Many companies are using Just-In-Time (JIT) control systems in their mixed-model multi-level production facilities. When scheduling these facilities the most important objective is to keep a constant rate of usage for every part used by the system. In this paper a mathematical model for scheduling these facilities is studied and techniques for determining good schedules are developed. There is now strong evidence that the chorioretinal degeneration associated with ornithine-delta-aminotransferase (OAT) deficiency is a consequence of hyperornithinemia. Therefore development of a metabolic system for clearing ornithine from the circulation is being pursued as a potential treatment. The skin is considered an attractive location for such a metabolic system because autologous cells can be safely and easily utilized. This study was undertaken to determine the ornithine metabolizing capacity of epidermal keratinocytes expressing normal and superphysiologic amounts of OAT. The data show that overexpression of OAT in keratinocytes cultured from a gyrate atrophy patient restores ornithine metabolism and results in a rate of ornithine disappearance from the medium that is significantly higher than the rate of disappearance from the medium bathing normal keratinocytes. In addition, OAT activity determined in soluble protein prepared from sonicates suggests that the capacity to maintain plasma ornithine within the normal range is contained within an accomplishable graft of keratinocytes overexpressing OAT. However, the actual rate of ornithine disappearance from the media was significantly less than predicted from enzyme activity assays. Following ornithine metabolite production by intact cells suggests that ornithine metabolism is limited primarily by clearance of downstream metabolites, as opposed to substrate delivery.  相似文献

The subcellular events responsible for release of mediators by mast cells may help to clarify roles for mast cells in health and disease. In this study we show that the granule-associated protease chymase is also within cytoplasmic vesicles in appropriately stimulated rat peritoneal mast cells. Rat peritoneal mast cells were recovered before or 1-10 sec after exposure to the secretogogue compound 48/80 (10 micrograms/ml) and then were examined by radioimmunoassay to quantify histamine release or were processed, using routine methods for postembedding immunoelectron microscopy, to identify the subcellular localization of chymase. In comparison to unstimulated cells, compound 48/80 stimulated cells in two independent experiments showed an increase (15%, 28%) in the surface area of the cell and a decrease (12%, 6%) in the surface area of the total granule compartment before degranulation channel formation. These global cellular changes occurred in a background of transient but significant (p < 0.01) increases in the area and number of chymase-immunoreactive vesicles per microns2 cytoplasm. These changes were detectable at 5 or 7 sec after stimulation with compound 48/80 but returned to near prestimulation levels by 9 or 10 sec after addition of compound 48/80 (total cumulative histamine release was 28% by 8 sec and 47% by 14 sec). These observations suggest that vesicles participate in the early stages of regulated secretion of chymase from rat peritoneal mast cells.  相似文献

AIMS/BACKGROUND: To characterise clinically a large kindred segregating retinitis pigmentosa and sensorineural hearing impairment in an autosomal dominant pattern and perform genetic linkage studies in this family. Extensive linkage analysis in this family had previously excluded the majority of loci shown to be involved in the aetiologies of RP, some other forms of inherited retinal degeneration, and inherited deafness. METHODS: Members of the family were subjected to detailed ophthalmic and audiological assessment. In addition, some family members underwent skeletal muscle biopsy, electromyography, and electrocardiography. Linkage analysis using anonymous microsatellite markers was performed on DNA samples from all living members of the pedigree. RESULTS: Patients in this kindred have a retinopathy typical of retinitis pigmentosa in addition to a hearing impairment. Those members of the pedigree examined demonstrated a subclinical myopathy, as evidence by abnormal skeletal muscle histology, electromyography, and electrocardiography. LOD scores of Zmax = 3.75 (theta = 0.10), Zmax = 3.41 (theta = 0.10), and Zmax = 3.25 (theta = 0.15) respectively were obtained with the markers D9S118, D9S121, and ASS, located on chromosome 9q34-qter, suggesting that the causative gene in this family may lie on the long arm (q) of chromosome 9. CONCLUSIONS: These data indicate that the gene responsible for the phenotype in this kindred is located on chromosome 9 q. These data, together with evidence that a murine deafness gene is located in a syntenic area of the mouse genome, should direct the research community to consider this area as a candidate region for retinopathy and/or deafness genes.  相似文献

Capacity and power control issues are considered for urban CDMA microcells that operate on channels characterized by log-normal shadowing and Nakagami fading. The effects of fading on the desired and interfering signals, variations in the shadow standard deviation, imperfect power control, and the requiredE b /N o for satisfactory link operation are investigated. It is shown that the reverse channel is relatively insensitive, and the forward channel very sensitive, to shadow and fading variations. Power control error is shown to significantly degrade reverse channel capacity. Twobiased forward channel power control strategies are introduced to enhance existing unbiasedbalancing forward channel power control algorithms.This research has been supported by BNR Inc.  相似文献

A method for predicting the long-term performance of solar energy systems, based on the analysis of system performance for one particular day—the typical meteorological day (TMD)—is presented. The TMD is constructed from the cumulative time distribution of insolation values on the collector aperture. The TMD method requires little calculational effort and a small data base relative to standard yearly computer simulations. Good agreement is found between the predictions of the new method and the corresponding results of the -f-chart method. The TMD method is of particular value for cases that may often be treated inaccurately by simple calculational methods: (1) high threshold problems; (2) systems with short response times (e.g., due to small storage); and (3) systems in which collectors other than flat plates are used (the method is applicable to all solar collector types).  相似文献

A structured in-depth interview employing standardized criteria was used to determine the prevalence of lifetime and current alcohol dependence (alcoholism) in unselected consecutive patients admitted to a regional Level I trauma center. Of 629 patients, 157 (25.0%) were current alcoholics at the time of injury. An additional 87 (13.8%) were diagnosed as lifetime non-current alcoholics. There was no significant difference in the rates of current alcohol dependence among patients injured in vehicular crashes (23.5%), other unintentional trauma victims (29.3%), and those injured as a result of violence (24.6%). Of BAC + (blood alcohol concentration positive) patients, 54.5% were current alcoholics. However, 14.4% of alcohol-negative patients were also diagnosed as alcohol dependent.  相似文献

Walking without vision results in veering, an inability to maintain a straight path that has important consequences for blind pedestrians. In this study, the authors addressed whether the source of veering in the absence of visual and auditory feedback is better attributed to errors in perceptual encoding or undetected motor error. Three experiments had the following results: No significant differences in the shapes of veering trajectories were found between blind and blindfolded participants; accuracy in detecting curved walking paths was not correlated with simple measures of veering behavior; and explicit perceptual cues to initial walking direction did not reduce veering. The authors present a model that accounts for the major characteristics of participants' veering behavior by postulating 3 independent sources of undetected motor error: initial orientation, consistent biases in step direction, and, most important, variable error in individual steps.
