Alport syndrome (AS) is a hereditary renal disorder with no etiological therapy. In the preclinical Col4a3-/- model of AS, disease progression and severity vary depending on mouse strain. The sodium-glucose cotransporter 2 (SGLT2) is emerging as an attractive therapeutic target in cardiac/renal pathologies, but its application to AS remains untested. This study investigates cardiorespiratory function and SGLT2 renal expression in Col4a3-/- mice from three different genetic backgrounds, 129x1/SvJ, C57Bl/6 and Balb/C. male Col4a3-/- 129x1/SvJ mice displayed alterations consistent with heart failure with preserved ejection fraction (HFpEF). Female, but not male, C57Bl/6 and Balb/C Col4a3-/- mice exhibited mild changes in systolic and diastolic function of the heart by echocardiography. Male C57Bl/6 Col4a3-/- mice presented systolic dysfunction by invasive hemodynamic analysis. All strains except Balb/C males demonstrated alterations in respiratory function. SGLT2 expression was significantly increased in AS compared to WT mice from all strains. However, cardiorespiratory abnormalities and SGLT2 over-expression were significantly less in AS Balb/C mice compared to the other two strains. Systolic blood pressure was significantly elevated only in mutant 129x1/SvJ mice. The results provide further evidence for strain-dependent cardiorespiratory and hypertensive phenotype variations in mouse AS models, corroborated by renal SGLT2 expression, and support ongoing initiatives to develop SGLT2 inhibitors for the treatment of AS. 相似文献

Because magnetic resonance imaging (MRI) contrast agents play a vital role in diagnosing diseases, demand for new MRI contrast agents, with an enhanced sensitivity and advanced functionalities, is very high. During the past decade, various inorganic nanoparticles have been used as MRI contrast agents due to their unique properties, such as large surface area, easy surface functionalization, excellent contrasting effect, and other size-dependent properties. This review provides an overview of recent progress in the development of nanoparticle-based T1-weighted MRI contrast agents. The chemical synthesis of the nanoparticle-based contrast agents and their potential applications were discussed and summarized. In addition, the recent development in nanoparticle-based multimodal contrast agents including T1-weighted MRI/computed X-ray tomography (CT) and T1-weighted MRI/optical were also described, since nanoparticles may curtail the shortcomings of single mode contrast agents in diagnostic and clinical settings by synergistically incorporating functionality. 相似文献

Fine powders of GaOOH nanocrystals are synthesized via a facile hydrolysis process through the solution–solution interface reactions of anhydrous GaCl3 and distilled water followed by subsequent solvothermal treatment at mild conditions. Well‐faceted α‐GaOOH hexagonal prism‐like nanorods are prepared through solvothermal treatment at 180°C with CTAB as the morphology controlling surfactant. Ga2O3 nanocrystals are fabricated via the pyrolysis of α‐GaOOH hexagonal prism‐like nanostructures at temperatures above 410°C. A peculiar back‐transformation from β‐Ga2O3 to α‐Ga2O3 has been observed to occur between about 557°C and 719°C, which is considered to be responsible for the coexistence of the two phases. The phase transformation mechanisms of Ga2O3 at elevated temperatures, as well as the possible transformation route, have been postulated from a thermodynamic point of view. 相似文献

Summary A series of cellulose esters bearing olig(oxyethylene)s with different degree of substitution (DS) and different length of the oxyethylene chain were synthesized by a homogeneous reaction of cellulose with corresponding monofunctional acid chloride in a 10% LiCl-dimethyl acetoamide (DMAc) solution. The effect of total DS value on the solubility of the derivatives in aqueous solution was investigated. It was found that the lower limit DS value for both water-soluble and amphiphilic derivatives decreases with increasing length of oxyethylene chains. The amphiphilic derivatives, which are soluble in both water and chloroform, precipitate out of aqueous solution on heating without gel forming, such a phase transition behavior was studied in terms of DS value, length of oxyethylene and concentration. The precipitation temperature (Tp) of the amphiphilic derivatives is range from 54°C to 96°C. It decreases with increasing the total DS value, and increases with an increase in the length of oxyethylene chains. The Tp value of the derivatives was found to be almost independent in the concentration range of 1–15 wt %, however the Tp value increases sharply with decreasing polymer concentration when the concentration is lower than 1 wt%. The ultrasonic guided waves are dispersive waves characterized by the phase and group velocities dispersion curves. In order to use guided waves in various industrial applications, their parameters must be known. Since these guided wave velocities depend on the frequency and thickness of the material, they propagate differently comparing to bulk ultrasonic waves. Therefore, to analyze the parameters of such waves, new measurement techniques should be proposed and possibilities of their application have to be investigated. In this paper possibilities to measure the group velocity based on the spectrum decomposition approach are presented. The investigations are carried out using the simulated and experimental signals of Lamb wave propagating in a 2 mm thickness aluminium plate. The two fundamental modes \(\hbox {A}_{0}\) and \(\hbox {S}_{0}\) are selected. Using the proposed technique, segments of the group velocity dispersion curves have been reconstructed and compared with the dispersion curves calculated by the SAFE method to estimate errors. Accordingly to the obtained lower absolute and relative errors, an optimal set of narrowband filters with bandwidth from 20 kHz up to 100 kHz for the \(\hbox {A}_{0}\) mode and from 80 kHz up to 160 kHz for the \(\hbox {S}_{0}\) mode are proposed. Applying the proposed optimal frequency sets of narrowband filters for the experimental signals, segments of the group velocity dispersion curves for both modes are reconstructed. The average relative error for the \(\hbox {A}_{0}\) mode is 1.7–2.2% (expanded relative uncertainty ± (2.2–2.8)%) and 0.78–1.2% (expanded relative uncertainty ± (0.5–0.8)%) for the \(\hbox {S}_{0}\) mode. 相似文献

针对在云机器人系统中,机器人端和云端之间消息需要转换格式才能传递,对异构机器人的兼容性较差,并且对网络环境的鲁棒性有待提高的情况,提出了基于ROS并只在本地端部署主节点的云机器人系统CloudROS。首先,本地和云端的服务均采用标准的ROS节点形式进行封装,云端的服务节点可以动态加入到机器人端的ROS网络,节点之间的消息格式无进行转换,提高了系统的兼容性;然后,在云端采用了授权认证和Docker虚拟化技术,保证了系统的安全性;最后,提出了服务质量(QoS)监测与调节策略,通过动态调整任务参数或切换本地与云端的服务,使QoS保持一个较优的值。实验结果表明,CloudROS可以有效地把机器人的计算任务卸载到云端,使机器人的性能得到提升,并对网络环境具有较好的鲁棒性。 相似文献
