Several studies have reported on the detection of perchlorate (ClO(4)(-)) in edible leafy vegetables irrigated with Colorado River water. However, there is no information on spinach as related to ClO(4)(-) in irrigation water nor on the effect of other anions on ClO(4)(-) uptake. A greenhouse ClO(4)(-) uptake experiment using spinach was conducted to investigate the impact of presence of chloride (Cl(-)) and nitrate (NO(3)(-)) on ClO(4)(-) uptake under controlled conditions. We examined three concentrations of ClO(4)(-), 40, 220, and 400 nmol(c)/L (nanomoles of charge per liter of solution), three concentrations of Cl(-), 2.5, 13.75, and 25 mmol(c)/L, and NO(3)(-) at 2, 11, and 20 mmol(c)/L. The results revealed that ClO(4)(-) was taken up the most when NO(3)(-) and Cl(-) were lowest in concentration in irrigation water. More ClO(4)(-) was detected in spinach leaves than that in the root tissue. Relative to lettuces, spinach accumulated more ClO(4)(-) in the plant tissue. Perchlorate was accumulated in spinach leaves more than reported for outer leaves of lettuce at 40 nmol(c)/L of ClO(4)(-) in irrigation water. The results also provided evidence that spinach selectively took up ClO(4)(-) relative to Cl(-). We developed a predictive model to describe the ClO(4)(-) concentration in spinach as related to the Cl(-), NO(3)(-), and ClO(4)(-) concentration in irrigation water.  相似文献

It is often speculated that progressive deterioration of left ventricular function in heart failure is due to ongoing loss of viable cardiocytes. In this study, we examined the possibility that cardiocyte loss in heart failure may be due, in part, to apoptosis, an active process of gene-directed cellular self-destruction. Studies were performed in left ventricular tissue obtained from 10 dogs with chronic heart failure produced by multiple intracoronary microembolizations (left ventricular ejection fraction 27 +/- 1%) and from 5 normal dogs. Evidence for cardiocyte apoptosis was based on transmission electron microscopy criteria and on in situ immunohistochemical labeling of nuclear DNA fragmentation. There was no evidence of apoptotic cardiocytes in normal dogs. Features of cardiocyte apoptosis were observed in dogs with heart failure primarily in regions bordering old infarcts. Electron microscopic features of cardiocyte apoptosis included (1) intact sarcolemma and inner organelles in the presence of compaction and segregation of nuclear chromatin into sharply delineated masses that about the nuclear envelope, (2) intact sarcolemma in the presence of cytoplasm shrinkage, blebbing, and nuclear fragmentation, and (3) intact sarcolemma in the presence of complete disorganization of inner organelles and disappearance of nucleolemma. A count of all of the apoptotic bodies positively labeled for nuclear DNA fragments showed that 11% were of cardiocyte origin confirmed by positive labeling with striated muscle antimyosin antibody. We conclude that morphological and biochemical features of cardiocyte apoptosis exist in the left ventricular myocardium of dogs with chronic heart failure.  相似文献

Nitrous oxide (concentrations of 30 to 50%) was administered to 22 healthy volunteer subjects via nasal prongs, rebreathing mask, and an airlines mask to assess the efficiency of systems of delivery and the hemodynamic effects. The end-expired concentration of nitrous oxide, expressed as a percentage of the inspired concentration of nitrous oxide, was 19% for nasal prongs, 34% for the rebreathing mask, and 95% for the airlines mask (most accurate delivery). The pulse rate fell from a mean of 79+/-3 beats per minute to 67+/-2 with nasal prongs, to 64+/-2 with the rebreathing mask, and to 64+/-2 with the airlines mask. Both systolic and diastolic pressures fell from means of 122+/-4 and 74+/-2 mm Hg, respectively, to 98+/-3 mm Hg and 64+/-2 mm Hg, respectively, with the airlines apparatus. The fall in systolic blood pressure (slope, -0.79) exceeded that in the diastolic (slope, -0.35). In additional eight normal subjects, administration of 30% nitrous oxide via airlines mask produced identical changes in blood pressure and pulse rate, but there was no effect from 30 minutes of administration of 30% nitrous oxide on the end-systolic volume index, cardiac index, ejection fraction, normalized wall velocity determined echocardiographically, ejection time, or the ratio of preejection period to ejection time.  相似文献

