α-Amylase K is a novel thermostable α-amylase from Bacillus subtilis which produces maltohexaose from starch. Various methods showed the existance of two forms which did not differ significantly in action and which may therefore be isoenzymes. α-Amylase K is stable at 60°C in the presence of additional calcium chloride with 80% activity remaining after 6d. It is completely deactivated after 10 min at 90°C and inhibited by tris-(hydroxymethyl)-aminomethane above pH 5.0. The purified α-amylase K fractions are less thermostable than the original enzyme solution. α-Amylase K is stable to freeze drying but reconstituted enzyme solution was less active at at pH-values below of 5.8.  相似文献

The development of controlled atmosphere storage technology for insect control requires an accurate prediction of the distribution of introduced gases in bulk grain. The published models are based on either diffusion theory or convective-diffusion theory. The momentum transport of gases, which is a key driving force for the gas mixture movements, has been neglected. In this paper, all the driving forces for gas transfer were investigated. A momentum-diffusive model is proposed. The model is based on the combined effects of concentration gradients, pressure gradients, and gravity on the transport of gases in the bulk grain. The experimental data for CO2 transfer through connected columns of hot and cold wheat were used to validate the model. The equations were solved using the finite difference method, and the predictions from the proposed model were in good agreement with the experimental results.  相似文献

Although filial therapy has traditionally been viewed as suitable for children of diverse cultures, there is limited research to support this perspective. This qualitative case study examined the perceived effectiveness of filial therapy for a Jamaican mother and the perceived effect on the mother-child relationship. Findings indicate that some aspects of filial therapy are congruent with the parenting practices and values of this Jamaican mother. The mother reported an increase in empathy, a heightened awareness of her child's needs, and a stronger relationship with her child. She expressed some difficulty with limit setting and tracking skills taught as part of the model. Future research directions include further examination of the compatibility of filial therapy with other Jamaican parents and the effectiveness of group filial therapy with Jamaican parents or other parents of Caribbean descent. This report provides X-ray diffraction and Raman spectral evidence that, when 2,6-dichlorobenzonitrile is present in the culture medium, Acetobacter xylinum, which is a model system for investigation of the biosynthesis of native cellulose, produces cellulose II, as well as cellulose I. The significance of the observations with respect to the mechanism of biosynthesis of cellulose is discussed briefly.  相似文献

The current study evaluated immune response to silicon dioxide in children born to women with silicone breast implants. In part one of the study, the T lymphocytes of 21 of 24 such children were significantly stimulated by silicon dioxide (silica). Part two consisted of eleven children, four born preimplantation and seven born postimplantation. None of the preimplant offspring showed T cell responses to silica; five of the seven postimplant children were positive for T cell memory for silica. Part three was a blinded study based on statistically significant differences in T cell stimulation with silicon dioxide between postimplant children and controls. These findings indicate a common immune reaction, that of T cell memory, occurs in mothers and their children born after exposure to silicone mammary implants placed prior to pregnancy. Since not all such children were breast fed the result favors transplacental passage of immunogens such as silicone oligomers or through maternofetal cellular traffic.  相似文献

High doses of morphine produce a state of behavioural inactivity and muscular rigidity. This type of 'catalepsy' is clearly different from the state which is produced by the administration of neuroleptics, e.g. haloperidol. While haloperidol-induced catalepsy can easily be antagonised by NMDA receptor antagonists, there has been a report that the non-competitive NMDA receptor antagonist (+)-5-methyl-10,11-dihydro-5H-dibenzo[a,d] cyclohepten-5,10-imine (MK-801) potentiates morphine-induced catalepsy. The aim of this study was to further examine the role of glutamate receptors in the mediation of morphine-induced catalepsy. To this end we coadministered morphine (20, 40, 60 mg/kg i.p.) with MK-801 (0.1 and 0.3 mg/kg i.p.), the competitive NMDA receptor antagonist DL-(E)-2-amino-4-methyl-5-phosphono-3-pentoic acid (CGP 37849) (2 and 6 mg/kg i.p.), or 1-(4-aminophenyl)-4-methyl-7,8-methylen-dioxy-5H-2,3- benzodiazepine (GYKI 52466) (2 and 4 mg/kg), an antagonist of the AMPA type of glutamate receptors, respectively. The degree of catalepsy was assessed using two different methods, the 'bar/podium/grid' test which is commonly used to measure neuroleptic-induced catalepsy, and a test for the presence or absence of righting reflexes after turning the animals into a supine position. It was found that in the 'bar/podium/grid' test coadministration of both NMDA receptor antagonists significantly and dose-dependently augmented morphine-induced catalepsy. The results using the AMPA receptor antagonist were less clear since the lower dose of GYKI 52466 tended to attenuate the morphine effect whereas the higher dose augmented morphine-induced catalepsy in some cases. While placing the animals on the bar and on the podium produced essentially the same results, the grid was found to be inapplicable for the measurement of morphine-induced catalepsy since the animals did not cling to the grid and fell off almost immediately after being released from the experimenter's hand. With respect to the righting reflexes it was found that the number of animals not showing these responses increased when MK-801 or CGP 37849 was coadministered with morphine. In contrast, most of the animals treated with GYKI 52466 and morphine displayed intact righting reflexes. It is concluded that glutamatergic transmission plays an important role in the mediation of morphine-induced catalepsy, though different to that of haloperidol-induced catalepsy, and that NMDA and AMPA receptors are differentially involved in different aspects of the associated behavioural state.  相似文献

A man, 23 years of age, had a typical erythema infectiosum, complicated by a severe bilateral brachial plexus neuritis. Motor function recovered slowly and only partially after 6 months. An infection by human parvovirus B19 was demonstrated, with strongly positive and gradually declining IgM antibodies and viral DNA detectable in serum for more than 3 months. There was also clear evidence of a recent infection by cytomegalovirus. The interaction between these two viruses could be responsible for this rare and severe complication of common infections in this patient.  相似文献
