We propose a novel type of immunosensor based on the diffraction of surface plasmon, which allows for in situ, real-time, and label-free observation of interfacial binding events. The inherent self-referencing mechanism of surface diffraction was found to be very effective for compensating fluctuations of the bulk, demonstrated by a temperature variation experiment. Possessing a stable baseline signal, the diffraction sensor offered picomolar sensitivity in directly detecting the binding of human chorionic gonadotropin (hCG) hormone. 相似文献

Digital methods to correct for imperfections in gamma ray cameras are reviewed. The two principal problems currently addressed are nonlinearities in the analog position signals, and variations with position in the magnitude of the energy signal. As an example of real-time processing to correct for these imperfections, the operation of the Medical Data Systems image preprocessor is outlined. Variable density random sampling patterns have recently become increasingly popular for accelerated imaging strategies, as they lead to incoherent aliasing artifacts. However, the design of these sampling patterns is still an open problem. Current strategies use model assumptions like polynomials of different order to generate a probability density function that is then used to generate the sampling pattern. The activity of ceramic honeycombs with varying material composition and degree of utilization is compared on a suitable model gas in a test plant. The tested honeycombs are standard products that have not been surface‐treated in any way. A propane‐air mixture was fed to the test plant and then oxidized in a high‐temperature furnace. The analysis of the conversion rate over the reaction temperature showed remarkable differences in activity. The honeycomb with the most promising results was further investigated with the exhaust gas from the drying process of a waste treatment plant. Fluorogenic oligonucleotide probes allow mRNA imaging in living cells. A key challenge is the cellular delivery of probes. Most delivery agents, such as cell-penetrating peptides (CPPs) and pore-forming proteins, require interactions with the membrane. Charges play an important role. To explore the influence of charge on fluorogenic properties and delivery efficiency, we compared peptide nucleic acid (PNA)- with DNA-based forced intercalation (FIT) probes. Perhaps counterintuitively, fluorescence signaling by charged DNA FIT probes proved tolerant to CPP conjugation, whereas CPP–FIT PNA conjugates were affected. Live-cell imaging was performed with a genetically engineered HEK293 cell line to allow the inducible expression of a specific mRNA target. Blob-like features and high background were recurring nuisances of the tested CPP and lipid conjugates. By contrast, delivery by streptolysin-O provided high enhancements of the fluorescence of the FIT probe upon target induction. Notably, DNA-based FIT probes were brighter and more responsive than PNA-based FIT probes. Optimized conditions enabled live-cell multicolor imaging of three different mRNA target sequences. 相似文献

To clarify the value of the serum inorganic phosphate concentration in the diagnosis of ischemic bowel disease, a retrospective study of 24 patients with various causes of intestinal ischemia was carried out. Only 25 percent of the patients had elevations of their serum phosphate concentrations. These patients had the combination of extensive bowel injury, acute renal insufficiency, and acidosis. Mortality was significantly increased in these patients. Thus, the serum phosphate concentration was not a sensitive indicator of ischemic bowel disease, but elevations did predict extensive injury and poor prognosis. 相似文献

We report the observation of live-cell dynamics by noncontact scanning near-field optical microscopy (SNOM) modified to work with living biological samples that are fully immersed in liquid. We did not use the SNOM setup in strictly near-field conditions (we used 1-mum constant-height mode); however, we could examine the dynamics of rhythmically beating cardiac myocytes in culture with extremely high vertical sensitivity below the nanometric range. We could halt scans at any point to record localized contraction profiles of the cell membrane. We show that the contractions of the organisms changed shape dramatically within adjacent areas. We believe that the spatial dependency of the contractions arises because of the measurement system's ability to resolve the behavior of individual submembrane actin bundles. Our results, combining imaging and real-time recording in localized areas, reveal a new, to our knowledge, noninvasive method for using SNOM setups for studying the dynamics of live biological samples. 相似文献
