Phospholipids of many cold-tolerant organisms have been reported to contain more unsaturated fatty acids than cold-susceptible organisms, a phenomenon known to maintain membrane fluidity at low temperature. However, we have obtained results to the contrary through a comparison of the membrane phospholipids of six temperate and subtropical species belonging to the Drosophila melanogaster species group. With enhancement of cold tolerance, the percentages of monoenoic acids increased but the percentages of dienoic acids decreased, that is, the number of double bonds in the phospholipid decreased without a marked variation in the percentages of unsaturated fatty acids. Concomitantly, the percentage of fatty acids containing 16 carbon atoms increased, while that of fatty acids with 18 carbon atoms decreased. Since phosphatidylethanolamine is a dominant phospholipid in Drosophila, these changes probably contribute to keeping the homeoviscosity of the cellular membranes in a manner different to that in phosphatidylcholine-rich membranes, thereby increasing cold tolerance.  相似文献

The observations in both mouse and rat models of experimental allergic encephalomyelitis (EAE) demonstrating restricted T-cell receptor (TCR) usage among pathogenic T cells has led to the generation of a new class of therapeutic vaccines composed of TCR V region peptides. Whether a similar approach will be of use in the treatment of human autoimmune disorders is still unclear. The experiments performed in our laboratory over the past several years have focused on two aspects of TCR peptide immunoregulation, namely, (1) how to identify the critical T-cell populations involved in the pathology of autoimmune disease, and (2) how to identify biologically relevant TCR peptides--those endogenous TCR peptides presented in association with MHC molecules on the surface of pathogenic T cells that are recognized by immunoregulatory T-cell populations. Results of our recently completed clinical studies regarding TCR V beta expression among CD4+ T cells in the cerebral spinal fluid (CSF) of patients with multiple sclerosis suggests that these cells may be an appropriate T-cell population to be targeted for TCR peptide therapy. In addition, our studies on the immune response to autologous, soluble TCR heterodimers may provide a strategy for the identification of new TCR peptide candidate vaccines.  相似文献

We report the case of a woman who, at the age of 27, developed a cerebral arterial occlusion. The laboratory investigations showed an anti-human beta2-glycoprotein I antibody, but no other biological sign of antiphospholipid antibody syndrome or autoimmune disorders. The patient otherwise presented with diabetes and moderate obesity. The species specificity of anti-beta2-glycoprotein I antibodies probably explains the discrepancy between false negative results for antiphospholipid antibodies assayed by clotting and ELISA studies and positivity for anti-human beta2-glycoprotein I. Further studies will be important to evaluate the frequency of such antibodies, as well as their value as a risk factor for venous and arterial thrombosis, and their signification within the antiphospholipid antibody syndrome.  相似文献

The continuity between early smoking experimentation and smoking at age 16 years was analysed for a birth cohort of New Zealand children. This analysis suggested that when due allowance was made for errors in reports of smoking behaviours, there was evidence of relatively strong continuity (r = 0.60) between early smoking experimentation and cigarette smoking at age 16 years. Further analysis suggested that the continuities between early smoking experimentation and later smoking arose from three pathways that linked early smoking experimentation to later smoking. First there was evidence to suggest that children who engaged in early smoking experimentation tended to affiliate with adolescent peer groups whose members smoked. In turn, these peer group affiliations reinforced pre-existing tendencies to cigarette smoking. Secondly, a small component of the apparent continuity between early smoking experimentation and later smoking arose because of common social, individual and contextual factors that were associated with both smoking experimentation and later smoking. Finally, there was evidence of moderate direct continuity in cigarette smoking behaviour over time. The implications of these findings for the development of smoking prevention programmes are discussed and it is concluded that effective programmes need to be embedded in a developmental approach which attempts to reduce both early smoking experimentation and the effects of peer pressure in adolescence on the development of cigarette smoking.  相似文献

We used Xenopus egg extracts to examine the effects of TPEN, a chelator with strong affinities for Zn2+, Fe2+, and Mn2+, on nuclear assembly in vitro. At concentrations above 1 mM, TPEN blocked the assembly of the nuclear lamina and produced nuclei that were profoundly sensitive to stress-induced balloon-like 'shedding' of nuclear membranes away from chromatin-associated membranes. TPEN-arrested nuclei were also defective for DNA replication, which could be explained as secondary to the lack of a lamina. Imaging of TPEN-arrested nuclei by field emission in-lens scanning electron microscopy (FEISEM) revealed clustered, structurally-perturbed nuclear pore complexes. TPEN-arrested nuclei were defective in the accumulation of fluorescent karyophilic proteins. All detectable effects caused by TPEN were downstream of the effects of BAPTA, a Ca2+/Zn2+ chelator that blocks pore complex assembly at two distinct early stages. Surprisingly, TPEN-arrested nuclei, but not control nuclei, remained active for replication in apoptotic extracts, as assayed by [32P]-dCTP incorporation into high molecular weight DNA, suggesting that TPEN blocks a metal-binding protein(s) required for nuclear destruction during programmed cell death.  相似文献

