Glutamate receptors are implicated in the genesis of opioid tolerance and dependence. Factors governing release of amino acids in systems chronically exposed to opiates, however, remain undefined. Using rats, each prepared with a spinal loop dialysis catheter and with a chronic lumbar intrathecal infusion catheter connected to a subcutaneous minipump, the release of amino acids before and during antagonist-precipitated withdrawal in unanesthetized rats was examined. Spinal infusion of morphine (20 nmol/micro l/hr) for 4 d had little effect on resting release of amino acids. In morphine-infused, but not saline-infused, rats naloxone (2 mg/kg, i.p.) evoked an immediate increase in the release of L-glutamate (299 +/- 143%) and taurine (306 +/- 113%) but not other amino acids. The magnitude and time course of the release of these amino acids significantly correlated with behavioral indices of withdrawal intensity. Acute intrathecal pretreatment immediately before naloxone with clonidine (20 microg; alpha2 agonist), MK-801 (3 microg; noncompetitive NMDA antagonist), or aminophosphonopentanoic acid (AP-5; 3 microg; competitive NMDA antagonist) suppressed naloxone-induced increases in spinal L-glutamate and taurine release and behavioral signs of withdrawal in spinal morphine-infused rats. Results point to a correlated increase in spinal L-glutamate release, which contributes to genesis of the opioid withdrawal syndrome. Agents such as clonidine that suppress opioid withdrawal may owe their action to an inhibition of excitatory amino acid release. The effects of MK-801 and AP-5 suggest a glutamate-evoked glutamate release.  相似文献

Despite all advances in the diagnosis and therapy of cardiovascular diseases, the mortality from malignant ventricular tachyarrhythmias is still a major health problem. In addition to established therapeutic strategies in the prevention of sudden cardiac death such as antiarrhythmic drug treatment, catheter ablation or antiarrhythmic drug treatment, cardioverter/defibrillator was introduced to clinical practice in 1980. The number of 50,000 overall implants reflects the current clinical status of the therapy with implantable cardioverter/defibrillators. Significant technical improvements in the defibrillator therapy may contribute to an increase in therapy acceptance. These advances include the introduction of nonthoracotomy lead systems, enhanced defibrillation efficacy, full programmable devices providing tiered electrical therapy, improved diagnostic Holter functions and enhanced arrhythmia detection algorithms. The major present goals of defibrillator therapy are detection and termination of malignant ventricular tachyarrhythmias, prevention of sudden cardiac death, reduction in patient's mortality and improvement in quality of life. The efficacy and safety of defibrillator therapy to prevent sudden arrhythmic death has been proven in several large clinical investigations In patients with this device the annual sudden cardiac death mortality is < 2% even in high-risk patient populations. Compared to sudden cardiac death rate there is a much higher rate of overall cardiac mortality because a defibrillator is not able to prevent nonarrhythmic cardiovascular deaths. There is a clinical impression that cardiovascular mortality is lower in patients treated with an implantable cardioverter/defibrillator compared to patients treated with other therapies. However, there are no results from controlled studies providing scientific evidence that defribillator therapy can decrease overall cardiovascular mortality.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Paclitaxel induces a cell cycle block at G2-M phase by preventing the depolymerization of microtubules and induces p53-independent apoptosis in many cancer cells. We observed that gastric cancer cells treated with paclitaxel have shown a cyclin-dependent kinase (CDK)4 down-regulation. This paclitaxel-induced CDK4 down-regulation resulted in a cell cycle arrest at G1-S phase. To confirm this observation, we prepared stable transfectants that overexpressed CDK4 and analyzed the cell cycle progression. Ectopic expression of CDK4 in SNU cells resulted in a release of paclitaxel-induced G1 arrest. The release of G1 arrest by enforced expression of CDK4 seems to make the cells more sensitive to paclitaxel-induced apoptosis. From this finding, we could then suggest that paclitaxel treatment induces both G1-S and G2-M blocks in the cell cycle progression of gastric cancer cells.  相似文献

2-Amino (6), 2-alkylamino (7-8), 2-methoxy (9), 2-acetamido (10), and 5,8-diacetoxy (11) derivatives of the lead compound 2,3-dichloro-5,8-dimethoxy-1,4-naphthoquinone (4) were synthesized, together with 6,7-dichloro-5,8dimethoxy-1,4-naphthoquinone (5), a positional isomer of 4. Antiplatelet, antiinflammatory, and antiallergic activities were evaluated, and most compounds were quite potent in all assays. Compounds 5 and 9-11 were especially active; however, 5 was ineffective against neutrophil superoxide formation, and 10 was ineffective against mast cell degranulation.  相似文献

