Hepatic lipid characteristics and histopathology of laying hens fed CLA or n−3 fatty acids 总被引:2,自引:0,他引:2
The effect of dietary CLA and n−3 PUFA on hepatic TAG accumulation, histopathology, and FA incorporation in lipid classes
by laying chickens was investigated. One hundred twenty 30-wk-old single-comb white leghorn laying hens were distributed randomly
to four treatments (3 replications of 10 birds) and were fed diets containing CLA and animal fat (Diet I), 18∶3n−3 (Diet II),
or long-chain n−3 FA (Diet III). A sunflower oil (n−6 FA)-based diet was the control. Feeding Diet I resulted in an increase
in hepatic total lipids (P<0.05). The liver TAG content was 32.2, 18.9, 29.4, and 18.7 mg/g for hens fed Diet I, Diet II, Diet III, and the control
diet, respectively (P<0.05). The serum TAG was lowest in bilds fed Diet II (P<0.05). Diet I resulted in an increase in the total number of fat vacuoles and lipid infiltration in hepatocytes (P<0.05). The number of cells with 75% or higher lipid vacuolation was observed only in birds fed Diet I. Feeding diets containing
CLA resulted in an increase in the content of the c9,t11 CLA isomer in liver TAG and PC (P<0.05). No difference was observed in the CLA concentration of hepatic PE fractions. The content of DHA (22∶6n−3) was higher
in the TAG, PC, and PE of hens fed Diet II and Diet III than Diet I and the control (P<0.05). Feeding CLA resulted in an increase in total saturated FA in the TAG and PC fractions (P<0.05). Long-term feeding of CLA in laying birds leads to an increase in liver TAG and may predispose birds to fatty liver

Structural genomics initiatives are determining thousands of new protein structures. Many of these structures are of unknown function, and computational methods for the rapid determination of functional information from protein structure are needed. We present details of how functional information is obtained from the structure using THEMATICS (Theoretical Microscopic Titration Curves). THEMATICS is a computational procedure that gives information about chemical reactivity, based on solution of the Poisson-Boltzmann equations for the electrical potential function. We show how anomalies in predicted titration curves are established. We show further that when residues with anomalous predicted titration curves form a cluster in physical space, these residues tend to be very highly conserved across species and such clusters are reliable predictors of the active site. Results are given for ten enzymes; detailed results are shown for the enzymes triosephosphate isomerase (from chicken), 6-hydroxymethyl-7,8-dihydropterin pyrophosphokinase (from E. coli), and papain (from papaya). 相似文献

A novel host–guest system was developed by the encapsulation of simple organic guest molecules in the hydrophilic molecular architecture of crosslinked polyacrylamide hydrogels. The crosslinking agents used for the preparation of the host systems were hexanediol dimethacrylate (HDDMA) and divinyl benzene (DVB). This enabled us to construct hydrogels with different hydrophobic–hydrophilic equilibria. The model guest system used for the studies was benzoic acid. The selections gave simple but excellent host–guest systems with fine polar–apolar balancing. Polyacrylamide hydrogels with encapsulated benzoic acid were prepared with varying crosslink densities (5, 10, 15, and 20 mol %) by the solution polymerization technique. The rate of release of the host from the host–guest assembly was studied in different swelling conditions. The rate of release depended on the interaction forces between the polymer and the solvents. Polar forces, dispersion forces, and hydrogen bonding all played a vital role. The swelling behavior of the host‐polymer system and the host–guest assembly was analyzed and compared by the Flory–Rehner method. The amount of benzoic acid encapsulated in the DVB‐crosslinked polymer was higher than in the HDDMA‐crosslinked polymer, and the rate of release was in the order 5 > 15 > 10 > 20% for the DVB‐crosslinked polymer. The rate of release for the HDDMA‐crosslinked host–guest assembly was in the order 10 > 5 > 15 > 20%. In drug delivery systems that use silicone elastomers as a diffusion matrix for the active drug, it is common to crosslink the material by the hydrosilylation reaction. In this platinum‐catalyzed reaction, vinyl groups on a polymer add to the methyl siloxane hydride (MHS) groups on a low molecular mass crosslinker. With an excess of crosslinker, a fast curing is achieved and a fully crosslinked material is formed. Unreacted MHS groups were shown to remain in the elastomer after curing because of the excess crosslinker. In this work, a simple procedure was developed to eliminate the unreacted MHS groups from the final product. We found that storage of the product at +40°C and 75% relative humidity for a few weeks will effectively destroy the residual MHS groups in the elastomer. The effects of varying levels of humidity, oxygen, and temperature on this postcuring procedure were studied. The amount of MHS groups was measured with NMR and IR spectroscopy. We also found that the hardness of the material increased by approximately 25% as a consequence of this postcuring treatment. This increase is probably due to a secondary crosslinking reaction between MHS and silanol groups. Heat treatment at higher temperatures led to an even further increase in the hardness and compression modulus. Because no MHS groups remained in the elastomer when this heat treatment was started, it is apparent that another secondary crosslinking reaction is occurring, probably silanol condensation. © 2002 Wiley Periodicals, Inc. AbstractThis article introduces a case study undertaken in the indigenous Penan community of Long Lamai, Upper Baram, Sarawak, Malaysia. In this community, there is concern about the negative image other cultural groups hold of the Penan. This case study explores co-design as a means to invite community members, together with a designer, to explore the identity that they would like to present to people outside the community. In preparing for an exhibition to challenge perceptions, it turned out to be important to embrace the culture of the community to facilitate self-expression, introducing new concepts such as technological interventions and design probes to stimulate reflection and creativity. However, it was indigenous material culture, when actively and encouragingly supported by the designer, that had a key role in developing the co-design and, with it, empathic understanding between designer and community. 相似文献

