vacB, a gene previously shown to be required for expression of virulence in Shigella and enteroinvasive Escherichia coli, has been found to encode the 3'-5' exoribonuclease, RNase R. Thus, cloning of E. coli vacB led to overexpression of RNase R activity, and partial deletion or interruption of the cloned gene abolished this overexpression. Interruption of the chromosomal copy of vacB eliminated endogenous RNase R activity; however, the absence of RNase R by itself had no effect on cell growth. In contrast, cells lacking both RNase R and polynucleotide phosphorylase were found to be inviable. These data indicate that RNase R participates in an essential cell function in addition to its role in virulence. The identification of the vacB gene product as RNase R should aid in understanding how the virulence phenotype in enterobacteria is expressed and regulated. On the basis of this information we propose that vacB be renamed rnr.  相似文献

Increasing emphasis is now being placed on the assessment of patient outcomes, both in evaluating medical interventions and in quality assurance initiatives. Clinicians, purchasers, managers and researchers need outcome measures that are valid, reliable and responsive. This paper describes the theory and practice underlying the development of outcome measures for two chronic conditions, asthma and diabetes, for application in ambulatory settings. Existing generic and condition-specific health status and health-related quality-of-life measures were administered to almost 1300 patients. The psychometric properties of these measures were examined to identify those that were of adequate validity and reliability in these population groups. Step-wise regression procedures were then used to identify a core set of scales that best predicted patients' general health perceptions, which could be used in measuring general health outcomes for each of these groups. These core sets consist of up to 40 items, spanning physical function, energy and vitality, emotional well-being and condition-specific aspects of health such as symptom control. Further analysis is being carried out to assess the responsiveness to change of these core item sets.  相似文献

NMR (1H and 31P) and HPLC techniques were used to study the partitioning of phosphoramide mustard (PM) and its aziridinium ions among alkylation and P-N bond hydrolysis reactions as a function of the concentration and strength of added nucleophiles at 37 degrees C and pH 7.4. With water as the nucleophile, bisalkylation accounted for only 10-13% of the product distribution given by PM. The remainder of the products resulted from P-N bond hydrolysis reactions. With 50 mM thiosulfate or 55-110 mM glutathione (GSH), bisalkylation by a strong nucleophile increased to 55-76%. The rest of the PM was lost to either HOH alkylation or P-N bond hydrolysis reactions. Strong experimental and theoretical evidence was obtained to support the hypothesis that the P-N bond scission observed at neutral pH does not occur in the parent PM to produce nornitrogen mustard; rather it is an aziridinium ion derived from PM which undergoes P-N bond hydrolysis to give chloroethylaziridine. In every buffer studied (bis-Tris, lutidine, triethanolamine, and Tris), the decomposition of PM (with and without GSH) gave rise to 31P NMR signals which could not be attributed to products of HOH or GSH alkylation or P-N bond hydrolysis. The intensities of these unidentified signals were dependent on the concentration of buffer.  相似文献

Of all the 95 primary tracheal carcinomas diagnosed in Finland in 1967-1985, 72% were squamous cell carcinomas. Fifty-three percent were located in the lower third of the trachea. main symptoms were dyspnea, cough and hemoptysis. Fifty-nine percent of the patients developed extratracheal growth during the follow-up of 12 years, with lungs, neck, liver, and bones being the most common sites. In addition, autopsy revealed mediastinal metastases in 25% of patients. Vocal cord palsy at the time of diagnosis was present in 33%. The size and location of tumor in the trachea did not affect survival significantly. Patients with adenoid cystic carcinomas had the best prognosis.  相似文献

Absorbance changes are reported associated with Photosystem II and showing a periodicity of two and four as a function of flash number. The absorbance changes showing a periodicity of two were found to occur in the presence of artificial electron donors as well and are presumably caused by the secondary electron acceptor R of Photosystem II. The absorbance difference spectra suggest that R is a plastoquinone molecule, which is reduced to its semiquinone anion after an uneven number of flashes. After an even number of flashes, the semiquinone is reoxidized. The absorbance changes showing a periodicity of four are tentatively ascribed to the charge accumulating donor complex of Photosystem II.  相似文献
