Simulation of the fatigue crack propagation in a Wendelstein 7‐X baffle module is performed in this study using both a finite element method‐based software and the UniGrow nonlinear model for small‐scale yielding (SSY) conditions. Some experimental fatigue tests of several cracked baffle modules have been performed through a servo‐hydraulic machine. One of these experimental tests has been considered to simulate fatigue crack propagation in the baffle module. Before starting the experimental test, a first crack partly contained in the welding seam and partly in the steel pipe is found. Subsequently, owing to the applied load, the crack propagated both into the welding seam and into the steel pipe until the plastic zone in the near field attains SSY conditions. Finally, owing to the increase in the extension of the plastic zone, SSY conditions are not more valid, and the breakage of the steel pipe is produced by plastic collapse.  相似文献

1. Anti-acetylcholine effects of pilsicainide, flecainide, disopyramide and propafenone on the acetylcholine (ACh)-induced K+ current (IK.ACh) were examined in dissociated guinea-pig atrial myocytes under whole-cell voltage clamp by the use of the 'concentration-clamp' technique. 2. The IK.ACh was activated with a latency of about 100 ms after 1 microM ACh application and desensitized to a steady-state level. The latent period and the time to peak response were shortened with increasing ACh concentration. 3. The values of half-maximal inhibition (IC50) on the peak and steady state responses were 25 and 25 microM for pilsicainide, 1.7 and 2.0 microM for disopyramide, 19 and 2.0 microM for flecainide and 0.7 and 0.2 microM for propafenone, respectively. 4. Pilsicainide and disopyramide increased the latent period and the time to peak of IK.ACh in a concentration-dependent manner. Flecainide and propafenone did not change the latent period, but shortened the time to peak and hastened the decay of IK.ACh in a voltage-independent manner. 5. The results suggest that the mechanisms underlying the anti-acetylcholine effect of antiarrhythmic drugs are different among these drugs: i.e., pilsicainide and disopyramide mainly block the muscarinic ACh receptors while flecainide and propafenone inhibit the K+ channel itself as open channel blockers.  相似文献

The study examined how social constraints on discussion of a traumatic experience can interfere with cognitive processing of and recovery from loss. Bereaved mothers were interviewed at 3 weeks (Tl ), 3 months (T2), and 18 months (T3) after their infants' death. Intrusive thoughts at Tl, conceptualized as a marker of cognitive processing, were negatively associated with talking about infant's death at T2 and T3 among socially constrained mothers. The reverse associations were found among unconstrained mothers. Controlling for initial level of distress, there was a positive relation between T1 intrusive thoughts and depressive symptoms over time among socially constrained mothers. However, higher levels of T1 intrusive thoughts were associated with a decrease in T3 depressive symptoms among mothers with unconstrained social relationships. The role of interior design elements in mitigating the negative relationship between residential crowding and psychological health was investigated. Residents of crowded homes with greater architectural depth (the number of spaces one must pass through to get from one room in the house to another) are less likely to socially withdraw or to be psychologically distressed than residents in crowded homes with relatively low depth. Additional analyses suggest that greater depth buffers the association between residential crowding and psychological distress because it reduces social withdrawal among residents of crowded homes. A longitudinal study examined the relative and joint effects of perceived social support and social conflict on psychological distress in 228 college students. Women had higher perceived social support from roommates and friends and less conflict with roommates than did men; there were no gender differences in level of conflict with friends or psychological distress. Roommate conflict predicted increases in psychological distress over time; this effect was attenuated by high levels of perceived social support from friends. Friend conflict also predicted increases in psychological distress over time; this effect was attenuated by high levels of perceived social support from roommates. These results show the importance of negative and positive aspects of social experiences to emotional functioning and the importance of compensatory social support for individuals facing social conflicts. Three‐dimensional mixed‐mode crack propagation simulations were performed by means of the dual boundary element method code BEASY and 2 finite element method‐based crack propagation codes: ZENCRACK (ZC) and CRACKTRACER3D (CT3D). The stress intensity factors (SIFs) along the front of an initial semielliptical crack, initiated from the external surface of a shaft, were calculated for 4 different load cases: bending, press fit, shear, and torsion. The methods used for the SIF assessment along the crack front were the J‐integral for BEASY and ZC and the quarter point element stress method for CT3D. Subsequently, crack propagation simulations were performed, with the crack growth rate evaluated by using Paris' law, calibrated for the material at stake (American Society for Testing and Materials A469 steel). The kink angles were evaluated by using the minimum strain energy density and maximum tangential stress criteria for BEASY, the maximum energy release rate and maximum tangential stress for ZC, and the maximum principal asymptotic stress for CT3D. The results obtained in terms of SIFs and crack propagation life show very good agreement among the 3 codes. Also, the shape of the propagated crack, which is significantly out‐of‐plane for the shear and torsion loading, matched very well.  相似文献

