A mass balance model for mercury in Lake Champlain was developed in an effort to understand the sources, inventories, concentrations, and effects of mercury (Hg) contamination in the lake ecosystem. To construct the mass balance model, air, water, and sediment were sampled as a part of this project and other research/monitoring projects in the Lake Champlain Basin. This project produced a STELLA-based computer model and quantitative apportionments of the principal input and output pathways of Hg for each of 13 segments in the lake. The model Hg concentrations in the lake were consistent with measured concentrations. Specifically, the modeling identified surface water inflows as the largest direct contributor of Hg into the lake. Direct wet deposition to the lake was the second largest source of Hg followed by direct dry deposition. Volatilization and sedimentation losses were identified as the two major removal mechanisms. This study significantly improves previous estimates of the relative importance of Hg input pathways and of wet and dry deposition fluxes of Hg into Lake Champlain. It also provides new estimates of volatilization fluxes across different lake segments and sedimentation loss in the lake. 相似文献

The ordering of synthetic liquid crystals near surfaces is known to be dependent on the nanoscopic structure and chemical functionality of surfaces. In this letter, we report that the orientational ordering of synthetic liquid crystals on surfaces decorated with viruses is also dependent on the structures of the viruses. Each of the four virions investigated had diameters of approximately 100 nm, but three of the viruses (influenza virus, La Crosse virus, and vesicular stomatitis virus) were enveloped in a lipid bilayer, whereas one virus (adenovirus) was not. We observed that lipid bilayer-enveloped viruses induce homeotropic (perpendicular) ordering of a nematic liquid crystal upon contact with the liquid crystal. In contrast, nonenveloped virus (adenovirus)-treated surfaces caused a near-planar orientation of the liquid crystal. We conclude that the homeotropic ordering of liquid crystals is a signature of the presence of enveloped viruses present on surfaces. These results suggest new approaches to the design of nanostructured materials that incorporate viruses as well as suggest methods that can be used to amplify the presence of nanoscopic virions into micrometer-sized domains of liquid crystal that can be optically probed. 相似文献

Maintaining the integrity of analytical data over time is a challenge. Years ago, data were recorded on paper that was pasted directly into a laboratory notebook. The digital age has made maintaining the integrity of data harder. Nowadays, digitized analytical data are often separated from information about how the sample was collected and prepared for analysis and how the data were acquired. The data are stored on digital media, while the related information about the data may be written in a paper notebook or stored separately in other digital files. Sometimes the connection between this “scientific meta-data” and the analytical data is lost, rendering the spectrum or chromatogram useless. We have been working with ASTM Subcommittee E13.15 on Analytical Data to create the Analytical Information Markup Language or AnIML—a new way to interchange and store spectroscopy and chromatography data based on XML (Extensible Markup Language). XML is a language for describing what data are by enclosing them in computer-useable tags. Recording the units associated with the analytical data and metadata is an essential issue for any data representation scheme that must be addressed by all domain-specific markup languages. As scientific markup languages proliferate, it is very desirable to have a single scheme for handling units to facilitate moving information between different data domains.At NIST, we have been developing a general markup language just for units that we call UnitsML. This presentation will describe how UnitsML is used and how it is being incorporated into AnIML. 相似文献

Thin film samples of CeCl3, CeBr3, and combinatorial compositions along the CeCl3-CeBr3 join were produced using thermal evaporation, which is being evaluated as a method for rapid screening of scintillator materials. The combinatorial thin films were shown to be compositionally reproducible from run-to-run within reasonable limitations. Analytical results suggest a continuous variation in the combinatorial samples in terms of their compositions, crystal structures, and luminescence characteristics. 相似文献

Digital dermatitis is an infectious disease of cattle and the leading cause of lameness. This disease is complicated by the reoccurrence of the lesions and the observation of lesions on more than one limb at different time points, indicating infection may not result in a protective immune response. The objective of this study was to characterize the peripheral blood cellular response in naturally infected and naïve cattle to bacterial antigens derived from pathogens associated with digital dermatitis lesions. Peripheral blood mononuclear cells were isolated from dairy cattle identified as having active or chronic lesions during routine hoof-trimming. Following bacterial antigen stimulation, cells were analyzed for proliferation and phenotype by flow cytometry, and culture supernatants were analyzed for IFN-γ secretion. Digital-dermatitis-infected animals had greater serum antibody titers to treponemal antigens, higher percentages of proliferating CD8+, γδ-T cells, and B cells, and increased IFN-γ secretion in vitro when compared with responses of naïve animals. No increase in proliferation of CD4+ T cells was detected in infected or naïve cattle. Although CD8+ and γδ-T cell responses may be antigen specific, the memory nature or long-lived response is yet unknown. The lack of responsiveness of CD4+ memory cells to treponemal antigens could explain the high rate of reoccurrence of digital dermatitis in infected animals. 相似文献