A hospital emergency department (ED) is a complex cognitive work system. ED providers routinely create, process and share various kinds of information in their work. They may constantly transform information using technological artifacts such as an electronic patient information system. The functionality in the technology, however, limits their tasks and activities. So, they create their own artifacts (such as handwritten notes on a post-it note), to share and process information. Taking advantage of the unique characteristics of an ambipolar carbon nanotube field effect transistor (CNTFET), a ‘p-n junction’ is simulated along the single-walled carbon nanotube channel using two separate gates close to the source and drain of the CNTFET, respectively. The current-voltage characteristics of the double-gated CNTFET are calculated using a semiclassical method based on the Schottky barrier field effect transistor mechanism. n−3 fatty acids (fish oil; FO), but were similar in their content of saturated and monounsaturated fatty acids and vitamin
E. At the end of the four-week feeding period, each dietary group was subdivided into two groups. One group received a single
placebo injection of α-tocopherol-stripped corn oil (TSCO); the other group received a single injection of the free radical
generator, methyl ethyl ketone peroxide (MEKP), in TSCO. Twenty-four hours after injection, the effect of dietary oil and
MEKP treatment on endogenous lipid peroxide (LPO) production (measured as methylene blue formed by the “Determiner LPO” assay),
glutathione (GSH) and vitamin E content, and fatty acid composition of phosphatidylcholine and phosphatidylethanolamine in
heart and liver from unfasted animals were measured. FO-fed rats had significantly heavier hearts and livers, increased levels
of n−3 fatty acids in membrane phospholipids, and higher liver LPO levels than CO-fed rats. MEKP treatment resulted in significantly
lower body weights and liver GSH levels. The data indicate that dietary n−3 fatty acids increase lipid peroxidation in liver
somewhat more than in heart. The study also demonstrates that the effect of induced oxidative stress due to a single dose
of MEKP on lipid peroxide formation and antioxidant status in tissues from unfasted animals was independent of the dietary

To examine the effect of fish oil supplementation on the fatty acid (FA) composition of human milk and maternal and infant
erythrocytes, five lactating women were supplemented with 6 g of fish oil daily for 21d. Usual maternal diets contained 1,147
mg of total n−3 FA, with 120 mg from very long-chain (>C18) n−3 FA. Supplementation increased dietary levels to 3,092 mg of total n−3 FA and 2,006 mg of very long-chain n−3 FA. Milk
samples were collected daily, prior to fish oil ingestion, and at 4-h intervals on days 1, 7, 14 and 21. Milk n−3 FA content
increased within 8 h and reached steady state levels within one week. The n−6 fatty acid content decreased. Erythrocyte eicosapentaenoic
acid content increased from 0.24% to 1.4% (P<0.01) in mothers and from 0.11% to 0.70% (P<0.05) in infants. Docosapentaenoic acid increased from 1.4% to 2.2% (P<0.05) in mothers and from 0.30% to 0.78% (P<0.01) in infants. There was no significant change in docosahexaenoic acid or n−6 fatty acid content. Maternal platelet aggregation
responses were variable. No differences in milk or plasma tocopherol levels were noted.

Coupled gas chromatography with electroantennographic detection (GC-EAD) using antennae of adult female Manduca sexta was employed to screen for olfactory stimulants present in headspace collections from four species of larval host plants belonging to two families: Solanaceae—Lycopersicon esculentum (tomato), Capiscum annuum (bell pepper), and Datura wrightii; and Martyniaceae—Proboscidea parviflora. Headspace volatiles were collected from undamaged foliage of potted, living plants. GC–EAD revealed 23 EAD-active compounds, of which 15 were identified by GC-mass spectrometry. Identified compounds included aliphatic, aromatic, and terpenoid compounds bearing a range of functional groups. Nine EAD-active compounds were common to all four host plant species: (Z)-3-hexenyl acetate, nonanal, decanal, phenylacetaldehyde, methyl salicylate, benzyl alcohol, geranyl acetone, (E)-nerolidol, and one unidentified compound. Behavioral responses of female moths to an eight-component synthetic blend of selected tomato headspace volatiles were tested in a laboratory wind tunnel. Females were attracted to the blend. A comparison of responses from antennae of males and females to bell pepper headspace volatiles revealed that males responded to the same suite of volatiles as females, except for (Z)-3-hexenyl benzoate. EAD responses of males also were lower for (Z)- and (E)-nerolidol and one unidentified compound. Electroantennogram EAG dose–response curves for the 15 identified EAD-active volatiles were recorded. At the higher test doses (10–100 g), female antennae yielded larger EAG responses to terpenoids and to aliphatic and aromatic esters. Male antennae did respond to the higher doses of (Z)-3-hexenyl benzoate, indicating that they can detect this compound. On the basis of ubiquity of the EAD-active volatiles identified to date in host plant headspace collections, we suggest that M. sexta uses a suite of volatiles to locate and identify appropriate host plants. 相似文献
