Use of [6,6-2H2]glucose and of low-enrichment [U-13C6]-glucose for sequential or simultaneous measurements of glucose turnover by gas chromatography-mass spectrometry

We developed gas chromatography-mass spectrometric methods for assaying the enrichment of 99 at.% [6,6-2H2]glucose and 30 at.% [U-13C6]glucose, although both tracers are mostly M + 2. 13C enrichment is determined either by the C-1 to C-5 fragment of glucose aldonitrile pentaacetate or by oxidation of glucose to glucarate. 2H enrichment is assayed as the difference between the 13C enrichment of glucarate and the 2H + 13C enrichment of glucose. The techniques, which were validated in in vivo experiments, are applicable to the determination of simultaneous or sequential measurements of the rate of glucose appearance before and after an intervention. They could also be applied to the simultaneous determination of (i) gluconeogenesis by incorporation of a 13C-labeled precursor into glucose and (ii) the rate of glucose appearance by [6,6-2H2]glucose infusion.

The murine monoclonal antibody A7 (MAb A7) is reactive against most human gastric cancer cell lines. Using a nude mouse peritoneal dissemination model of human gastric cancer, we investigated targeted chemotherapy using a conjugate of neocarzinostatin (NCS) with MAb A7 (A7-NCS). After demonstrating cytotoxicity of the complex against the human gastric cancer cell line MKN45 in vitro, we intraperitoneally injected A7-NCS, NCS or saline into nude mice bearing peritoneally disseminated human gastric cancer. A7-NCS inhibited peritoneal dissemination significantly more effectively than NCS. MAb A7 may prove to be an effective carrier for antineoplastic drugs in patients with peritoneal dissemination of gastric cancer.     

Contribution to modelling of process planning

The process planning activities have been performed up to now manually and based on personal experience of those who are engaged. Development in computer based planning attempts to make the planning process to be carried out easier for the staff and to make use of experiences gained in the past with help of expert systems. The two principles used are the variant and the generic approach.     

Patterns of chloroform-induced regenerative cell proliferation in BDF1 mice correlate with organ specificity and dose-response of tumor formation

Drawing arm movements in four different directions: a) upward vertical (0 degree), b) upward oblique (45 degrees), c) downward vertical (180 degrees) and d) downward oblique (135 degrees), and at two different speeds, normal and fast, were executed by eight subjects. Movements of the arm were recorded using an optoelectronic (2 TV, 100 Hz) system which allowed the computer reconstruction of joint motion. Analyses focused upon pen kinematics in the frontal plane. Velocity profiles were unimodal for all conditions. The ratio of acceleration time to total movement time changed significantly as a function of the direction and the speed of the movement. Movement time and was not affected by movement direction and consequently changes in gravitational torques, for both speeds tested. Results from this study provide indirect evidence that the CNS executes movements by taking advantage of gravitational force.
