This paper demonstrates the effect of large scale structures found in free shear layers on the dispersion of solid particles. Numerical simulation of a free shear layer using a psuedo-spectral code were carried out with the addition of discrete particles. It was shown that the dispersion of particles is strongly dependent on the Stokes number with the possibility of particle dispersions being greater than the fluid dispersion at certain values of the Stokes number. These trends compare qualitatively with flow visualization of a free shear flow with solid particles. 相似文献

We prepared large unilamellar vesicles (LUVs) with three different stratum corneum lipid compositions: constant amounts of ceramides (55 wt %) and fatty acids (15%) with varying amounts of cholesterol sulfate (0-15%) and cholesterol (15-30%). One of the compositions served as a model for normal stratum corneum, while the second one served as a model for recessive X-linked ichthyosis stratum corneum. The third composition consisted of no cholesterol sulfate. Intervesicle lipid interactions in these LUVs were monitored by fluorescence methods for content leakage, and contents mixing at pH 9, in the absence and presence of Ca2+, and at pH 6. Since the content leakage and contents mixing assays were originally developed for phospholipid vesicles, we characterized the probe binding and the probe quenching properties for stratum corneum LUV systems, and modified the assays slightly accordingly. The time-dependent fluorescence intensity changes in the probe-containing LUVs at pH 9 and 6 and in response to the addition of calcium were monitored. Our results demonstrated that all three types of LUVs were relatively stable at pH 9. Addition of Ca2+ or decreasing the pH to 6 activated intervesicle lipid mixing followed by vesicle fusion and lysis. We found that the LUVs with no cholesterol sulfate and 30% cholesterol exhibited a more extensive Ca2+- or low-pH-activated intervesicle lipid interaction than LUVs with either 5% cholesterol sulfate and 25% cholesterol or 15% cholesterol sulfate and 15% cholesterol. These results suggest that fusogenic agents such as Ca2+ and H+ act to neutralize the fatty acids in the lipid bilayer of stratum corneum vesicles. The inclusion of 5-15% cholesterol sulfate helps to prevent the collapse of fused vesicles into other structures. 相似文献

Spectrin of the erythrocyte membrane skeleton is composed of alpha- and beta-spectrin, which associate to form heterodimers and tetramers. It has been suggested that a fractional domain (helix C) in the amino-terminal region of alpha-spectrin (Nalpha region) bundles with another fractional domain in the carboxyl-terminal region of beta-spectrin (Cbeta region) to yield a triple alpha-helical bundle and that this helical bundling is largely responsible for tetramer formation. However, there are certain objections to assigning a preeminent role to this helical bundling in the tetramerization reactions. We prepared several recombinant peptides of alpha-spectrin fragments spanning only the Nalpha region (lacking the dimer nucleation site) and quantitatively studied their interaction with beta-spectrin. We found that a majority of the interactions were localized, as expected, in the Nalpha-helix C region but that there was also some contribution from the nonhomologous region. More importantly, the temperature and ionic strength dependence of this interaction in our model peptides was different from that in intact spectrin. We suggest that, although the regions involving the putative helical bundling in alpha- and beta-spectrin undoubtedly play a significant role in tetramerization, regions distal to the Nalpha-helix C region in spectrin are also involved in tetramer formation. Companies conducting compliance testing are required to analyze audit samples at the time they collect and analyze the stack samples if audit samples are available. Eastern Research Group (ERG) provides technical support to the EPA's Emission Measurements Center's Stationary Source Audit Program (SSAP) for developing, preparing, and distributing performance evaluation samples and audit materials. These audit samples are requested via the regulatory Agency and include spiked audit materials for EPA Method 29-Metals Emissions from Stationary Sources, as well as other methods. To provide appropriate audit materials to federal, state, tribal, and local governments, as well as agencies performing environmental activities and conducting emission compliance tests, ERG has recently performed testing of blank filter materials and preparation of spiked filters for EPA Method 29. For sampling stationary sources using an EPA Method 29 sampling train, the use of filters without organic binders containing less than 1.3 microg/in.2 of each of the metals to be measured is required. Risk Assessment testing imposes even stricter requirements for clean filter background levels. Three vendor sources of quartz fiber filters were evaluated for background contamination to ensure that audit samples would be prepared using filters with the lowest metal background levels. A procedure was developed to test new filters, and a cleaning procedure was evaluated to see if a greater level of cleanliness could be achieved using an acid rinse with new filters. Background levels for filters supplied by different vendors and within lots of filters from the same vendor showed a wide variation, confirmed through contact with several analytical laboratories that frequently perform EPA Method 29 analyses. It has been necessary to repeat more than one compliance test because of suspect metals background contamination levels. An acid cleaning step produced improvement in contamination level, but the difference was not significant for most of the Method 29 target metals. As a result of our studies, we conclude: Filters for Method 29 testing should be purchased in lots as large as possible. Testing firms should pre-screen new boxes and/or new lots of filters used for Method 29 testing. Random analysis of three filters (top, middle, bottom of the box) from a new box of vendor filters before allowing them to be used in field tests is a prudent approach. A box of filters from a given vendor should be screened, and filters from this screened box should be used both for testing and as field blanks in each test scenario to provide the level of quality assurance required for stationary source testing. 相似文献

High-performance green buildings require close integration of building systems with a special focus on energy, daylighting, and material analysis during their design processes. Design process modeling and use of visualization tools can facilitate better communication and collaboration between team members; hence better integration in the design process. This paper presents a case study of the integrated design process of an Early Childhood Learning Center as performed by a team of university students and faculty. A process modeling approach of key decisions, required consultants, and virtual prototypes of the building was used during the design development stages of the case study. This case illustrates the use of process modeling and visualization tools to provide an accurate building information system for integrated teams. Through this experience, process modeling and visualization tools were found to be useful mechanisms to achieve high performance design goals and minimize design process waste. 相似文献

The addition of both elements (Cr and Ni) and carbides (SiC and WC) during laser surface alloying under different processing speeds produced surfaces with both enhanced hardness wear resistance and corrosion properties compared to the base AISI 4340 steel material. These effects were due to the evolution of unique microstructures within the laser-processed region, which includes austenite, ferrite, martensite, Fe- and Si-based carbides and the retention of the original carbides (SiC and WC) in various combinations. The chromium and nickel stabilized the austenite and ferrite but reduced the formation of martensite that is useful to increase the hardness and prevent cracking. Also, the substantial dissociation of the original carbides (SIC and WC) into elemental silicon and tungsten supplemented the stabilization of ferrite and reduction in the hardness. The presence of the undissociated carbides and some martensite formation provided substantial increases in the microhardness. The improvement of both the mechanical properties and corrosion resistance might be self-exclusive due to the reduction of the carbides and the subsequent inability of the matrix to prevent cracking. 相似文献
