Pregnancy is characterized by adaptations in the function of several maternal body systems that ensure the development of the fetus whilst maintaining health of the mother. The renal system is responsible for water and electrolyte balance, as well as waste removal. Thus, it is imperative that structural and functional changes occur in the kidney during pregnancy. However, our knowledge of the precise morphological and molecular mechanisms occurring in the kidney during pregnancy is still very limited. Here, we investigated the changes occurring in the mouse kidney during pregnancy by performing an integrated analysis involving histology, gene and protein expression assays, mass spectrometry profiling and bioinformatics. Data from non-pregnant and pregnant mice were used to identify critical signalling pathways mediating changes in the maternal kidneys. We observed an expansion of renal medulla due to proliferation and infiltration of interstitial cellular constituents, as well as alterations in the activity of key cellular signalling pathways (e.g., AKT, AMPK and MAPKs) and genes involved in cell growth/metabolism (e.g., Cdc6, Foxm1 and Rb1) in the kidneys during pregnancy. We also generated plasma and urine proteomic profiles, identifying unique proteins in pregnancy. These proteins could be used to monitor and study potential mechanisms of renal adaptations during pregnancy and disease.  相似文献

The medical community recommends that infant formulae should mimic human milk as far as possible, particularly in regards to long-chain polyunsaturated fatty acids (LC-PUFA). These include arachidonic acid (AA) and docosahexaenoic acid (DHA), both of which provide biochemical and functional benefits to neonates. However, LC-PUFAs are highly susceptible to oxidation and the composition of formulae must be carefully controlled. In this study, the stability of two types of LC-PUFA-supplemented milk-based powdered infant formula was evaluated over the course of 18 months storage at 25 °C and 40 °C. One contained egg yolk phospholipids (IF-EPL) and the other contained triacylglycerides (DHA and AA) synthesized by single cell oils (IF-SCO). The following parameters were monitored: peroxide values, volatile content (propanal, pentanal and hexanal), fatty acid profiles, and potential and free furfural content (5-hydroxymethyl-2-furaldehyde and 2-furaldehyde). In addition, these formulae were subjected to sensory evaluations by a panel of experts. The parameters studied revealed acceptable lipid stability in both types of formula, with better results for IF-EPL. At the end of the study period, significant deficits (p < 0.05) in linoleic acid were noted in both formulae. However, no significant decreases were observed in the other fatty acids, including AA and DHA. In regards to furfural content, both formulae exhibited a similar increase, indicative of the typical Maillard reaction characteristic of products stored for long periods.  相似文献

In many categories, weight has been found to influence how users perceive and appraise products. However, to date, the influence of the weight of the dish in which food is served on people’s perception has not been studied empirically. This exploratory study was therefore designed to investigate whether the weight of the container would exert a significant influence on people’s sensory and hedonic responses to the food consumed from it. Three bowls, identical except for the fact that they were different weights, were filled with exactly the same yoghurt. Consumers evaluated the yoghurt samples from the three bowls holding them with one hand, one at a time. Participants rated flavor intensity, density, price expectation, and liking using 9-point likert scales. Significant effects were found for all attributes except for flavor intensity. The effects on both density and price expectation ratings were highly significant.These findings are potentially relevant for designers and those working in restaurants, the hospitality sector, and food production, since the design and choice of dishes (or packages) of various weights could potentially be used to help enhance and/or modify the way in which consumers perceive and experience the food consumed from them.  相似文献

