A newly developed comparative molecular field analysis (CoMFA) technique, the cross-validated r2-guided region selection (CoMFA/q2-GRS) method, has been used to build a quantitative structure-activity relationship (3D-QSAR) for nonsteroidal estrogen receptor (ER) ligands. Ligands included in this study belong to a series of diethylstilbestrol (DES) and indenestrol analogues whose affinities for the mouse ER (mER) have been determined in our laboratory. The final model utilized 30 compounds and yielded a q2GRS (cross-validated r2, guided region selection) of 0.796, as compared to a q2 of 0.720 for conventional CoMFA, with a standard error of prediction of 0.594 at 3 principal components. This model was used to visualize steric and electrostatic features of the ligands that correspond with ER binding affinity. Results obtained from the CoMFA steric and electrostatic plots of this model have also been compared to information from the ER binding affinities of substituted estradiol analogues. This is in an effort to determine structural features of compounds in the CoMFA analysis that may correspond to those of the estradiol analogues and to further clarify the mode of binding of nonsteroidal ER ligands.  相似文献

We have measured the room-temperature lasing behavior of NdP5OI4(NdPP) transverselypumped at 058-wm wavelength by a CW dye laser in an excitation geometrysimulatingthat of an LED or laser diode. For acrystal 760 Nm long, the lowest transversethreshold was 4 mW. From thisthresholdandatheoreticalrelationshipbetween excitation efficiency andpump wavelength, we predict that a threshold of approximately 8.5 mW should be possible for excitation by an edge-emitting 0.8-wm wavelength LED. Usingthin NdP P samples with reflecting back surfaces for better excitation efficiency should lower this threshold by -60 percent, andgiveminimumoptical-to-optical con- version efficiencies of - 3.5 percent.  相似文献

Oral administration of high dosages of the dithiocarbamate pesticides maneb and mancozeb was teratogenic in rats but not in mice. The malformations, severe limb and craniofacial defects, were pronounced after maneb treatment but less so after mancozeb and propineb, zinc-containing compounds. The teratogenic effect of maneb was progressively reduced by simultaneously administering increasing amounts of zinc acetate. The mechanism of the teratogenic effect may involve the compounds being chelating agents, trapping zinc required for many important enzyme systems.  相似文献

Ras proteins are membrane-associated guanine nucleotide-binding proteins that serve as molecular switches for signal transduction pathways in a diverse array of organisms. Various cellular factors are known to interact with Ras proteins. In order to find the novel cellular factors that are associated with Ras function, we have constructed synthetic lethal mutants of the ras1+ gene in Schizosaccharomyces pombe and used them to identify the genes that are functionally dependent on the Ras1. We first constructed S. pombe strains in which chromosomal ras1+ gene is placed under the nmt1 promoter that is regulated by thiamine. This strain shows ras1+ phenotype in the absence of thiamine, whereas it shows ras1- phenotype in the presence of thiamine. Second, we mutated the constructed strains with ultraviolet light (UV) and selected two synthetic lethal mutants that could not grow when Ras1 function was repressed (ras1-). One of the mutants, KSC3, showed a swollen cell shape, aberrant deposition of septum materials, and aberrant nuclei. The other mutant, KSC4, showed sensitivity to hyper-osmolarity when Ras1 function is absent. These mutants, however, grow normally when Ras1 is expressed (ras1+). These two novel synthetic lethal mutants of ras1 provide the means to isolate the corresponding genes that function in association with Ras1 in S. pombe. Screening of a genomic library of S. pombe complementing the mutant phenotype allowed us to identify several novel genes associated with Ras1 of S. pombe.  相似文献

In this study the desizing, scouring, bleaching and slack mercerising of cotton fabric by a one–step process at different sodium hydroxide concentrations, impregnation temperatures and curing times were examined. The results indicate that when the fabrics are mercerised at lower temperature, the strength retention and colour strength are enhanced. When the sodium hydroxide concentration is increased, these properties also increase. When cotton fabrics are mercerised for 3 min at 40C and cured for 30 s at 120C, their physical properties are similar to those obtained using a conventional two–step approach.  相似文献
