This study evaluated the direct effect of a phytochemical, hesperidin, on pre-osteoblast cell function as well as osteogenesis and collagen matrix quality, as there is little known about hesperidin’s influence in mineralized tissue formation and regeneration. Hesperidin was added to a culture of MC3T3-E1 cells at various concentrations. Cell proliferation, viability, osteogenic gene expression and deposited collagen matrix analyses were performed. Treatment with hesperidin showed significant upregulation of osteogenic markers, particularly with lower doses. Mature and compact collagen fibrils in hesperidin-treated cultures were observed by picrosirius red staining (PSR), although a thinner matrix layer was present for the higher dose of hesperidin compared to osteogenic media alone. Fourier-transform infrared spectroscopy indicated a better mineral-to-matrix ratio and matrix distribution in cultures exposed to hesperidin and confirmed less collagen deposited with the 100-µM dose of hesperidin. In vivo, hesperidin combined with a suboptimal dose of bone morphogenetic protein 2 (BMP2) (dose unable to promote healing of a rat mandible critical-sized bone defect) in a collagenous scaffold promoted a well-controlled (not ectopic) pattern of bone formation as compared to a large dose of BMP2 (previously defined as optimal in healing the critical-sized defect, although of ectopic nature). PSR staining of newly formed bone demonstrated that hesperidin can promote maturation of bone organic matrix. Our findings show, for the first time, that hesperidin has a modulatory role in mineralized tissue formation via not only osteoblast cell differentiation but also matrix organization and matrix-to-mineral ratio and could be a potential adjunct in regenerative bone therapies. 相似文献

Oxidative destruction of perfluorooctane sulfonate using boron-doped diamond film electrodes 总被引:1,自引:0,他引:1
This research investigated the oxidative destruction of perfluorooctane sulfonate at boron-doped diamond film electrodes. Experiments measuring oxidation rates of PFOS were performed over a range in current densities and temperatures using a rotating disk electrode (RDE) reactor and a parallel plate flow-through reactor. The oxidation of PFOS yielded sulfate, fluoride, carbon dioxide, and trace levels of trifluoroacetic acid. Reaction rates in the RDE reactor were zeroth order in PFOS concentration. Reaction rates in the flow-through reactor were mass-transfer-limited and were pseudo-first-order in PFOS concentration, with a half-life of 5.3 min at a current density of 20 mA/cm2. Eyring analysis of the zeroth order rate constants at a fixed electrode potential yielded an apparent activation energy of 4.2 kJ/mol for PFOS oxidation. Density functional theory (DFT) simulations were used to calculate activation barriers for different possible reaction mechanisms, including oxidation by hydroxyl radicals at different sites on the PFOS molecule, and direct electron transfer. A comparison of the experimentally measured apparent activation energy with those calculated using DFT indicated that the most likely rate-limiting step for PFOS oxidation was direct electron transfer. 相似文献

Dreissenid (zebra and quagga) mussels are widely recognized as having strong, adverse ecological and economic impacts, e.g., biofouling and loss of water column primary production. We assessed perceptions and values associated with two less often considered ecological outcomes of dreissenid mussel influences on coastal ecosystems along Lake Ontario and the western St. Lawrence River in New York State. One, the generation of water clarity through filtration, we define as an ecosystem service; the other, the production of large amounts of nuisance algae (e.g., Cladophora and Microcystis) is defined as an ecosystem disservice. Surveys of business owners and homeowners quantified their preferences and the formation of values regarding these products of zebra mussel influence. Water clarity increased greatly, particularly in the eastern portion of Lake Ontario, and algal problems increased throughout. Businesses attributed increases and decreases in revenues associated with water clarity and algae; homeowners reported analogous changes in property values. Water clarity was positively associated, and algae negatively associated, with changes in revenues and property values. Threshold responses of costs as functions of filamentous algae were evident. Given the likely continued influx of invasive species due to human activities, further development of the ecosystem service concept should consider potential “goods” and “bads” of invasives and their influence on ecosystem and social system resiliency. 相似文献

