Microneedles are small needle‐like structures that are almost invisible to the naked eye. They have an immense potential to serve as a valuable tool in many medical applications, such as painless vaccination. Microneedles work by breaking through the stratum corneum, the outermost barrier layer of the skin, and providing a direct path for drug delivery into the skin. A lot of research has been presented over the past two decades on the applications of microneedles, yet the fundamental mechanism of how they interact, pressure, and penetrate the skin in its native state is worth examining further. As such, a major difficulty with understanding the mechanism of microneedle–skin interaction is the lack of an artificial mechanical human skin model to use as a standardized substrate. In this research news, the development of an artificial mechanical skin model based on a thorough mechanical study of fresh human and porcine skin samples is presented. The artificial mechanical skin model can be used to study the mechanical interactions between microneedles and skin, but not diffusion of molecules across skin. This model can assist in improving the performance of microneedles by enhancing the reproducibility of microneedle depth insertions for optimal drug delivery and biosensing.

Many industrially important catalysts consist of late transition metal particles supported on the surfaces of oxide materials. Our studies of such systems using model catalysts consisting of metal films vapor deposited onto the surfaces of single-crystalline oxides are reviewed here. Systems studied include Cu on ZnO, Pt on ZnO, Au on TiO2 and Cu, Ag and Pb on MgO. A unique adsorption microcalorimeter was developed to measure directly the energetic stability of the metal atoms on the oxide surfaces and the adhesion energy at the metal/oxide interface, which clarify the structural and chemisorption properties of the ultrathin metal particles. The structure of the oxide surface and the metal particles was elucidated by low-energy electron diffraction (LEED), low-energy ion scattering spectroscopy (ISS), angular-resolved X-ray photoelectron spectroscopy (XPS) and X-ray photoelectron diffraction (XPD). The electronic character of the metal particles was revealed by XPS, Auger electron spectroscopy (AES), band-bending and work function measurements. Sintering rates were measured by temperature-programmed ion scattering spectroscopy (TPISS). The chemisorption properties of these particles and their catalytic reactivity were monitored by mass spectroscopy and temperature-programmed desorption (TPD).  相似文献

Microcellular foams in polypropylene containing rubber particles were produced in an injection molding process. The foams are generated because of the thermodynamic instability and are controlled by formation process. The effect of processing parameters on microcellular foaming was investigated in the injection molding process. Injection speed and pressure are less important factors but packing pressure plays an important role in controlling the foam density. A critical packing pressure, about 5 × 106 Pa, was found to generate microcellular foams in our polypropylene material system. Rubber particles inside the polypropylene seem to stabilize the microcellular foams.  相似文献

We describe a drug-design strategy termed REPLACE (REplacement with Partial Ligand Alternatives through Computational Enrichment) in which nonpeptidic surrogates for specific determinants of known peptide ligands are identified in silico by using a core peptide-bound protein structure as a design anchor. In the REPLACE application example, we present the effective replacement of two critical binding motifs in a lead protein-protein interaction inhibitor pentapeptide with more druglike phenyltriazole and diphenyl ether groups. These were identified through docking of fragment libraries into the volume of the cyclin-binding groove of CDK2/cyclin A vacated through truncation of the inhibitor peptide-binding determinants. Proof of concept for this strategy was obtained through the generation of potent peptide-small-molecule hybrids and by the confirmation of inhibitor-binding modes in X-ray crystal structures. This method therefore allows nonpeptide fragments to be identified without the requirement for a high-sensitivity binding assay and should be generally applicable in replacing amino acids as individual residues or groups in peptide inhibitors to generate pharmaceutically acceptable lead molecules.  相似文献

The objective of this study was to assess the effectiveness of an engineered shallow water cover in reducing the oxidation of sulfidic mine tailings and thus preventing the development of acid rock drainage. Fresh tailings were submerged under a 0.3-m water cover in experimental field cells. From 1996 to 1998, we followed the chemistry of the interstitial water near the tailings-overlying water interface using in situ dialysis, and determined pH and dissolved oxygen (DO) profiles across the tailing water interface using micro-electrodes. Penetration of DO into the tailings was limited to <7 mm, even in the presence of DO produced by benthic periphyton. Anoxia in the tailings was further demonstrated by the appearance of dissolved sigmaH2S, Fe and Mn in pore water at depths -1.5 cm below the interface. However, there was clear evidence of surface oxidation of the mine tailings at the mm scale (i.e., DO depletion, coupled with localized increases in [H+] and [SO4(2-)]). Mobilization of Cd and Zn from this surface layer was indicated by the presence of sub-surface peaks in the concentrations of these two metals in the tailings interstitial water and by a change in their solid phase partitioning from refractory to more labile fractions. In contrast, mobilization of Cu from tailings was less evident. Unlike previous reports, which suggested that submerged tailings were effectively inert, our results show alteration of the superficial layer over time.  相似文献

The inactivation of bioactive ingredients during spray drying is often matrix specific. Therefore, the design of new processes or the optimisation of existing spray drying processes is usually highly product specific and requires numerous experiments. Rapid experimentation methods that facilitate fast data generation are therefore desired. A novel method for drying single droplets to mimic spray drying is proposed. The approach involves droplet deposition on a hydrophobic flat surface followed by controlled drying. A heat and mass transfer model is applied to predict the drying history of the single droplets. The approach is successfully evaluated through studying the inactivation of β-galactosidase during drying. The heat and mass transfer model supplemented with inactivation kinetics provided reasonable prediction of the residual enzyme activity after drying. In addition, the inactivation kinetics could be directly extracted from single droplet experiments rather than using the kinetics from separate heating experiments. Finally, it was demonstrated that the inactivation kinetics found with the single-drop experiments could satisfactorily predict the residual activity of β-galactosidase dried with a laboratory-scale spray dryer.  相似文献

The avalanche buildup time of an avalanche photodiode can be determined from the frequency response of the noise power as a function of the dc multiplication M0. In this paper we report on the first measurements of the avalanche buildup time of InP/InGaAsP/ InGaAs avalanche photodiodes with separate absorption, grading, and multiplication regions (SAGM-APD's). Measurements on several different device structures reveal that the avalanche buildup time (gain-bandwidth product) decreases (increases) with increasing carrier concentration in the multiplication region. The shortest buildup time that we have observed wasM_{0} times 4.2ps which corresponds to a gain-bandwidth Product of 38 GHz.  相似文献

为了解决目前人工晶状体不能实现大跨度调节问题,根据人体生物眼测量基础数据及调节机制,建立了人工晶状体襻结构生物力学模型.基于结构拓扑优化方法,对人工晶状体襻结构进行了拓扑优化分析,得到布局合理的基本骨架结构.经机构运动学分析,给出了机构目标点位移值的变化规律.采用柔性建模理论方法,考虑材料特性及关节柔性,深入研究了该放大机构的实际运动规律,给出了该柔性机构位移放大的一般规律.经本模型优化后得到柔性机构位移输出值可达输入值的3.34倍,光学部前置位移随之增加,理论上可实现视觉质量的提高.  相似文献