Helminth parasite eggs in low quality water represent main food safety and health hazards and are therefore important indicators used to determine whether such water can be used for irrigation. Through sedimentation helminth eggs accumulate in the sediment, however resuspension of deposited helminth eggs will lead to increased concentration of suspended eggs in the water. Our study aimed to determine the erodibility (erosion rate and erosion threshold) and settling velocity of Ascaris and Trichuris eggs as well as cohesive sediment at different time points after incorporation into the sediment. Cohesive sediment collected from a freshwater stream was used to prepare a sediment bed onto which helminth eggs were allowed to settle. The erodibility of both sediment and helminth eggs was found to decrease over time indicating that the eggs were incorporated into the surface material of the bed and that this material was stabilized through time. This interaction between eggs and bulk sediment was further manifested in an increased settling velocity of suspended eggs when sediment was present in the suspension as compared to a situation with settling in clean water. The incorporation into the sediment bed and the aggregation with sediment particles decrease the mobility of both helminth egg types. Our findings document that helminth eggs should not be viewed as single entities in water systems when modelling the distribution of eggs since both erodibility and settling velocity of eggs are determined by mobility of the sediment present in the water stream. Recalculation of the erosion threshold for helminth eggs and sediment showed that even at relatively low current velocities i.e. 0.07-0.12 m s−1 newly deposited eggs will be mobile in open irrigation channels. Large Eddy Simulations (LESs) were performed for a confined rectangular liquid jet with a co-flow and compared in detail with particle image velocimetry (PIV) measurements. A finite-volume CFD library, OpenFOAM, was used to discretize and solve the filtered Navier–Stokes equation. The effects of grid resolution, numerical schemes and subgrid models on the LES results were investigated. The second and fourth order schemes gave similar performance, while the fourth order scheme costs much more computationally. Subgrid model comparison showed that the locally dynamic procedure is necessary for complex flow simulation. Model validation was performed by comparing LES data for the point-wise velocity statistics such as the mean and the root-mean-square velocity, shear stress, correlation coefficient, velocity skewness and flatness with the PIV data. In addition, LES data for the two-point spatial correlations of velocity fluctuations that provide structural information were computed and compared with PIV data. Good agreement was observed leading to the conclusion that the LES velocity field accurately captures the important characteristics of all the turbulent length scales present in the flow, from the fully resolved energy-containing eddies to the subgrid-scale dissipative eddies. 相似文献

The objective of this study was to characterize the distribution of Mycobacterium paratuberculosis (Map) in the environment of infected and uninfected Minnesota dairy farms. Eighty herds known to be infected from Minnesota's Johne's Disease Control Program (JDCP) and 28 herds known to be uninfected from Minnesota Voluntary Johne's Disease Herd Status Program (VJDHSP) were sampled. Fecal samples from up to 100 cows in each herd were cultured in pools of 5 cows. Two environmental samples were obtained from each farm from various locations. All samples were tested using bacterial culture for Map. Eighty percent of the JDCP herds had at least one positive pool. Environmental samples were cultured positive in 78% of the JDCP herds. Two (7%) of the VJDHSP herds had one positive pool, and one herd had one positive environmental sample. Environmental samples were cultured positive in cow alleyways (77% of the herds), manure storage (68%), calving area (21%), sick cow pen (18%), water runoff (6%), and postweaned calves areas (3%). There was an association between maximum level of colonies per tube from cow alleyways and manure storage and fecal pool prevalence. Herds with both areas cultured negative were estimated to have 0.3 to 4% fecal pool prevalence. Herds with both areas having a heavy load of bacteria were estimated to have 53 to 73% fecal pool prevalence. The study results indicate that targeted sampling of cow alleyways and manure storage areas appears to be an alternative strategy for herd screening and Johne's infection status assessment and for estimating herd fecal prevalence. 相似文献
