Significance of adrenal hormones in brain energy metabolism

Intensity of glycolysis was studied under conditions of immobilization stress and adrenalectomy as well as with administration of adrenaline and hydrocortisone. The processes of aerobic glycolysis, activity of enzymes of the tricarboxylic acid cycle and mitochondria respiratory chain, content of total, free and protein-bound glycogen, glucose-I-phosphate, glucose-6-phosphate, pyruvate, alpha-ketoglutarate, cytrate and macroergs in the brain are shown to depend on the level of adrenal hormones producing a multilateral effect on a series of energy metabolism links.     

Effect of split doses of N-methyl-N-nitrosourea on DNA repair synthesis in cultured mammalian cells

DNA repair was measured in human fibroblasts, mouse C3H 10 T 1/2 fibroblsts and rat hepatocytes by the non-semi-conservative incorporation of [3H]-TdR during DNA repair synthesis using liquid scintillation techniques. Confluent monolayers of these cells grown on cover slips were exposed to split doses (125 or 250 microgram/ml) of the mutagenic and carcinogenic alkylating agent MNU and DNA repair synthesis compared with that produced by a single dose (500 microgram/ml). No significant difference in DNA repair capacity was detected in the three cell lines treated with a single dose or split doses of MNU.     

Performance monitoring of the EC - 1030 system by a microprocessor - based monitor

This paper presents a M6800 microprocessor based hardware monitor for measuring certain selected parameters of the EC-1030 computer system. The motivation for this work was to get an insight into the operation of the system under the present work load and use these results towards improvement and design studies. The three main classes of quantitative indices of computer system performance/productivity, responsiveness and use have been measured using hardware and software tools. This paper describes the hardware monitor design and discusses the measurements made.     

Effects of atmospheric ozone on microarray data quality

A data anomaly was observed that affected the uniformity and reproducibility of fluorescent signal across DNA microarrays. Results from experimental sets designed to identify potential causes (from microarray production to array scanning) indicated that the anomaly was linked to a batch process; further work allowed us to localize the effect to the posthybridization array stringency washes. Ozone levels were monitored and highly correlated with the batch effect. Controlled exposures of microarrays to ozone confirmed this factor as the root cause, and we present data that show susceptibility of a class of cyanine dyes (e.g., Cy5, Alexa 647) to ozone levels as low as 5-10 ppb for periods as short as 10-30 s. Other cyanine dyes (e.g., Cy3, Alexa 555) were not significantly affected until higher ozone levels (> 100 ppb). To address this environmental effect, laboratory ozone levels should be kept below 2 ppb (e.g., with filters in HVAC) to achieve high quality microarray data.     

Synthesis of carbon-nitride films using a fast-switched dual-source low energy ion beam deposition system

Thin C_xN_y films were deposited in UHV using alternating low energy ion beams of C⁺ and N⁺ or N₂⁺ in the energy range of 5 to 100 eV. The ion beam deposition system is equipped with two Freeman ion sources, mass analysis and fast automated beam switching, allowing perpendicular bombardment of the target with a single ion beam at a time. The composition and density of the films were studied by ARS (in situ), XPS and RBS. The dependence of the film properties and growth mechanisms on ion energy, beam switching rate, and C-to-N arrival ratio have been investigated. The influence of the deposition parameters on the film stoichiometry is discussed. Exposure of the film to atmosphere leads to oxygen incorporation, resulting in a lowered surface concentration of nitrogen. The XPS N 1s and C Is binding energies vary in a relatively broad range indicating that several bond states may be present. The influence of the substrate material on film growth has also been studied. On Si{100}, film growth commences with the formation of an interfacial silicon nitride. No film growth was observed on gold, however deposition was possible on tantalum and molybdenum.     

P2X3 is expressed by DRG neurons that terminate in inner lamina II

The P2X3 receptor subunit, a member of the P2X family of ATP-gated ion channels, is almost exclusively localized in sensory neurons. In the present study, we sought to gain insight into the role of P2X3 and P2X3-containing neurons in sensory transmission, using immunohistochemical approaches. In rat dorsal root ganglia (DRG), P2X3-immunoreactivity (-ir) was observed in small- and medium-sized neurons. Approximately 40% of DRG neuronal profiles in normal rats contained P2X3-ir. In rats that had received neonatal capsaicin treatment, the number of P2X3-positive neurons was decreased by approximately 70%. Analysis of the colocalization of P2X3-ir with cytochemical markers of DRG neurons indicated that approximately 94% of the P2X3-positive neuronal profiles were labelled by isolectin B4 from Bandeiraea simplicifolia, while only 3% contained substance P-ir, and 7% contained somatostatin-ir. In dorsal horn of rat spinal cord, P2X3-ir was observed in the inner portion of lamina II and was reduced subsequent to dorsal rhizotomy, as well as subsequent to neonatal capsaicin treatment. Finally, P2X3-ir accumulated proximal to the site of sciatic nerve ligation, and was seen in nerve fibres in skin and corneal epithelium. In summary, our results suggest that P2X3 is expressed by a functionally heterogeneous population of BSI-B4-binding sensory neurons, and is transported into both central and peripheral processes of these neurons.
