Through proteolysis and peptide mass determination using mass spectrometry, a peptide mass map (PMM) can be generated for protein identification. However, insufficient peptide mass accuracy and protein sequence coverage limit the potential of the PMM approach for high-throughput, large-scale analysis of proteins. In our novel approach, nonlabile protons in particular amino acid residues were replaced with deuteriums to mass-tag proteins of the S. cerevisiae proteome in a sequence-specific manner. The resulting mass-tagged proteolytic peptides with characteristic mass-split patterns can be identified in the data search using constraints of both amino acid composition and mass-to-charge ratio. More importantly, the mass-tagged peptides can further act as internal calibrants with high confidence in a PMM to identify the parent proteins at modest mass accuracy and low sequence coverage. As a result, the specificity and accuracy of a PMM was greatly enhanced without the need for peptide sequencing or instrumental improvements to obtain increased mass accuracy. The power of PMM has been extended to the unambiguous identification of multiple proteins in a 1D SDS gel band including the identification of a membrane protein.  相似文献

ZnO nano layer was prepared by sol–gel method using zinc acetate dihydrate, methanol and diethanolamine as precursor, solvent and stabilizer, respectively. According to the thermal analysis results, the heat treatment process after producing ZnO nano layer can be divided into three stages. The structure of the nano layer showed a significant dependency on heat treatment temperature in the low temperature heat treatment stage, which is performed to evaporate the organic components. The high temperature heat treatment stage has no significant effect on the structure of the films. The temperature employed in the first stage of heat treatment process during the initial 10 min of heat treatment was found to be important in determining the structure of the films. The results determined that the elimination condition of the organic components is very important for achieving high crystallinity.  相似文献

Encapsulation of cellulose chain into carbon nanotubes and boron nitride nanotubes was investigated to find out the possibility of band gap engineering in these nanotubes. The structural stability and the electronic properties of the zigzag carbon nanotubes and boron nitride nanotubes filled with cellulose chain were studied using density functional theory. It was found that encapsulation of cellulose chain into nanotubes was an exothermic process. The metallic properties of the carbon nanotubes did not change by cellulose encapsulation. The semiconductor and insulator nanotubes filled with cellulose were shown semiconducting properties. The energy band gap of these tubes was decreased by cellulose encapsulation. The results demonstrated the ability of band gap engineering through the encapsulation of cellulose chain into carbon nanotubes and boron nitride nanotubes.  相似文献

In this paper the statistical data of fifty days' wind speed measurements at the MERC-solar site are used to find out the wind energy density and other wind characteristics with the help of the Weibull probability distribution function. It is emphasized that the Weibull and Rayleigh probability functions are useful tools for wind energy density estimation but are not quite appropriate for properly fitting the actual wind data of low mean speed, short-time records. One has to use either the actual wind data (histogram) or look for a better fit by other models of the probability function.  相似文献

Green chemical method could be a promising route to achieve large scale synthesis of nanostructures for biomedical applications. Here, we describe a green chemical synthesis of silver nanoparticles (Ag NPs) on chitosan‐based electrospun nanofibers using Eucalyptus leaf extract. A series of silver salt (AgNO3) amounts were added to a certain composition of chitosan/polyethylene oxide aqueous acetic acid solution. The solutions were then electrospun to obtain nanofibrous mats and then, morphology and size of nanofibers were analyzed by scanning electron microscopy (SEM). Incubation of AgNO3‐containing mats into Eucalyptus leaf extract led to the formation of Ag NP clusters with average diameter of 91 ± 24 nm, depicted by SEM and transmission electron microscopy. Surface enhanced Raman spectroscopy also confirmed formation of Ag NPs on the nanofibers. The mats also showed antimicrobial activity against Escherichia coli and Staphylococcus aureus bacteria with bigger inhibition zone for extract‐exposed mats against S. aureus. © 2015 Wiley Periodicals, Inc. Microstructures have profound effects on the hardness and strength of Cu-Cr alloys. The microstructures of a Cu-Cr alloy cast in a water-cooled copper mold were studied in the present work. Tunable rigidity materials have potentially widespread implications in robotic technologies. They enable morphological shape change while maintaining structural strength, and can reversibly alternate between rigid, load bearing and compliant, flexible states capable of deformation within unstructured environments. In this review, we cover a range of materials with mechanical rigidity that can be reversibly tuned using one of several stimuli (e.g. heat, electrical current, electric field, magnetism, etc.). We explain the mechanisms by which these materials change rigidity and how they have been used for robot tasks. We quantitatively assess the performance in terms of the magnitude of rigidity, variation ratio, response time, and energy consumption, and explore the correlations between these desired characteristics as principles for material design and usage.  相似文献
