some of the models for one particular type of scene often encountered in geographic information systems, namely certain triangulated irregular networks.  相似文献

Waste Electrical and Electronic Equipment (WEEE) comprises a complex mix of engineering plastic grades often containing flame retardants. Whereas post-shredder recycling strategies are widely implemented, considerably higher economic and environmental returns can be expected based on information-rich dismantling scenarios. This paper therefore presents the results of an extensive technological feasibility study on diversified recycling strategies for a dismantling based approach. Results of mechanical and flammability tests as well as injection moulding trials are moreover presented to show the quality and potential of the recycling strategies. Subsequently, the environmental impact reduction potential is assessed using Life Cycle Assessment.  相似文献

30 vol.% 2 and 30 μm diamond dispersed Si3N4 matrix composites were prepared by pulsed electric current sintering (PECS) for 4 min at 100 MPa in the 1550–1750 °C range. The densification behaviour, microstructure, Si3N4 phase transformation and stiffness of the composites were assessed, as well as the thermal stability of the dispersed diamond phase. Monolithic Si3N4 with 4 wt% Al2O3 and 5 wt% Y2O3 sintering additives was fully densified at 1550 °C for 4 min and 60 MPa. The densification and α to β-Si3N4 transformation were substantially suppressed upon adding 30 vol.% diamond particles. Diamond graphitisation in the Si3N4 matrix was closely correlated to the sintering temperature and grit size. The dispersed coarse grained diamonds significantly improved the fracture toughness of the diamond composite, whereas the Vickers hardness was comparable to that of the Si3N4 matrix ceramic. The Elastic modulus measurements were found to be an excellent tool to assess diamond graphitisation in a Si3N4 matrix.  相似文献

Atomic layer deposition (ALD) of thin Al2O3 (≤10 nm) films is used to improve the rear surface passivation of large‐area screen‐printed p‐type Si passivated emitter and rear cells (PERC). A blister‐free stack of Al2O3/SiOx/SiNx is developed, leading to an improved back reflection and a rear recombination current (J0,rear) of 92 ± 6 fA/cm2. The Al2O3/SiOx/SiNx stack is blister‐free if a 700°C anneal in N2 is performed after the Al2O3 deposition and prior to the SiOx/SiNx capping. A clear relationship between blistering density and lower open‐circuit voltage (VOC) due to increased rear contacting area is shown. In case of the blister‐free Al2O3/SiOx/SiNx rear surface passivation stack, an average cell efficiency of 19.0% is reached and independently confirmed by FhG‐ISE CalLab. Compared with SiOx/SiNx‐passivated PERC, there is an obvious gain in VOC and short‐circuit current (JSC) of 5 mV and 0.2 mA/cm2, respectively, thanks to improved rear surface passivation and rear internal reflection. Since the discovery of a flavin-dependent thymidylate synthase (ThyX or FDTS) that is absent in humans but crucial for DNA biosynthesis in a diverse group of pathogens, the enzyme has been pursued for the development of new antibacterial agents against Mycobacterium tuberculosis, the causative agent of the widespread infectious disease tuberculosis (TB). In response to a growing need for more effective anti-TB drugs, we have built upon our previous screening efforts and report herein an optimization campaign of a novel series of inhibitors with a unique inhibition profile. The inhibitors display competitive inhibition toward the methylene tetrahydrofolate cofactor of ThyX, enabling us to generate a model of the compounds bound to their target, thus offering insight into their structure–activity relationships.  相似文献
