The N-methyl-{d}-aspartate (NMDA) subtype of the excitatory amino acid receptor has been implicated in several kinds of learning and memory, as well as in long-term potentiation (LTP), a putative cellular mechanism for learning and memory. This experiment examined the role of the NMDA receptor in patterned single-alternation (PSA) learning in preweanling rats following intraperitoneal injections of 0.05 mg/kg MK-801, a selective NMDA antagonist. MK-801 significantly inhibited PSA at both 60-sec and 30-sec intervals (ITIs). and attenuated, but did not block Iearning at 8-sec . These results are compared with effects on PSA, a form of nonspatial, memory-based learning, observed; after early postnatal exposure to alcohol, infant hippocampal lesions, and infant exposure to X-irradiation, and they add strongly to these earlier demonstrartions of the role of the hippocampus in learning and memory that is clearly nonspatial and non-cognitive-map-related. No chilling effect and higher recovery are the two chief advantages which have turned steel producers toward exothermic ferroalloys—a blend of an oxidizing agent, a reducing agent, and a conventional ferroalloy. The reducing and the oxidizing agents, upon contact with the molten metal, produce sufficient heat to deliver the ferroalloy in molten state. Satisfaction has been such that use of exothermic ferroalloys has greatly multiplied since exothermic ferrochrome was introduced in 1939 by the Chromium Mining & Smelting Corp.Exothermic ferromanganese has found the broadest applications of this group of alloys and was the subject of a special session at this year’s National Open Hearth Conference. Social housing of preweaned calves can benefit their welfare, but housing and cross sucking are potential barriers to adoption for farmers. For farms using outdoor hutches, an option is to pair adjacent hutches with a shared fence. Our objective was to investigate milk- or starter-feeding strategies to mitigate cross sucking in this system. Holstein heifers were housed in pairs (n = 32 pairs) and divided into 4 treatments (n = 8 pairs each) in a 2 × 2 factorial design: milk in an open bucket with starter in only a bucket (OB-SB), milk in an open bucket and starter in both a bucket and a specialized teat bottle (Braden bottle; OB-BB), milk in a slow-flow teat bucket with starter in only a bucket (TB-SB), or milk in a slow-flow teat bucket and starter in both a bucket and a specialized teat bottle (TB-BB). When starter was first offered (d 6 ± 1 of life, mean ± SD), calf latency to approach was recorded, averaged within pairs, and compared between starter treatments using a linear mixed model with fixed effect of treatment and random effect of pair within treatment. Calves were initially bottle fed; milk treatments began on d 14 ± 1 of life and ended when calves were completely weaned (d 53 ± 1). Calves were observed for behaviors such as drinking milk and cross sucking twice weekly for 30 min during the afternoon milk meal using continuous video, with values averaged within pairs. Linear mixed models were run separately before (wk 3–6) and after (wk 7–8) weaning, with fixed effects of milk- and starter-feeding treatments, week, and the 2- and 3-way interactions, with week as repeated measure and pair as subject. Pairs with Braden bottles and buckets approached starter sooner than those with only buckets (Braden bottles vs. no Braden bottles: 13.1 ± 6.1 vs. 33.2 ± 6.1 min, LSM ± SEM). Before weaning, pairs with open buckets for both milk and starter cross sucked for at least twice as long (OB-SB: 2.9 ± 0.3 min) as all other treatments (OB-BB: 1.5 ± 0.3 min; TB-SB: 0.4 ± 0.3 min; TB-BB: 0.5 ± 0.3 min). This pattern held during weaning, when cross sucking increased overall (OB-SB: 3.9 ± 0.4 min; OB-BB: 1.8 ± 0.4 min; TB-SB: 0.9 ± 0.4 min; TB-BB: 1.6 ± 0.4 min). Regardless of starter treatment, calves spent less time cross sucking when fed milk in teat buckets, which extended the milk meal relative to open buckets (teat bucket vs. open bucket: preweaning = 7.0 ± 0.2 vs. 1.6 ± 0.2 min; weaning = 3.0 ± 0.1 vs. 0.6 ± 0.1 min). When calves are fed milk in open buckets, a novel option for reducing cross sucking is to provide starter through a specialized bottle. Nonetheless, providing milk in slow-flow teat buckets had the greatest effect on reducing cross sucking by directing suckling to the teat instead of another calf or pen objects.  相似文献