METHODS: Three hundred forty-two patients with lung cancer and 99 patients with nonneoplastic lung diseases (control group) underwent intraoperative pleural lavage with 300 ml physiologic saline solution before (lavage I) and after resection (lavage II). RESULTS: Studies of the lavage fluid in all control patients were negative, that is, there were no false positive findings. Tumor cells were found in lavage I in 132 patients (38.6%) and also in lavage II in 99 of them. In stage I (pT1 N0, pT2 N0) lung cancer, tumor cell detection was possible in 47 patients (28.6%). The 4-year survival of patients with resected non-small-cell lung cancer was 24% (95% confidence interval, 16% to 32%) if lavage I results were positive and 52% (95% confidence interval, 45% to 59%) if lavage I results were negative (all stages, p = 0.007). For patients with stage I disease (n = 164) the 4-year survival was 35% (95% confidence interval, 18% to 52%) if lavage I results were positive (n = 47), and 69% (95% confidence interval, 60% to 78%) if lavage I results were negative (n = 117) (p = 0.037). On multivariate analysis the positive cytologic result in intraoperative pleural lavage was an additional prognostic factor for our patients. To prove how the tumor cells enter the pleural cavity, we performed tissue cultures of tumor-free parenchyma in 23 cases of lung cancer. Tumor cell detection by histology and immunohistology was possible in 16 cases (69.6%). Detection of tumor cells in pleural lavage fluid before resection proves that tumor cells have spread into the pleural cavity. CONCLUSION: The positive result in pleural lavage seems to be a prognostic predictor for patients with lung cancer.  相似文献

Epoxidized soybean oil (ESO) was thermally cured using methylhexahydrophthalic anhydride (MHHPA) curing agent in the presence of 2-ethyl-4-methylimidazole (EMI) catalyst. The curing characteristics of ESO/MHHPA/EMI systems were characterized using Fourier transform infrared spectroscopy (FTIR), a dynamic mechanical analyzer (DMA) and a differential scanning calorimeter (DSC). FTIR spectra showed that the polyesterification rate in ESO/MHHPA/EMI systems increased with increasing of the catalyst concentration. DSC thermograms indicated that EMI-catalyzed ESO/MHHPA systems experienced enthalpy relaxation at low EMI concentration whereas the extent decreased with increasing of the EMI concentration. There is a direct relationship between the degree of conversion and crosslink density of the thermal cured ESO/MHHPA/EMI systems with EMI concentration. The curing characteristics of thermal curable ESO thermosetting resins were found to have influence on the thermal properties of the ESO systems. It was determined that the glass transition temperature (T g) and storage modulus (E′) of cured ESO increased with increasing the EMI concentration whereas the damping properties of the ESO/MHHPA/EMI systems exhibited the reverse trend. It was found that the thermally curable ESO thermosetting resins experienced a two-stage thermal decomposition process.  相似文献

Epoxidized soybean oil (ESO)/diglycidyl ether of bisphenol A (DGEBA) in various blend ratios (i.e. 100/0, 90/10, 80/20, 70/30, 60/40, 50/50) was thermally cured using methylhexahydrophthalic anhydride in the presence of 2‐ethyl‐4‐methylimidazole catalyst. The tensile properties and fracture toughness of the ESO/DGEBA thermoset blends were determined. Thermal properties of the blends were characterized using dynamic mechanical analysis, differential scanning calorimetry and thermogravimetric analysis. Blending of ESO and DGEBA gave synergistic effects on the modulus, strength, glass transition temperature and thermal stability. However, the fracture toughness and elongation at break of ESO/DGEBA blends are lower than those of ESO, as expected. The enhancement in certain mechanical and thermal properties of ESO/DGEBA can be associated with the crosslink density, gel content and possible interpenetrating network of the resulting thermoset blends. © 2013 Society of Chemical Industry  相似文献

Although double‐skin façade (DSF) is an environmental‐friendly architectural feature, its fire behaviour is a deep concern. The interior glass system including the glass pane, metal frame and associated accessories will be hotter than the exterior glass system as demonstrated by earlier studies. The glass pane above the fire room will be broken to spread flame into the upper compartment. Aprons are proposed to protect the air cavity of DSF in a way similar to those outside a single‐skin façade. In this paper, the effect of aprons in protecting against fire spread from an underlying compartment to the compartments above by preventing glass breakage of the inner glass pane was studied. Fire and smoke from a post‐flashover room fire adjacent to the DSF would be trapped in the air cavity between the two glass panes. Spreading of hot gases with different apron widths was studied by numerical simulations with CFD first. Fire environment with and without breaking the apron immediately above the fire room was studied. Full‐scale burning tests on part of an experimental DSF rig were then carried out to demonstrate the performance of horizontal apron in the DSF rig of 6 m tall and air cavity depth of 2 m with different apron widths.
