This two-phase trial involved 83 Holstein heifers. The rearing phase consisted of two diets (alfalfa silage plus corn grain for ad libitum intake vs. corn silage plus urea for ad libitum intake) and two breeding age groups (13 vs. 16 mo). The lactation phase compared the above treatments plus two lactation feeding systems: concentrate fed individually to production versus a TMR. The heifers were assigned randomly to the rearing phase at 7 wk of age and fed their respective diets until 14 d prepartum. They were placed on preassigned lactation diets 14 d prepartum and remained on the lactation phase for 550 consecutive d. Daily gains and height at the withers were similar between forage groups during the first half of the rearing phase; in the second half, the heifers fed alfalfa silage were taller at the withers. Those fed corn silage consumed less DM and CP throughout the rearing phase and gained more BW than the heifers fed alfalfa in the latter half. There were no differences in daily gain or DMI between the breeding age groups. In the lactation phase, the group fed alfalfa for ad libitum intake consumed more DM and gave more milk during the first 90 d of lactation than heifers fed corn silage. However, after 90 d the difference in cumulative milk production was not significant. There were no differences in milk production. FCM, or DMI between breeding age groups. The cows fed concentrate consumed more DM and gave more milk during the first 90 d of lactation. By 550 d, there were no differences. Feeding high levels of corn silage can cause heifers to have lower DMI in early lactation. These differences tend to disappear during the second lactation.  相似文献

At 55 to 70 degrees C, thermal inactivation D-values for Escherichia coli O157:H7, Salmonella, and Listeria monocytogenes were 19.05 to 0.038, 43.10 to 0.096, and 33.11 to 0.12 min, respectively, in ground turkey and 21.55 to 0.055, 37.04 to 0.066, and 36.90 to 0.063 min, respectively, in ground beef. The z-values were 5.73, 5.54, and 6.13 degrees C, respectively, in ground turkey and 5.43, 5.74, and 6.01 degrees C, respectively, in ground beef. In both ground turkey and beef, significant (P < 0.05) differences were found in the D-values between E. coli O157:H7 and Salmonella or between E. coli O157:H7 and L. monocytogenes. At 65 to 70 degrees C, D-values for E. coli O157:H7, Salmonella, and L. monocytogenes were also significantly (P < 0.05) different between turkey and beef. The obtained D- and z-values were used in predicting process lethality of the pathogens in ground turkey and beef patties cooked in an air impingement oven and confirmed by inoculation studies for a 7-log (CFU/g) reduction of E. coli O157:H7, Salmonella, and L. monocytogenes.  相似文献

ABSTRACT: Thermal inactivation D (decimal reduction time at a certain heating temperature) values of Escherichia coli O157:H7 at 55 °C to 70 °C were 21.36 to 0.031 min in raw franks and 24.91 to 0.038 min in fully cooked franks. Although statistically significant differences were found on the D values of E. coli O157:H7 between raw and fully cooked franks, the z value of E. coli O157:H7 in raw franks (5.07 °C) and fully cooked franks (5.08 °C) was not significantly different. The obtained D and z values were used to validate the process lethality of the pathogen in the raw franks that were processed according to a multistage cooking/cooling schedule or in the fully cooked franks that were pasteurized in-packages via steam. In this study, the calculated process lethality for both the cooking (process lethality = 254 min) and post-cook pasteurization (process lethality = 39 min) processes was far greater than the processing time that was needed for achieving a 7D reduction of E. coli O157:H7 during cooking and post-cook pasteurization of the franks.  相似文献

The objective of this research was to evaluate the consumer controlled factors of cooking method and degree of doneness on top loin steaks from different USDA quality grades (Low Choice, High Select or Low Select) and breed-types (English, Continental European Cross or Brahman Cross). In addition, cities within the same region were evaluated for differences in consumer controlled factors and palatability responses. The in-home product test was conducted in Dallas and San Antonio, Texas, USA. Consumers (n=173) evaluated steaks for overall like (OSAT), tenderness (TEND), juiciness (JUIC), and flavor (FLAV) using 23-point hedonic scales. Respondents in Dallas cooked their steaks to higher degrees of doneness than did those in San Antonio. Outdoor grilling was the most frequently used method of cookery for steaks in both cities. Generally, consumers in San Antonio gave higher palatability ratings to Choice steaks and Dallas consumers gave higher ratings to Select steaks. The interactions of city×cooking method, breed-type×cooking method, and degree of doneness×cooking method were significant for all palatability attributes. In addition, the interaction of cooking method×quality grade was significant for TEND, JUIC, and FLAV. Warner-Bratzler shear (WBS) force was determined on a steak from each strip loin. Steaks from Continental European Cross cattle and Low Choice carcasses had the lowest WBS values. Differences in consumer preparation of beef top loin steaks present very unique challenges for the beef industry. Consumer information programs may serve a valuable role in connecting consumer perceptions with the preparation techniques needed to consistently achieve satisfaction.  相似文献