The pBR322 recombinant plasmids DNA products, which purified by phenol-extract and equilibrium centrifugation in CsCl-ethidium bromide gradients, have been shown as mainly spatial conformation of covalently closed circular DNA (i.e. cccDNA) by agarose gel electrophoresis for homogeneity detection. Laser Raman spectra in the region 450-1750 cm-1 have been obtained for the circular double-stranded plasmids DNA molecule in an aqueous solution, indicating not only contain those marker peaks of secondary structure as in conventional B-form of DNA, but also present both 854 and 1083 cm-1 diagnostic bands of reflecting the vibration state of deoxyribosyl phosphodiester backbone. The present analysis have demonstrated relationship between the superhelical state for cccDNA and the two conformational marker bands that can be considered as the tertiary structure marker on plasmids DNA. Because of markedly Raman hypochromicity of charecter band 1378cm-1 of dT in contrast to liear DNA molecule, as well as the carbonyl double bond vibration line of dT have shifted to higher wave number position, the base stacking analysis represent occurrence of both increase in staking reaction activity of dT, and injury of a number of the Hoogsteen hydrogen bond between dA and dT with exist supercoil structure in cccDNA.  相似文献

The current study was designed to evaluate tubing sets with either polymeric phospholipids or ionically bound heparin in six bovine experiments (body weight, 70 +/- 5 kg). No heparin was given systemically. Left heart bypass was started with 300 ml of clear priming solution and maintained over 6 hours (50 ml/kg/min). Coagulation studies included platelet counts, activated coagulation time (ACT), thrombin time (TT), fibrinogen (Factor I), antithrombin III (AT III), and fibrinopeptide A (FPA). Normalized platelet levels dropped from 100 +/- 12% before to 86 +/- 13% after 6 hours of left heart bypass for heparin, compared with 100 +/- 46% to 90 +/- 44% for phospholipid coating (NS). The ACT increased from 146 +/- 7 sec at 10 min to 159 +/- 16 sec after 6 hours for heparin, compared with 122 +/- 4 to 126 +/- 5 sec for phospholipid (p < 0.05). Thrombin time changed from 18 +/- 0 sec before to 19 +/- 1 sec after 6 hours for heparin, as compared with 16 +/- 1 sec to 18 +/- 1 sec for phospholipid (NS). Factor I levels decreased from 1.5 +/- 0.3 g/L to 1.3 +/- 0.1 g/L for heparin, compared with 1.5 +/- 0.2 g/L to 1.4 +/- 0.3 g/L for phospholipid. Antithrombin III levels changed from 102 +/- 26% to 91 +/- 7% for heparin, compared with 123 +/- 12% to 118 +/- 12% for phospholipid. Fibrinopeptide A levels changed from 100 +/- 60% to 130 +/- 13% for heparin, compared with 100 +/- 11% to 99 +/- 6% for phospholipid (P < 0.05).
