The function and nature of inhibition of neurons in the visual cortex have been the focus of both experimental and theoretical investigations. There are two ways in which inhibition can suppress synaptic excitation. In hyperpolarizing inhibition, negative and positive currents sum linearly to produce a net change in membrane potential. In contrast, shunting inhibition acts nonlinearly by causing an increase in membrane conductance; this divides the amplitude of the excitatory response. Visually evoked changes in membrane conductance have been reported to be nonsignificant or weak, supporting the hyperpolarization mode of inhibition. Here we present a new approach to studying inhibition that is based on in vivo whole-cell voltage clamping. This technique allows the continuous measurement of conductance dynamics during visual activation. We show, in neurons of cat primary visual cortex, that the response to optimally orientated flashed bars can increase the somatic input conductance to more than three times that of the resting state. The short latency of the visually evoked peak of conductance, and its apparent reversal potential suggest a dominant contribution from gamma-aminobutyric acid ((GABA)A) receptor-mediated synapses. We propose that nonlinear shunting inhibition may act during the initial stage of visual cortical processing, setting the balance between opponent 'On' and 'Off' responses in different locations of the visual receptive field.  相似文献

As we demonstrated earlier, platelets adhere to intact endothelium provided they are activated and convectively transported against the endothelial surface. To identify the platelet receptors involved we superfused cultured endothelium with activated platelet rich plasma (PRP) by means of the Stagnation Point Flow Adhesio- Aggregometer while blocking various platelet receptors. Inhibition was performed with the tetrapeptide RGDS, the non-peptide Ro-43-8857, or a monoclonal antibody directed against integrin alphaIIbeta3. Platelet deposition was video-recorded and quantified by image analysis. Infusion of RGDS or Ro-43-8857 into ADP-stimulated PRP completely prevented adhesion as well as subsequent aggregation. Interrupting the inhibitor infusion while ADP stimulation persisted, prompted adhesion and aggregation, demonstrating the reversibility of the inhibition. Platelet adhesion was irreversibly blocked by preincubation of the PRP with the moab against alphaIIbeta3. Its specific binding was confirmed by immunoelectron microscopy. Our results suggest that platelet adhesion to intact endothelium is mediated via platelet integrin alphaIIbeta3.  相似文献

Before appropriate therapy can be instituted for a cancer patient, the presence and extent of tumor must be evaluated. Deciding which imaging technique to use depends on tumor location, type, and biologic behavior. Conventional radiography provides important information at a relatively low cost compared with other imaging modalities. Ultrasound is a valuable adjunct to radiography, but does not replace it because both imaging modalities provide unique information. Nuclear medicine procedures contribute additional, unique data by providing physiological information, but specificity is lacking. Both CT and MRI provide images with exquisite anatomic detail, but availability and cost prohibit their general use.  相似文献

Male and female subjects (n = 125) were measured on a battery of anthropometric tests every six months for a period of five years. Using maturity status as the temporal component (pubescent assessment stages one through five), proportionality characteristics of subjects were monitored as they matured toward the adult form. Subjects were assigned to one of the Hi, Mid and Lo sub-groups based on their score at PA stage one for each of 15 proportionality characteristics. Subsequently, those in the Mid group were removed from the analysis, while male and female subjects were pooled for the final ANOVA model. The results showed that a number of variables remained stable from PA stages one through five. That is, differences between the Hi and Lo groups were maintained throughout maturation. Predominantly, these included the proportional body segment breadth measures. Proportionality characteristics of the upper limbs appeared to stabilise from mid-adolescence. However, those related to the lower limb were not stable. In other words, a significant difference for the lower limb variables at PA stage one was not maintained through stages four and five. The results raised some doubts as to the suitability of using lower limb proportions as selection criteria in talent identification programs.  相似文献

PURPOSE: To compare the outcome for patients with squamous cell carcinoma of cervical lymph nodes metastatic from an unknown primary site who were irradiated to both sides of the neck and potential mucosal sites with opposed photon beams, and for those irradiated to the ipsilateral side of the neck alone with an electron beam. METHODS AND MATERIALS: Fifty-two patients with squamous cell carcinoma metastatic to cervical lymph nodes from an unknown primary site were irradiated by two different methods. Thirty-six were irradiated with a bilateral technique (BT), i.e., to both sides of the neck, including the naso-oro-hypopharyngeal mucosa, and 16 were irradiated with an electron beam (EB) to the ipsilateral side of the neck alone. Twenty patients of the BT group and 11 of the EB group had cervical lymph node dissections, and the remaining 21 patients had lymph node biopsies, prior to radiotherapy. RESULTS: Tumor control in the ipsilateral side of the neck did not differ for either radiation technique, but was significantly higher after lymph node dissection than after biopsy (90 vs. 48%; p = 0.0004). Control of subclinical metastases in the contralateral cervical lymph nodes was higher for patients irradiated with BT than for patients irradiated with EB (86 vs. 56%; p = 0.03). The occult primary was later discovered in 8% of the patients in the BT group and 44% of the EB group (p = 0.0005). The disease-free survival rate at 5 years for patients who had lymph node dissection prior to irradiation was 61%, and was 37% for those who had biopsy (p = 0.05). Only 20% of patients who subsequently developed an occult primary were salvaged and survived for 5 years after salvage treatment. CONCLUSION: Bilateral neck and mucosal irradiation is superior to ipsilateral neck irradiation in preventing contralateral cervical lymph node metastases and the subsequent appearance of an occult primary cancer. Both techniques combined with cervical lymph node dissection were equally effective in controlling the ipsilateral neck disease.  相似文献

The 0.1/dL copper sulfate solution may be used to replace distilled water for humidifying oxygen. It can reduce the carrier rate and prolong the using time. There is little copper-ion in the oxygen while the 0.5/dL copper sulfate solution were used continuously for 9 days. The oxygen humidified by copper sulfate solution has no peculiar smell.
