Physiological regulation of early and late stages of megakaryocytopoiesis by thrombopoietin

Thrombopoietin (TPO) has recently been cloned and shown to regulate megakaryocyte and platelet production by activating the cytokine receptor c-mpl. To determine whether TPO is the only ligand for c-mpl and the major regulator of megakaryocytopoiesis, TPO deficient mice were generated by gene targeting. TPO-/- mice have a >80% decrease in their platelets and megakaryocytes but have normal levels of all the other hematopoietic cell types. A gene dosage effect observed in heterozygous mice suggests that the TPO gene is constitutively expressed and that the circulating TPO level is directly regulated by the platelet mass. Bone marrow from TPO-/- mice have decreased numbers of megakaryocyte-committed progenitors as well as lower ploidy in the megakaryocytes that are present. These results demonstrate that TPO alone is the major physiological regulator of both proliferation and differentiation of hematopoietic progenitor cells into mature megakaryocytes but that TPO is not critical to the final step of platelet production.     

Excess mortality in general hospital patients with delirium: a 5-year follow-up of 519 patients seen in psychiatric consultation

Mortality was determined in 519 patients with delirium who were seen in psychiatric consultation in two general hospitals. Among 419 patients with simple delirium (DSM-III: 293.00) in-hospital mortality was 26%. As compared to average hospital patients the age adjusted in-hospital excess mortality ratio varied from 6.2 for patients with malignancies to 2.1 for patients with motor system disease. After hospital discharge the 5-yr cumulative mortality was 51%. As compared to the general population excess mortality was noted in most, but not in all diagnostic subgroups. The age and sex adjusted excess mortality ratio varied from 14.1 for malignancies to 1.3 for motor system disease. The figures underline a general notion that delirium may be an indicator of disorders of grave prognosis, but mortality appears to depend more on the medical condition than on the presence of delirium.     

Influence of water and accelerated aging on the shear fracture properties of glass/epoxy composite

The degradation of interlaminar shear strength and shear fracture toughness of glass/epoxy composites due to uptake of distilled water and sea water has been studied. The composites were immersed in water for up to eight months at temperatures up to 70 °C. Unreinforced matrix resin samples were also immersed for periods up to 2 years. Sea water was absorbed less rapidly than distilled water. Weight gains below 1% did not influence the shear strength while higher weight gains reduced shear strength up to 25%. The loss in apparent interlaminar shear strength was uniquely related to specimen weight gain. Mode II fracture toughness, G _IIc, also decreased with increasing immersion time after an initial incubation period, but the accelerated tests were found to reduce G _IIc less than the room temperature tests at comparable weight gains.     

Compact optical millimetre-wave source using a dual-modesemiconductor laser

The authors demonstrate the optical generation of extremely narrow linewidth millimetre-wave signals between 40 and 60 GHz using a single-chip semiconductor laser. A dual-mode long-cavity multisection DFB semiconductor laser is driven at a subharmonic of the free-running-mode beat signal frequency to produce phase-locked millimetre waves with a 3 dB linewidth of less than 10 Hz and a 3 dB locking range of ~500 MHz     

The peeling of flexible probabilistic fasteners

Probabilistic fasteners are attachment devices consisting of two elements each of which is covered by an array of outgrowths or protuberances which interact or interlock with those on the opposing surface to form a bond which can be made and detached many times. A number of peel tests are described in which the behaviour of such bonds, specifically those formed by Velcro® hooks and loops and Dual Lock®, has been explored. The value of the specific energy of detachment of such mechanically latched systems can be related to the geometry and material properties of the individual fixing elements. Similar arguments can be applied, both qualitatively and quantitatively, to some natural systems such as the spines on some cacti.     

Transient conduction in a slab with chemical reaction

The heat-balance integral technique is used to analyse a one-dimensional transient conduction process. Heat is transferred by convection to the surface of a slab and when the surface temperature reaches a certain value, an endothermic reaction is triggered which propagates as an isothermal front in the same direction as the conduction front. Two possible routes to the fully developed heat transfer condition are treated in a way which allows the approximate temperature distribution in the slab to be predicted as a function of the main process variables.     

Designer deletion strains derived from Saccharomyces cerevisiae S288C: a useful set of strains and plasmids for PCR-mediated gene disruption and other applications

A set of yeast strains based on Saccharomyces cerevisiae S288C in which commonly used selectable marker genes are deleted by design based on the yeast genome sequence has been constructed and analysed. These strains minimize or eliminate the homology to the corresponding marker genes in commonly used vectors without significantly affecting adjacent gene expression. Because the homology between commonly used auxotrophic marker gene segments and genomic sequences has been largely or completely abolished, these strains will also reduce plasmid integration events which can interfere with a wide variety of molecular genetic applications. We also report the construction of new members of the pRS400 series of vectors, containing the kanMX, ADE2 and MET15 genes.
