Drugs that decrease drug-maintained responding at doses that do not decrease other behaviors in animals may be suitable candidates for development as medications to treat drug abuse in humans. The present study examined whether this effect could be obtained with phentermine, a drug that has been reported to decrease cocaine intake in humans. Rhesus monkeys were trained under multiple fixed-ratio 30-response schedules of food and i.v. cocaine delivery. Phentermine was always given as a slow, i.v. infusion. Acute treatment with phentermine (0.3-10 mg/kg) decreased cocaine-maintained responding at doses that did not decrease, or decreased less, food-maintained responding for each of three unit doses of cocaine (10-100 microg/kg/injection). Subacute treatment with phentermine (3 or 5.6 mg/kg, daily) also decreased cocaine-maintained responding more than food-maintained responding. After subacute treatment was terminated, rates of cocaine-maintained responding generally recovered to levels comparable to those seen during untreated control sessions. Phentermine (0.3-3 mg/kg) did not generally increase responding associated with a very low (1 microg/kg/injection) unit dose of cocaine, suggesting that the decrease in cocaine-maintained responding at higher unit doses was not the result of a leftward shift in the cocaine unit dose-effect function. Phentermine (0.1-3 mg/kg) decreased responding maintained by 1-[2-[bis(4-fluorophenyl) methoxy]ethyl]-4-[3-phenylpropyl] piperazine (GBR 12909) (30 microg/kg/injection) at doses similar to those that decreased food-maintained responding. These results show that phentermine is effective in decreasing cocaine self-administration and suggest that it may be an effective medication for cocaine abuse. 相似文献

In order to optimise use of the limited resources (time, power) of an autonomous underwater vehicle (AUV) with a miniaturised X-ray fluorescence (XRF) spectrometer on board to carry out in situ autonomous chemical mapping of the surface of sediments with desired resolution, a genetic algorithm for rapid curve fitting is reported in this paper. This method quickly converges and provides an accurate in situ assessment of metals present, which helps the control system of the AUV to decide on future sampling locations. Methods for analyzing for silicon and silicone in biological matrixes were developed. A silicone-specific technique involved microwave digestion of samples in acid solution to rapidly break down the biological matrix while hydrolyzing silicones to monomeric species. The resulting monomeric silanol species were then capped with trimethylsilyl groups, extracted into hexamethyldisiloxane, and analyzed by gas chromatography. In serum, positive identification of silicone species with detection limits below 0.5 microgram of Si/mL are possible with this technique. The technique is compared with a silicone-specific technique, 29Si NMR, and a non-silicone-specific technique, ICP-AES. 29Si NMR was far less sensitive, with a detection limit of only 64 micrograms of Si/mL in serum when analyzing for one compound with a single sharp resonance. Inductively coupled plasma-atomic emission spectroscopy (ICP-AES) has potentially lower detection limits, but the technique is not silicone-specific and suffers from species-dependent responses. 相似文献

Use of poly(tetrafluoroethylene)s as a sample support for the MALDI-TOF analysis of DNA and proteins
A proline rich antigen (PRA), which protects mice against Coccidioides immitis, has been analyzed for differential expression and variation among isolates. Northern blots of RNA from different stages of growth were probed with previously cloned cDNA and showed that mRNA for PRA increased as spherules transformed and matured from mycelia. This pattern corresponds to the relative potency of whole cell vaccines from similar preparations. The PRA gene was then cloned from a genomic library of the Silveira strain of C. immitis and its sequence analyzed. Eight other coccidioidal isolates, selected for diversity in geographic origin and resulting clinical disease, demonstrated heterogeneity in Southern blots and in sequences of polymerase chain reaction products. Silveira differed from other California isolates at 33 of 555 bases, whereas it differed from non-California isolates by <=2 bases. Only one of these substitutions affected protein sequence. Thus, tests or vaccines based on this gene are likely to cover most isolates. 相似文献

Emulsifiable oils are often used in posttensioned construction to reduce friction losses and provide temporary corrosion protection for tendons prior to grouting. This paper addresses the effects of two emulsifiable oils and three duct types on bond and friction losses. Bond test results indicate that corrugated galvanized steel ducts provide better anchorage than corrugated HDPE ducts. Rigid steel pipes performed poorly because of failure at the duct-concrete or grout-duct interface. Bond test results also indicate that the ultimate strength of posttensioned specimens with oiled tendons is similar to or better than the ultimate strength of specimens with unoiled tendons. However, specimens with oiled tendons experienced greater slip at a given load than specimens with unoiled tendons. Friction test results indicate that current recommended design values for the coefficient of friction for steel pipes and galvanized ducts are accurate. However, the measured coefficient for HDPE ducts is significantly less than the AASHTO-recommended value. Friction tests also indicate that lubrication of the tendon reduces the friction coefficient by 15% in rigid steel pipes and HDPE ducts if stressing occurs while the oil is fresh. 相似文献

Cement clinker produced using a high sulfur petroleum coke fuel has been analysed to determine the identity of the sulfate‐containing phases. Quantitative X‐ray diffraction methods were used in conjunction with extraction procedures to concentrate or extract the sulfate phases. The minerals of interest were anhydrite (CaSO4), aphthitalite (3K2SO4·Na2SO4), arcanite (K2SO4), calcium langbeinite (K2SO4·2CaSO4) and thenardite (Na2SO4). Overall sulfur content of the clinker increased in proportion with the amount of sulfur in the fuel. The clinker produced using the high sulfur fuel was found to contain a significantly increased concentration of aphthitalite but a reduced amount of thenardite. Comparison of the experimental results with theoretical predictions based on the Bogue formulae shows differences in respect of calcium langbeinite, which is not detected in the clinker, and thenardite, which is detected.