Improvements in methods for disinfecting fresh-cut cantaloupe could reduce spoilage losses and reduce the risk of foodborne illness from human pathogen contamination. The objective of this study was to investigate the feasibility of using hot-water treatment in combination with low-dose irradiation to reduce native microbial populations while maintaining the quality of fresh-cut cantaloupe. Whole cantaloupes were washed in tap water at 20 or 76 degrees C for 3 min. Fresh-cut cantaloupe cubes, prepared from the washed fruit, were then packaged in clamshell containers, and half the samples were exposed to 0.5 kGy of gamma radiation. Native microflora populations and sensory qualities were evaluated during the subsequent 7 days of storage at 4 degrees C. The hot-water surface pasteurization reduced the microflora population by 3.3 log on the surface of whole fruits, resulting in a lower microbial load on the fresh-cut cubes compared with cubes cut from fruit treated with cold water. Irradiation of cubes prepared from untreated fruit to an absorbed dose of 0.5 kGy achieved a low microbial load similar to that of cubes prepared from hot-water-treated fruit. The combination of the two treatments was able to further reduce the microflora population. During storage, the headspace atmosphere of the packages was not significantly influenced by any of the treatments. Color, titratable acidity, pH, ascorbic acid, firmness, and drip loss were not consistently affected by treatment with irradiation, hot water, or the combination of the two. Cubes prepared from hot-water-treated whole fruit had slightly lower soluble solids content. The combination of hot-water pasteurization of whole cantaloupe and low-dose irradiation of packaged fresh-cut melon can reduce the population of native microflora while maintaining the quality of this product. 相似文献

The present study used perspectives from the general literature on college alcohol consumption to examine mediational influences of peer, environmental, and parental variables on heavy drinking for student athlete and nonathlete samples. Eight hundred thirty-five freshmen who differed in organized sports involvement were compared on heavy drinking outcomes, peer norms, environmental influences, and parental communication. College athletes reported significantly more heavy drinking experiences than nonathletes. Peer norms, environmental influences, and parental communication were all significant mediators of the athlete-heavy drinking relationship. Athletes reported a higher perception of peer drinking, peer approval of drinking, higher alcohol availability, and direct drink offers, which, in turn, were related to higher rates of heavy drinking. Parental communication mediated the athlete-heavy drinking relationship differently, depending on the specific topic of conversation. Discussion surrounding the importance of incorporating a variety of interventions aimed at reducing collegiate athlete drinking on the basis of the peer, environmental, and parental influences observed in the present analyses are presented. This research examines the application of ultrasonic Rayleigh surface waves to the non-destructive evaluation (NDE) of asphaltic concrete in the frequency range 40–100 kHz. Two ultrasonic parameters, the phase velocity and amplitude attenuation, are considered to be sensitive to local compositional and structural variations and are measured. A wedge technique is used to generate Rayleigh surface waves in an asphaltic concrete beam with uniformly distributed aggregate, and this technique is shown to be effective in launching Rayleigh waves in this highly viscoelastic (absorptive) and heterogeneous medium. Three different ultrasonic detection setups using contact and non-contact transducers as receivers are examined and their results are compared. The experimental results show that the wedge generation technique along with an air-coupled receiving transducer with a finite-size aperture is the most reliable for characterizing Rayleigh waves in asphaltic concrete. It is also experimentally demonstrated that the proposed setup may be used to detect aggregate segregation in asphaltic concrete. 相似文献

Although the interactions of cell-penetrating peptides (CPPs) with mammalian cells have been widely studied, much less is known about their interactions with fungal cells. To study how the properties of CPPs affect translocation into fungal cells, we designed variants of the peptides pVEC and SynB with altered levels of charge and hydrophobicity and evaluated the translocation of the variants into the important human fungal pathogen Candida albicans. Charge played a greater role in translocation efficacy of the peptides than hydrophobicity, with a higher net positive charge leading to higher level of translocation into C. albicans and a higher level of cytosolic localization. Hydrophobicity had little effect on translocation efficacy, but a low level of hydrophobicity did lead to increased vacuolar localization and an energy-dependent translocation mechanism. Our results suggest that CPPs can be designed for desired levels of cargo delivery into fungal cells and for desired translocation mechanisms. 相似文献
