Raman microspectroscopy provides chemo‐selective image contrast, sub‐micrometer resolution, and multiplexing capabilities. However, it suffers from weak signals resulting in image‐acquisition times of up to several hours. Surface‐enhanced Raman scattering (SERS) can dramatically enhance signals of molecules in close vicinity of metallic surfaces and overcome this limitation. Multimodal, SERS‐active nanoparticles are usually labeled with Raman marker molecules, limiting SERS to the coating material. In order to realize multimodal imaging while acquiring the rich endogenous vibronic information of the specimen, a core–shell particle based on “Nanorice”, where a spindle‐shaped iron oxide core is encapsulated by a closed gold shell, is developed. An ultrathin layer of silica prevents agglomeration and unwanted chemical interaction with the specimen. This approach provides Raman signal enhancement due to plasmon resonance effects of the shell while the optical absorption in the near‐infrared spectral region provides contrast in photoacoustic tomography. Finally, T2‐relaxation of a magnetic resonance imaging (MRI) experiment is altered by taking advantage of the iron oxide core. The feasibility for Raman imaging is evaluated by nearfield simulations and experimental studies on the primate cell line COS1. MRI and photoacoustics are demonstrated in agarose phantoms illustrating the promising translational nature of this strategy for clinical applications in radiology. 相似文献

Nephropathic cystinosis is an autosomal recessive lysosomal storage disease characterized by renal failure at 10 years of age and other systemic complications. The gene for cystinosis, CTNS, has 12 exons. Its 2.6-kb mRNA codes for a 367-amino-acid putative cystine transporter with seven transmembrane domains. Previously reported mutations include a 65-kb "European" deletion involving marker D17S829 and 11 small mutations. Mutation analysis of 108 American-based nephropathic cystinosis patients revealed that 48 patients (44%) were homozygous for the 65-kb deletion, 2 had a smaller major deletion, 11 were homozygous and 3 were heterozygous for 753G-->A (W138X), and 24 had 21 other mutations. In 20 patients (19%), no mutations were found. Of 82 alleles bearing the 65-kb deletion, 38 derived from Germany, 28 from the British Isles, and 4 from Iceland. Eighteen new mutations were identified, including the first reported missense mutations, two in-frame deletions, and mutations in patients of African American, Mexican, and Indian ancestry. CTNS mutations are spread throughout the leader sequence, transmembrane, and nontransmembrane regions. According to a cystinosis clinical severity score, homozygotes for the 65-kb deletion and for W138X have average disease, whereas mutations involving the first amino acids prior to transmembrane domains are associated with mild disease. By northern blot analysis, CTNS was not expressed in patients homozygous for the 65-kb deletion but was expressed in all 15 other patients tested. These data demonstrate the origins of CTNS mutations in America and provide a basis for possible molecular diagnosis in this population. 相似文献

Rivermouth ecosystems in the Laurentian Great Lakes represent complex hydrologic mixing zones where lake and river water combine to form biologically productive areas that are functionally similar to marine estuaries. As urban, industrial, shipping, and recreational centers, rivermouths are the focus of human interactions with the Great Lakes and, likewise, may represent critical habitat for larval fish and other biota. The hydrology and related geomorphology in these deltaic systems form the basis for ecosystem processes and wetland habitat structure but are poorly understood. To this end, we examined hydrogeomorphic structure and lake-tributary mixing in three rivermouths of intermediate size using water chemistry, stable isotopes, and current profiling over a five-month period. In rivermouths of this size, the maximum depth of the rivermouth ecosystem influenced water mixing, with temperature-related, density-dependent wedging and layering that isolated lake water below river water occurring in deeper systems. The inherent size of the rivermouth ecosystem, local geomorphology, and human modifications such as shoreline armoring and dredging influenced mixing by altering the propensity for density differences to occur. The improved scientific understanding and framework for characterizing hydrogeomorphic processes in Great Lakes rivermouths across a disturbance gradient is useful for conservation, management, restoration, and protection of critical habitats needed by native species. 相似文献

Recent efforts to influence the efficiency and timing of urban indoor water use through education, technology, conservation, reuse, economic incentives, and regulatory mechanisms have enabled many North American cities to accommodate population growth and buffer impacts of drought. It is unlikely that this approach will be equally successful into the future because the source of conservation will shift from indoor to outdoor use. Outdoor water is climate sensitive, difficult to measure, hard to predict, linked to other components of complex and dynamic urban resource systems, imbued with behavioral and cultural dimensions, and implicated in societal conflicts about climate risk, modern lifestyles, social justice, and future growth. Outdoor water conservation is not a traditional management problem focused on the water sector, assuming a stationary climate, and set aside from public debate. Instead, outdoor water is an adaptation problem, involving complex and uncertain system dynamics, the need for cross-sector coordination, strategies for dealing with climatic uncertainty, and mechanisms for engaging stakeholders with differing goals. This paper makes the case for treating outdoor water as an adaptation problem and offers a six-point strategy for how cities can better prepare their water systems for the uncertainties of climate and societal change. 相似文献

Percolation theory of transport in random composites is used to explain the correlation between the residual saturation of nonwetting phase in porous media after displacement by a wetting phase and the capillary number, this number being a measure of the ratio of Darcy-law viscous force in the wetting liquid to interfacial tension force in curved menisci between the two phases. Statistical concepts of percolation theory give estimates of the length distribution of blobs created when the nonwetting phase loses continuity because of displacement by the wetting phase. These estimates agree with the few experimental data. Simple blob mobilization theory and experiments establish that the capillary number required to mobilize a blob is inversely proportional to its length in the direction of the Darcy-law pressure gradient; this and the predictions of percolation theory account for the observed capillary number correlation. 相似文献

The accuracy and precision of thin-film interfacial mixing as measured with atom probe tomography (APT) are assessed by considering experimental and simulated field-evaporation of a Co/Cu/Co multilayer structure. Reconstructions were performed using constant shank angle and Z-scale reordering algorithms. Reconstruction of simulated data (zero intermixing) results in a 10-90% intermixing width of ∼0.2 nm while experiential intermixing (measured from multiple runs) was 0.47±0.19 and 0.49±0.10 nm for Co-on-Cu and Cu-on-Co interfaces, respectively. The experimental data were collected in analysis orientations both parallel and anti-parallel to film growth direction and the impact of this on the interfacial mixing measurements is discussed. It is proposed that the resolution of such APT measurements is limited by the combination of specimen shape and reconstruction algorithms rather than by an inherent instrumentation limit. 相似文献