Unique high mass negative ions in the -200 to -400 mass/charge range with repetitive spacings of 12, 14, and 16 units, representative of oligomeric species, have been detected in single ambient submicrometer aerosol particles using real-time single-particle mass spectrometry during the Study of Organic Aerosols field campaign conducted in Riverside, CA (SOAR) in August and November 2005. These oligomer-containing particles represented 33-40% of the total detected particles and contained other indicators of aging including oxidized organic carbon, amine, nitrate, and sulfate ion markers. Overall, the highest mass oligomeric patterns were observed in small acidic 140-200 nm particles in the summer. Also during the summer, increased oligomer intensities were observed when the particles were heated with a thermodenuder. We hypothesize that heat removed semivolatile species, thereby increasing particle acidity, while concentrating the oligomeric precursors and accelerating oligomer formation. Differences in oligomer behavior with respect to particle size and heating can be attributed to seasonal differences in photochemical oxidation, the relative amount of ammonium, and particle acidity.  相似文献

Thermoduric bacteria (TDB), including sporeformers and their spores, can be present in milk and dairy products even after pasteurization. They have the potential to adversely affect the quality and shelf life of products. The objectives of this study were to identify the origin and common species of heat-resistant bacteria occurring during summer and winter on Midwest dairy farms. Bulk tank milk samples were taken from 10 dairy farms located along the South Dakota section of Interstate 29, with herd sizes ranging from 650 to 3,500 lactating dairy cows. Milk samples were profiled for the prevalence of TDB and spore counts (SC). Corn silage samples and swabs of the milking clusters were also taken at the dairies to further profile the potential sources of TDB and SC. The samples were taken 3 times during 2 seasons [winter (January–March) and summer (June–August)] to track seasonal changes in the farm bacterial flora. During winter, the average TDB counts in bulk tank milk were 2.61 log compared with 2.76 log TDB counts in the summer. The SC was 1.08 log in the winter, which was half the 2.06 log SC present in the summer season. Corn silage sampled in winter contained a 7.57 log TDB count compared with an increased 10.77 log TDB count during summer sampling. Concentrations of SC in corn silage reached an average of 6.3 log in winter compared with 11.81 log for summer. The seasonal effect was evident with an increase in summer counts across the board for TDB and SC, both in the feed and bulk tank milk samples. Bacillus licheniformis was the predominant species identified in 62.4% of winter (85 total) and 49.4% of summer (83 total) samples. Bacillus subtilis made up 9.4% of the remaining winter isolates, followed by Bacillus sonorensis at 8.2%. Conversely, B. sonorensis made up 12% of the summer isolates followed by Bacillus pumilus at 10.8%. Bacillus licheniformis is a ubiquitous microbe and was isolated from both TDB and sporeformer categories in all 3 sample types. There were larger increases in SC than TDB, indicating that summer temperatures and conditions may favor proliferation of sporeforming bacteria over that of TDB. In conclusion, samples from bulk tank milk, milking cluster swabs, and corn silage samples at each of the 10 sites indicated that B. licheniformis was the major contaminant species, regardless of season. In this experiment, corn silage was the major environmental source of both TDB and SC with higher concentrations in summer when compared with winter.  相似文献