Soil fumigants are used to control a wide variety of soil-borne pests in high-cash-value crops. Application of soil fumigants through drip irrigation systems is receiving increasing attention as a method to improve the uniformity of fumigant application. Little information is available on the emissions and soil distribution of fumigants following subsurface drip application, or the effect of plastic tarp on fumigant emissions in these systems. In these experiments, the fumigant compounds 1,3-dichloropropene (1,3-D), Vapam (a methyl isothiocyanate (MITC) precursor), and propargyl bromide (PrBr) were applied to soil beds via drip irrigation at 15 cm depth. Beds were tarped with either standard 1-mil high-density polyethylene (HDPE) or a virtually impermeable film (VIF), leaving the furrows bare. Cumulative emissions of 1,3-D, MITC, and PrBr in these tarped bedded systems was very low, amounting to <10% of the applied mass. These experiments were conducted in the winter months, with average air temperatures of 12-15 degrees C. Cumulative emissions of MITC and 1,3-D from a sandy loam field soil were decreased by > or =80% by tarping the bed with VIF rather than HDPE. A large fraction of the 1,3-D and PrBr flux was from the untarped furrows in VIF-tarped plots, indicating that inhibiting volatilization from the furrow will be important in further reducing emissions in these systems. Monitoring the fumigant distribution in soil indicated that tarping the bed with VIF resulted in a more effective containment of fumigant vapors compared to use of a HDPE tarp.  相似文献

Phenolic phytochemicals are associated with many health benefits, and it would be useful to develop improved methods for their extraction from bean. In this work, we showed that microwave-assisted extraction was an effective method. Eight bean types important in the Northarvest region of North America, which includes North Dakota and Minnesota, were chosen for this study. Four temperatures (25, 50, 100 and 150 °C) and three solvents (water, 50% ethanol in water and 100% ethanol) were investigated. As expected, colored beans contained higher concentrations of extractable phenolics, and in most cases the concentration of phenolics was much higher in the hull (testa) than in the cotyledon. Extraction efficiency was superior at higher extraction temperatures. The most effective extraction was achieved at a temperature of 150 °C using 50% ethanol. Total phenolics contents determined by microwave-assisted extraction with water at 100 °C were two to three times those determined by conventional extraction with water at the same temperature.  相似文献

Abnormalities of contractile function have been identified in cardiomyocytes isolated from failed human hearts and from hearts of animals with experimentally induced heart failure (HF). The mechanism(s) responsible for these functional abnormalities are not fully understood. In the present study, we examined the relationship between action potential duration, pattern of contraction and relaxation, and associated intracellular Ca2+ transients in single cardiomyocytes isolated from the left ventricle (LV) of dogs (n = 7) with HF produced by multiple sequential intracoronary microembolizations. Comparisons were made with LV cardiomyocytes isolated from normal dogs. Action potentials were measured in isolated LV cardiomyocytes by perforated patch clamp, Ca2+ transients by fluo 3 probe fluorescence, and cardiomyocyte contraction and relaxation by edge movement detector. HF cardiomyocytes exhibited an abnormal pattern of contraction and relaxation characterized by an attenuated initial twitch (spike) followed by a sustained contracture ('dome') of 1 to 8 s in duration and subsequent delayed relaxation. This pattern was more prominent at low stimulation rates (58% at 0.2 Hz, n = 211, 21% at 0.5 Hz, n = 185). Measurements of Ca2+ transients in HF cardiomyocytes at 0.2 Hz manifested a similar spike and dome configuration. The dome phase of both the contraction/relaxation pattern and Ca2+ transients seen in HF cardiomyocytes coincided with a sustained plateau of the action potential. Shortening of the action potential duration by administration of saxitoxin (100 nM) or lidocaine (30 microM) reduced the duration of the dome phase of both the contraction/relaxation profile as well as that of the Ca2+ transient profile. An increase of stimulation rate up to 1 Hz caused shortening of the action potential and disappearance of the spike-dome profile in the majority of HF cardiomyocytes. In HF cardiomyocytes, the action potential and Ca2+ transient duration were not significantly different from those measured in normal cells. However, the contraction-relaxation cycle was significantly longer in HF cells (314 +/- 67 ms, n = 21, vs. 221 +/- 38 ms, n = 46, mean +/- SD), indicating impaired excitation-contraction uncoupling in HF cardiomyocytes. The results show that, in cardiomyocytes isolated from dogs with HF, contractile abnormalities and abnormalities of intracellular Ca2+ transients at low stimulation rates are characterized by a spike-dome configuration. This abnormal pattern appears to result from prolongation of the action potential.  相似文献

Today, drivers are faced with many in-vehicle activities that are potentially distracting. In many cases, they are not passive recipients of these tasks; rather, drivers decide whether or not (or how) to perform them. In this study, we examined whether drivers, given knowledge of the upcoming road demands, would strategically delay performing in-vehicle activities until demands were reduced. Twenty drivers drove an instrumented van around a closed track that was divided into sections of varying demands and difficulty. Drivers were asked to perform one of four in-vehicle tasks (e.g., phone conversation; read a text message; find an address; pick up an object on the floor); however, they were free to decide when to initiate these tasks, provided they finish them before a given deadline. Although drivers were fully aware of the relative demands of the road, they did not tend to strategically postpone tasks—a finding that was consistent across the different tasks (p > .05). Rather, drivers tended to initiate tasks regardless of the current driving conditions. This strategy frequently led to driving errors. Given the control that drivers have over many in-vehicle distractions, interventions that focus on strategic decisions and planning may have merit.  相似文献