Graft copolymers of crosslinked poly(N‐isopropyl acrylamide) (PNIPAAm) and homopolyguluronic acid (GG) and homopolymannuronic acid (MM) fractions of alginic acid were synthesized. MM and GG block fractions were obtained by partial acid hydrolysis of the alkaline extract from the brown seaweed Macrocystis pyrifera. The conjugation of these block fractions with the synthetic polymer was achieved by amidation with crosslinked PNIPAAm functionalized with an amino group at the end of the polymer chain. The structure of conjugates was determined by Fourier transform infrared and NMR spectroscopy. Atomic force microscopy of the graft copolymer GG‐g‐PNIPAAm showed a regular porous pattern, whereas the MM‐g‐PNIPAAm graft copolymer showed a regular netlike structure. Aqueous solutions of the synthesized graft copolymers afforded hydrogels by stirring with 0.1M CaCl2. The hydrogels showed a well‐defined stimulus–response to temperature and pH. The swelling, thermal, and pH characterizations demonstrated the superior properties of the GG‐g‐PNIPAAm hydrogel over the MM‐g‐PNIPAAm hydrogel. © 2015 Wiley Periodicals, Inc. J. Appl. Polym. This work aimed to optimize the extraction of an extracellular protease produced by the cold-adapted yeast Rhodotorula mucilaginosa L7 using aqueous two-phase systems (ATPS) comprising polyethylene glycol (PEG) and sodium citrate or sodium tartrate. First, the biocompatibility of the phase forming agents was assessed. The results obtained with PEG-2000, PEG-4000, and PEG-6000 demonstrated that even at large PEG concentrations (32 wt%) the protease maintains its activity after 3 h of reaction, whereas an increase in salt concentration provokes a gradual decrease in protease stability. Subsequently, the partitioning of the protease in both types of ATPS was assessed, evaluating the effect of temperature, molecular weight, and concentration of PEG on protease purification, using two 23-full factorial designs. The best partitioning conditions were obtained in PEG-6000/sodium tartrate-based ATPS, at 30ºC (with a yield of 81.09 ± 0.66% and a purification factor of 2.51 ± 0.03). Thus, considering the biodegradable characteristics of the system, the PEG/sodium tartrate ATPS is a viable and economic low-resolution step in protease purification, with a strong potential for future industrial application.  相似文献

Flaxseed oil is an alpha linolenic acid source important in the growth and body development stage; furthermore, this acid acts on adipose tissue and bone health. The aim of this study was to evaluate body composition, fatty acid composition, hormone profile, retroperitoneal adipocyte area and femur structure of pups at weaning, whose mothers were fed a diet containing flaxseed oil during lactation. After birth, pups were randomly assigned: control (C, n = 12) and flaxseed oil (FO, n = 12), rats whose mothers were treated with diet containing soybean or flaxseed oil. At 21 days, the pups were weaned and body mass, length, body composition, biochemical parameter, leptin, osteoprotegerin, osteocalcin, fatty acids composition, intra‐abdominal fat mass and femur structure were analyzed. FO showed (p < 0.05): higher body mass (+12 %) and length (+9 %); body fat mass (g, +45 %); bone mineral density (+8 %), bone mineral content (+55 %) and bone area (+35 %), osteocalcin (+173 %) and osteoprotegerin (+183 %). Arachidonic acid was lower (p < 0.0001), alpha‐linolenic and eicosapentaenoic were higher (p < 0.0001). Intra‐abdominal fat mass was higher (+25 %), however, the retroperitoneal adipocytes area was lower (?44 %). Femur mass (+10 %), distance between epiphyses (+4 %) and bone mineral density (+13 %) were higher. The study demonstrates that adequate flaxseed oil content during a lactation diet plays an important role in the development of pups.  相似文献

The birefringence and turbidity of a polystyrene/poly(methyl methacrylate) (PMMA) blend, with the concentration of the PMMA dispersed phase ranging up to 1%, were measured in both a slit channel with a constant cross section and a planar hyperbolic contraction/expansion (8:1:8). The measurements were performed by the attachment of a modular rheo‐optical die to a twin‐screw extruder. The optical arrangement had a red light‐emitting diode as the source and two photoresistors, with one of them measuring the turbidity and the other one measuring the transmitted intensity between cross‐polarizers. The experimental procedure consisted of the stopping of the extruder feeding, while the screw rotation was kept constant. Because the form birefringence could be associated with the shape of the droplets, these measurements were used to infer information about the PMMA droplet deformation and breakup. © 2016 Wiley Periodicals, Inc.