In vitro maturation of oocytes has enormous potential in assisted reproductive technology, but its use has been limited due to insufficient knowledge of oocyte physiology during this dynamic period and lack of an adequate maturation system. The aim of this study was to characterize the metabolic profiles of three groups of oocytes throughout maturation: cumulus-oocyte complexes (COCs), denuded oocytes, and denuded oocytes co-cultured with cumulus cells. Mouse oocytes were collected from 28-day-old unstimulated females and matured in a defined medium. Oocytes were matured individually and transferred into fresh 0.5 microl drops of medium at 4 h intervals until 16 h. Ultramicrofluorimetry was used to quantitate carbohydrate consumption from and metabolite release into the medium. Glucose consumption and lactate production of COCs increased (P < 0.001) over the maturation interval (0-16 h). Glucose consumption by COCs that subsequently fertilized was higher between 8-12 h of maturation than by COCs that did not fertilize (38 versus 29 pmol/COC per h, respectively; P < 0.01). Lactate production by COCs that subsequently fertilized was higher between 8-16 h of maturation, than by oocytes that did not fertilize (8-12 h, 66 versus 46 pmol/COC per h, P < 0.01; 12-16 h, 56 versus 40 pmol/COC per h, respectively; P < 0.05). These data indicate that the final hours of maturation may hold a unique marker of oocyte competence, as during this time fertilizable COCs take up more glucose and produce more lactate than those not subsequently fertilized.  相似文献

Anaerobic degradation of cis-1,2-dichloroethene (cis-1,2-DCE) and 1,2-dichloroethane (1,2-DCA) was studied in microcosms derived from a laboratory-scale upflow treatment wetland system used to biodegrade chlorinated compounds present in groundwater from a Superfund site. Dechlorination kinetics of cis-1,2-DCE (0.94-1.57 d(-1)) and 1,2-DCA (0.15-0.71 d(-1)) were rapid, and degradation proceeded to completion with ethene or ethane as terminal dechlorination products. Hydrogen concentrations, measured simultaneously during dechlorination, were significantly different for the two compounds, approximately 2.5 nM for cis-1,2-DCE and 38 nM for 1,2-DCA. Methanogenesis proceeded during the degradation of 1,2-DCA when H2 concentrations were high but not during the dechlorination of cis-1,2-DCE when H2 concentrations were below published thresholds for methanogenesis. A 16S rRNA gene-based approach indicates that microorganisms closely related to Dehalococcoides ethenogenes were present and that they were distributed throughout the bottom, middle, and top of the upflow treatment wetland system. These results coupled with consideration of hydrogen thresholds, degradation kinetics, daughter products, and measurements of methanogenesis strongly suggest that halorespirers were responsible for dechlorination of cis-1,2-DCE and that 1,2-DCA dechlorination was co-metabolic, likely mediated by acetogens or methanogens. Rapid dechlorination potential was distributed throughout the wetland bed, both within and below the rhizosphere, indicating that reductive dechlorination pathways can be active in anaerobic environments located in close spatial proximity to aerobic environments and plants in treatment wetland systems.  相似文献

In order to investigate the distributions and speciation of (129)I (and (127)I) in a contaminated F-Area groundwater plume of the Savannah River Site that cannot be explained by simple transport models, soil resuspension experiments simulating surface runoff or stormflow and erosion events were conducted. Results showed that 72-77% of the newly introduced I(-) or IO(3)(-) were irreversibly sequestered into the organic-rich riparian soil, while the rest was transformed by the soil into colloidal and truly dissolved organo-iodine, resulting in (129)I remobilization from the soil greatly exceeding the 1 pCi/L drinking water permit. This contradicts the conventional view that only considers I(-) or IO(3)(-) as the mobile forms. Laboratory iodination experiments indicate that iodine likely covalently binds to aromatic structures of the soil organic matter (SOM). Under very acidic conditions, abiotic iodination of SOM was predominant, whereas under less acidic conditions (pH ≥5), microbial enzymatically assisted iodination of SOM was predominant. The organic-rich soil in the vadose zone of F-Area thus acts primarily as a "sink," but may also behave as a potentially important vector for mobile radioiodine in an on-off carrying mechanism. Generally the riparian zone provides as a natural attenuation zone that greatly reduces radioiodine release.  相似文献