In this study we report the development and application of a Multiple-Locus Variable number of tandem repeat Analysis (MLVA) strategy for subtyping Listeria monocytogenes. Genome profiles of a collection of forty-five food-borne L. monocytogenes isolates were compared using MLVA. These isolates were obtained as part of an active surveillance programme of foods in the south-east region of Ireland. MLVA successfully discriminated amongst the isolates. The method was easy to perform, relatively fast and could be deployed in any molecular laboratory with basic laboratory equipment. This approach is a valuable tool, which has the capability to provide comparable results when compared with other more established typing methods, including pulsed-field gel electrophoresis (PFGE).  相似文献

Post partum negative energy balance (NEB) in dairy cattle is associated with a delayed return to ovarian cyclicity and reduced fertility. This study compared the IGF system of pre-recruitment ovarian follicles between cows in mild (n = 6) or severe (n = 6) NEB during early lactation. Ovaries were collected in the second week post partum, when circulating concentrations of IGF-I and glucose were lower (P < 0.01) in severe NEB cows. mRNA expression for IGF-II, type 1 IGF receptor (IGF-1R) and IGF-binding proteins (IGFBP)-1 to IGFBP-6 was determined by in situ hybridisation in individual follicles using radiolabelled oligonucleotide probes. Follicles were classified as very small (1-2.5 mm) or small (2.5-5 mm) and healthy or atretic. Relative mRNA concentrations were measured as optical density (OD) units using image analysis. Thecal IGF-II mRNA expression was highest in very small, healthy follicles (P < 0.05). Granulosa cell IGFBP-2 was the only component to change with EB status, with higher mRNA expression in mild compared with severe NEB cows (P < 0.05). IGFBP-1 and IGFBP-3 mRNA expression were undetectable. IGF-1R, IGFBP-4 and IGFBP-5 mRNA expression were not significantly altered by follicle size or health, but IGFBP-5 tended to increase in atretic follicles. The pattern of IGFBP-6 mRNA expression in theca paralleled that of IGF-II mRNA, with higher (P < 0.05) levels in healthy, very small follicles. In conclusion, the reduced expression of IGFBP-2 mRNA in severe NEB cows may alter the bioavailability of circulating IGF-I and locally produced IGF-II to modulate the pre-recruitment stages of follicles required to maintain normal post partum ovarian cyclicity.  相似文献

A boiling water reactor SVEA-96+ fresh fuel lattice has been used as the basis for a benchmark study of the void reactivity coefficient at assembly level in the full voidage range. Results have been obtained using the deterministic codes CASMO-4, HELIOS, PHOENIX, BOXER and the probabilistic code MCNP4C, combined for almost all cases with different cross section libraries. A statistical analysis of the results obtained showed that the void reactivity coefficient tends to become less negative beyond 80% void and that the discrepancies between codes tend to increase from less than 15% at voidages lower than 40% to more than 25% at voidages higher than 70%. The void reactivity coefficient results and the corresponding differences between codes were isotopically decomposed to interpret discrepancies. The isotopic decomposition shows that the minimum observed in the void reactivity coefficient between 80% and 90% void is largely due to the decrease in the relative importance of the 157Gd(n, γ) rate with increasing voidage, and that the fundamental discrepancies between codes or libraries are mainly governed by the different predictions of the 238U(n, γ) variation with voidage.  相似文献

Dental research studies often produce relatively small data sets in which observations are serially or spatially correlated. Rerandomization tests are presented as alternatives to analysis of variance and multivariate analysis for assessing group differences using such data. Rerandomization tests are particularly useful when the investigator is unwilling to make strong assumptions about the nature of the serial correlation or the distribution of the data. It is now well understood that the bone marrow (BM) compartment can sense systemic inflammatory signals and adapt through increased proliferation and lineage skewing. These coordinated and dynamic alterations in responding hematopoietic stem and progenitor cells (HSPCs), as well as in cells of the bone marrow niche, are increasingly viewed as key contributors to the inflammatory response. Growth factors, cytokines, metabolites, microbial products, and other signals can cause dysregulation across the entire hematopoietic hierarchy, leading to lineage-skewing and even long-term functional adaptations in bone marrow progenitor cells. These alterations may play a central role in the chronicity of disease as well as the links between many common chronic disorders. The possible existence of a form of “memory” in bone marrow progenitor cells is thought to contribute to innate immune responses via the generation of trained immunity (also called innate immune memory). These findings highlight how hematopoietic progenitors dynamically adapt to meet the demand for innate immune cells and how this adaptive response may be beneficial or detrimental depending on the context. In this review, we will discuss the role of bone marrow progenitor cells and their microenvironment in shaping the scope and scale of the immune response in health and disease.  相似文献