We have identified several strains of Escherichia coli which contain immunoglobulin-binding activity on the cell surface. Affinity-purified antibodies ordinarily used as secondary antibodies in immunodetection protocols were bound by 6 of 72 strains of the ECOR reference collection of E. coli. The Fc fragments of both human and sheep immunoglobulin G (IgG) were also bound, demonstrating the nonimmune nature of the phenomenon. Binding of conjugated IgG Fc directly to unfixed cells was observed by fluorescence microscopy. Western blots showed that the immunoglobulin-binding material occurs in the form of multiple bands, with the apparent molecular masses of the most prominent bands exceeding 100 kDa. No two of the strains have the same pattern of bands. The binding activity in extracts was sensitive to proteinase K. The binding activity of intact cells was reduced preferentially by trypsin digestion, demonstrating exposure at the cell surface. Expression of binding activity in Luria-Bertani broth cultures was favored by a temperature of 37 degrees C and entry into stationary phase of growth.  相似文献

Both silent and spontaneously firing spiny projection neurons have been described in the neostriatum, but the reason for their differences in firing activity are unknown. We compared properties of spontaneously firing and silent spiny neurons in urethan-anesthetized rats. Neurons were identified as spiny projection neurons after labeling by intracellular injection of biocytin. The threshold for action-potential firing was measured under three different conditions: 1) electrical stimulation of the contralateral cerebral cortex, 2) brief directly applied current pulses, and 3) spontaneous action-potentials occurring during spontaneous episodes of depolarization ( state). The average membrane potential and the amplitude of noiselike fluctuations of membrane potential in the state were determined by fitting a Gaussian curve to the membrane-potential distribution. All neurons in the sample exhibited spontaneous membrane potential shifts between a hyperpolarized state and a depolarized state, but not all fired action potentials while in the state. The difference between the spontaneously firing and the silent spiny neurons was in the average membrane potential in the state, which was significantly more depolarized in the spontaneously firing than in the silent spiny neurons. There were no significant differences in the threshold, the amplitude of the noiselike fluctuations of membrane potential in the state, or in the proportion of time that the membrane potential was in the state. In both spontaneously firing and silent neurons, the threshold for action potentials evoked by current pulses was significantly higher than for those evoked by cortical stimulation. Application of more intense current pulses that reproduced the excitatory postsynaptic potential rate of rise produced firing at correspondingly lower thresholds. Because the membrane potential in the state is mainly determined by the balance between the synaptic drive and the outward potassium conductances activated in the subthreshold range of membrane potentials, either or both of these factors may determine whether firing occurs in response to spontaneous afferent activity.  相似文献

The susceptibilities of 143 Porphyromonas gingivalis, Prevotella intermedia, and Prevotella nigrescens isolates to 18 antimicrobial agents were tested. All P. gingivalis isolates were susceptible. In contrast, some Prevotella spp. (17%) were resistant to beta-lactams, erythromycin, clindamycin, or tetracycline and carried resistance genes, ermF or tetQ, or beta-lactamases.  相似文献

The authors present an electrical design inspection (EDI) methodology that combines advanced power circuit simulation techniques and RISC (reduced instruction set computing) workstation hardware to use simulation in the day-to-day design of electronic power supplies. This methodology makes use of circuit simulation to detect design faults in electronic power supplies and prevent them from propagating further in the product realization process. A hierarchy of inspections which form the basis of EDI methodology, is introduced. The methodology has been embedded in a prototype electrical design inspection system which has been tested on a Sun Sparcserver 4/490 dedicated to circuit simulation. The power of this methodology has been illustrated by its application to a self-oscillating variable-frequency DC-DC power converter with peak current control. It is demonstrated that EDIS can automatically execute inspections requiring an accurate determination of the steady-state solution of the circuit, and process these results. The steady-state accelerator capability within the SIMPLIS circuit simulator has made it possible to achieve this in an unprecedentedly short CPU time  相似文献
