Allograft rejection is associated with infiltration of inflammatory cells and deposition of extracellular matrix proteins. The extent to which diversity in the extracellular matrix regulates inflammatory cell function in transplants remains unclear. One group of extracellular matrix proteins, termed fibronectins (FNs), exhibits inherent diversity as a consequence of alternative splicing in three segments: EIIIA, EIIIB, or V. Although the EIIIA segment has documented functions in mesenchymal cell differentiation, neither this segment nor the EIIIB segment have been tested for effects specific to leukocyte functions. By contrast, the V region can include the CS-1 segment to which leukocytes may adhere through alpha 4 beta 1 integrins. In this study, we demonstrate that EIIIA+, EIIIB+, and V+ FN variants are synthesized, primarily by macrophages in distinct temporal and spatial patterns in two rat cardiac transplant models: either with antigenic challenge, allografts, or without challenge, isografts. The ratio of EIIIA inclusion into FN increases by day 1 in allografts and isografts and remains high until allografts are rejected (approximately 7 days) but falls to normal levels in tolerated isografts (day 6). EIIIB+ FN ratios in allografts peak later than do EIIIA+ FNs (day 4). EIIIB+ FN ratios remain relatively low in isografts. Interestingly, EIIIA+ and EIIIB+ FNs are deposited prominently in the myocardium of rejecting allografts in close association with infiltrating leukocytes, and FN expression and deposition are prominent at sites of infarction. By contrast, these FNs are largely restricted to the epicardium and to a lesser degree in the immediately adjacent myocardium in isografts. CS-1+ FNs increase in allografts and isografts at 3 hours after transplantation but are particularly prominent in allografts 1 to 3 days before rejection. Our data suggest that FN splicing variants have a differential role in the effector functions of leukocytes in allografts and isografts and provide a foundation for testing their function on leukocytes and a rationale for FN-based therapeutics to modulate allograft rejection in transplant recipients.  相似文献

The tarnishing originated on a hammered gold mask was analysed. Red tarnishing was observed after three years of storage in an indoor environment in the Gold Museum of Banco de la República, Bogotá, Colombia. Silver sulphide (Ag2S) and silver sulphate (Ag2SO4) compounds were identified as the origin of the tarnishing phenomenon, which is attributed to environmental contamination. Baseline concentration levels of As, B, Ca, Cd, Cr, Cu, Co, Fe, Mg, Mn, Mo, Ni, P, Pb, Se, V, and Zn were determined for Porphyra columbina and Ulva sp. collected from three locations along San Jorge Gulf, in Patagonia Argentina. Elements were quantified by inductively coupled plasma-optical emission spectrometry, with the exception of lead and cadmium in some samples which were determined by electrothermal atomic absorption spectrometry. Three stations with different exposure degree to human activities, Bahía Solano, the mouth of Arroyo La Mata stream and Punta Maqueda, were selected as sampling points. The results showed a wide range of metal retention capacity between the two studied species. Regarding the levels of pollutants found in the researched sites, Punta Maqueda seemed to be less influenced by anthropogenic activities than the other two sites except for Cd. Taking into account their toxicities seasonal variations in Pb and Cd levels were studied in both algae in Punta Maqueda. Maximum concentrations of Cd (9.8 microg g(-1) dry wt.) were observed in P. columbina during winter, and maximum levels of Pb (0.82 microg g(-1) dry wt.) were detected in Ulva sp. during summer. We evaluated ethanol- and HCl-induced mucosal damages in developing rats. The degree of damage induced by ethanol and HCl was greatest in 1-week-old rats and decreased significantly with age until 4 weeks; thereafter it increased again. To evaluate the effect of weaning on the maturational changes in mucosal defense, we compared ethanol-induced mucosal damage among three groups of newborn rats: (1) receiving milk only; (2) receiving only rat chow from 14 days of age, and (3) having free access to milk and chow. There were no significant differences at 18 and 21 days of age. The mucus thickness increased with age until 8 weeks and was not affected by weaning. In conclusion, developmental changes occur in gastric mucosal protection in rats. Weaning does not have a significant effect on these changes.  相似文献

Dioscorea spp. tubers are known commonly as camote de cerro to the state of Jalisco, México. Besides its use as food, it contains diosgenin, a raw material used widely for synthesising steroid hormones. The purpose of the present research was to determine diosgenin contents by gas chromatography‐mass spectrometry (GC‐MS), as well as physical and chemical characteristics of sixty Dioscorea spp. accessions cultivated in mesh shade coming from eleven localities in the state of Jalisco. Extraction protocol for diosgenin was maceration combined with ethanol 80%. Significant statistical differences for diosgenin content were found between accessions, finding levels between 0.02 and 0.16 mg kg?1 in dry basis. Length, weight tuber, proportion pulp/skin, percentage of pulp and percentage of skin showed significant differences. Chemical composition based in fresh weight presented in moisture percentage a range of 71.93–83.26%, crude protein 1.42–1.74%, ashes 0.80–1.16%, lipid 0.13–0.17%, crude fibre 3.80–4.02% and total carbohydrates 13.97–25.44%. Results show that tubers from Jalisco can be an important source of nutrients for consumers.  相似文献
