An accurate analysis of the morphological changes which take place during pathological processes of the posterior pole is important for a correct diagnosis and therapeutic approach. The purpose of the study was to determine the intraobserver and interobserver reproducibility of the Image-net system 100 (Topcon, Japan) to take measurements on the retina. The program 'Linear/Areal Measurement functions' of Image-net system 100 which is an image digitalization technique, was tested. Twelve patients were consecutively selected from the patients of the Retina Center of the Department of Ophthalmology, University of Genoa. Three images of each eye were taken from each subject and only the best image was used in this study. The intraobserver and interobserver reproducibility of both the distance between two pre-set points (linear measurement), and the perimeter and area of preselected retinal zones were calculated. The repeatability (or intraobserver reproducibility) of the linear sizes was measured by the coefficient of variation and ranged from 0.32% to 7.38%, while the interobserver reproducibility ranged from 0.46% to 5.22%. The repeatability and reproducibility of the perimeters ranged from 0.72% to 9.63% and from 0.6% to 5.7%, respectively, while the repeatability and reproducibility of the areas ranged from 0.72% to 9.63% and from 0.6% to 5.7%, respectively. Although the results were quite good, the quality of the image of the fundus and the number of observers influenced the coefficient of variation; furthermore, the anatomy of the areas to be measured and the computer 'mouse' could increase the value of the coefficient of variation.  相似文献

Lipocortin 1 (LC1) and S100-beta, two Ca(2+)-binding proteins that serve as specific markers for microglia and astrocytes, respectively, have been used to study postnatal gliogenesis in the rat optic nerve. Computerized image analysis was used to quantify and map the stained and unstained glia in transverse sections (10 microns thick) taken 1-2 mm from the chiasm in optic nerves from rat pups at postnatal day 0 (P0), P7, P14, P21, P28, P38 and adults. The number of astrocytes was remarkably constant (100 per section) at all ages. Because the area of the nerve increases 10-fold from P0 to adult, the population density of astrocytes begins al > 5000 mm-2 and drops to 400 mm-2 in the mature nerve; however, because the nerve length increases two-fold, the number of astrocytes doubles over the same period. In contrast, the number of LC1 + cells per section initially is sparse (4 at P0), increases rapidly up to 36 at P21 and levels off at 49 in adults. The microglia population density is relatively stable throughout development (200-300 mm-2) except during the peak of oligodendroblast apoptosis (P21) when it rises to 450 mm-2. Neonatally, LC1 immunoreactivity predominantly labels spherical-ameboid cells; but by P28 they are replaced by mature ramified microglia. The number of unstained cells (putative oligodendrocytes) per section increases from 11 at P0 to a peak of 308 at P21, and declines slightly to 269 in adults. While generally confirming concepts of astrocyte and oligodendrocyte ontogeny from the literature, the present report adds considerable detail regarding microglia, which often have been ignored. Microglia identified by LC1 immunoreactivity comprise 12% of the glia in adult optic nerve near the chiasm.  相似文献

Experimental investigations of eukaryotic enhancers suggest that multiple binding sites and trans-acting regulatory factors are often required for wild-type enhancer function. Genetic analysis of the stripe 2 enhancer of even-skipped (eve), an important developmental gene in Drosophila, provides support for this view. Given the importance of even-skipped expression in early Drosophila development, it might be predicted that many structural features of the stripe 2 enhancer will be evolutionarily conserved, including the DNA sequences of protein binding sites and the spacing between them. To test this hypothesis, we compared sequences of the stripe 2 enhancer between four species of Drosophila: D. melanogaster, D. yakuba, D. erecta and D. pseudoobscura. Our analysis revealed a large number of nucleotide substitutions in regulatory protein binding sites for bicoid, hunchback, Kruppel and giant, as well as a systematic change in the size of the enhancer. Some of the binding sites in D. melanogaster are either absent or modified in other species. One functionally important bicoid-binding site in D. melanogaster appears to be recently evolved. We, therefore, investigated possible functional consequences of sequence differences among these stripe 2 enhancers by P-element-mediated transformation. This analysis revealed that the eve stripe 2 enhancer from each of the four species drove reporter gene expression at the identical time and location in D. melanogaster embryos. Double staining of native eve protein and transgene mRNA in early embryos showed that the reporter gene mimicked native eve expression and, in every case, produced sharply defined stripes at the blastoderm stage that were coincident with eve stripe 2 protein. We argue that stripe 2 eve expression in Drosophila evolution can be viewed as being under constant stabilizing selection with respect to the location of the anterior and posterior borders of the stripe. We further hypothesize that the stripe 2 enhancer is functionally robust, so that its evolution may be governed by the fixation of both slightly deleterious and adaptive mutations in regulatory protein binding sites as well as in the spacing between binding sites. This view allows for a slow but continual turnover of functionally important changes in the stripe 2 enhancer.  相似文献

In this article,a novel unordered classification rule list discovery algorithm is presented based on Ant Colony Optimization(ACO). The proposed classifier is compared empirically with two other ACO-based classification techniques on 26 data sets,selected from miscellaneous domains,based on several performance measures. As opposed to its ancestors,our technique has the flexibility of generating a list of IF-THEN rules with unrestricted order. It makes the generated classification model more comprehensible and easily interpretable.The results indicate that the performance of the proposed method is statistically significantly better as compared with previous versions of AntMiner based on predictive accuracy and comprehensibility of the classification model.  相似文献

Anonymization is a practical approach to protect privacy in data. The major objective of privacy preserving data publishing is to protect private information in data whereas data is still useful for some intended applications, such as building classification models. In this paper, we argue that data generalization in anonymization should be determined by the classification capability of data rather than the privacy requirement. We make use of mutual information for measuring classification capability for generalization, and propose two k-anonymity algorithms to produce anonymized tables for building accurate classification models. The algorithms generalize attributes to maximize the classification capability, and then suppress values by a privacy requirement k (IACk) or distributional constraints (IACc). Experimental results show that algorithm IACk supports more accurate classification models and is faster than a benchmark utility-aware data anonymization algorithm.  相似文献

Various synthetic fibres, namely, polyethylene terephthalate, nylon 6.6, acrylic and spandex fabrics, were dyed with indigo blue using an exhaust technique. The factors studied were sodium dithionite and sodium hydroxide concentrations, liquor/material ratio, dyeing temperatures and process times. The pH levels of the dyebaths were achieved by controlling the ratios of sodium dithionite and sodium hydroxide concentrations. Dyebath analysis at the end of dyeing showed that, in different pH regions, different forms of the indigo existed and each one exhibited different degrees of substantivity for synthetic fibres. The nonionic form exhibited substantivity for all the synthetic fibres, while the mono‐sodium phenolate form showed substantivity only in the acidic range. Synthetic fibres could best be dyed in the pH range of 5.5–6. Reflections spectra showed that indigo in synthetic fibres and when dissolved in dimethyl formamide absorbed maximally at 610 nm, while in cotton indigo absorbed at 660 nm. The fastness properties were measured and the results showed that washing and rubbing were good, while light and sublimation were moderate to poor.  相似文献
