The tear film is a multi-layer fluid that covers the corneal and conjunctival epithelia of the eye and provides lubrication, nutrients, and protection from the outside environment. Tear fluid contains a high concentration of proteins and has thus been recognized as a potential source of biomarkers for ocular disorders due to its proximity to disease sites on the ocular surface and the non-invasive nature of its collection. This is particularly true in the case of dry eye disease, which directly impacts the tear film and its components. Proteomic analysis of tear fluid is challenging mainly due to the wide dynamic range of proteins and the small sample volumes. However, recent advancements in mass spectrometry have revolutionized the field of proteomics enabling unprecedented depth, speed, and accuracy, even with small sample volumes. In this study using the Orbitrap Fusion Tribrid mass spectrometer, we compared four different mass spectrometry workflows for the proteomic analysis of tear fluid collected via Schirmer strips. We were able to establish a method of in-strip protein digestion that identified >3000 proteins in human tear samples from 11 healthy subjects. Our method offers a significant improvement in the number of proteins identified compared to previously reported methods without pooling samples.  相似文献

Despite data that traditional laboratory-based outcome measures in dialysis are improving over time, population-based data indicate that mortality rates are not improving in parallel. With increased focus on performance measures based on laboratory-based outcomes (e.g., hematocrit, albumin, and parathyroid hormone), less emphasis has been placed on other markers, some of which may be stronger predictors of mortality. We performed a systematic review to interpret the predictive value of laboratory-based outcome measures in dialysis. We identified studies with data regarding the predictive value of laboratory-based outcomes for mortality in dialysis. We calculated the sample size-weighted pooled relative risk of death with dichotomized "high" vs. "low" levels of each measure. We rank-ordered predictors by scaling the pooled relative risk of each measure by its pooled standard deviation. There were 5171 titles, of which 128 (representing 44 laboratory-based outcomes) were selected. Nine were significantly associated with mortality, in order of decreasing scaled effect size: (1) tumor necrosis factor-α, (2) hematocrit, (3) interleukin-6, (4) troponin T, (5) Kt/Vurea, (6) prealbumin, (7) urea reduction ratio, (8) serum albumin, and (9) C-reactive protein. Other oft-cited measures such as calcium phosphate product and parathyroid hormone were not significantly associated with mortality in pooled analysis. Quality improvement efforts to improve traditional laboratory-based outcomes in end-stage renal disease are necessary, but likely insufficient, to improve overall mortality in dialysis. Renewed consideration of cardiovascular, inflammatory, and nutritional markers that are especially strong predictors of mortality may have important implications for risk stratification and targeted therapeutic interventions.  相似文献

Understanding what factors influence positive youth development has been advocated by youth development researchers (P. L. Benson, 2006; J. S. Eccles & J. A. Gootman, 2002). Consequently, the purpose of this study was to examine whether perceptions of a caring youth sport context influenced prosocial and antisocial behavior through efficacy-related beliefs, that is, positive and negative affective self-regulatory efficacy (ASRE) and empathic self-efficacy (ESE). Multiethnic youths taking part in summer sport programs (N = 395) completed a questionnaire that measured perceptions of the caring climate, ESE, ASRE, and social behavior. Structural equation modeling was used to test whether ASRE and ESE mediated the relationship between caring and social behaviors. Findings revealed that perceptions of caring positively predicted ASRE and ESE. In turn, positive ASRE positively predicted ESE. Prosocial behaviors were positively linked to ESE, whereas antisocial behaviors were negatively predicted by positive ASRE. The results suggest that caring influences prosocial and antisocial behavior because such contexts develop youths' ability to monitor, manage, and control positive affect, which in turn enhances their belief in their ability to empathize. Internet based data collection methods have many advantages for psychological assessment when compared with more traditional paper formats, including, reduced costs, and greater convenience for both the researcher and the participant. However, prior to the wide-spread adoption of these methods, equivalence with paper tests formats must be established. The present study compared questionnaire data from the computer with that from a traditional paper format in a sample of college students both directly and through an opinion survey. Three types of questionnaires that represent distinct areas commonly assessed in psychological research included: quality of life (SF-36), depression (BDI-II) and personality (NEO). A within-subjects design counterbalanced order across the computer and paper formats. Overall the findings showed no differences for the Neuroticism and Extraversion facts of the NEO, for the BDI and for the subscales and composite scores of the SF-36. Significant differences were found between the paper and computer formats for the Openness, Agreeableness, and Conscientiousness facets of the NEO. Additionally, the computer was perceived to be convenient, user-friendly, comfortable, and secure. Results suggest that a computerized format is an efficient way to conduct quality of life research, especially for the assessment of distress levels and quality of life.  相似文献

Real-time, rich-media data communication between the construction site and the off-site design office is becoming one of the important research areas in information technology for construction. This paper presents the concept of a telematic digital workbench, a horizontal tabletop user interface integrating mobile computing and wireless communication to facilitate synchronous construction site to office collaboration. We demonstrate the capabilities and potentials of this system concept for construction defect management. The on-site crew uses a handheld mobile device to collect defect information and transfers the information to the design office through wireless communication by sending the information to a database listener. The digital workbench application monitors the database and synchronizes the location of the visual information on the site with the 3D model on the server. Integrated with 3D viewing capability in a CAD system, designers can interact with the combined model/site data using a horizontal and vertical screen. A case study compared the telematic digital workbench against paper-based and Pocket PC-based methods for defect management in a controlled laboratory experiment. The case study results show that the telematic digital workbench has the potential to improve the accuracy of matching site data to digital data and reduce information loss between site and office collaboration.  相似文献

A growing awareness of the potential impacts of climate change has led to the consideration of a variety of future outcomes by water supply companies, regulators and local authorities within England and Wales. These strategies recognise the possibility of episodes of flooding within catchments, with the Environment Agency's fourth Periodic Review requiring water companies to explore future demand and supply scenarios up to 2030. However, there is a gap in the literature concerning how those tasked to implement water policy, and those subject to it, view these change scenarios. This paper outlines a study undertaken within the River Nene catchment, exploring how water resource governance bodies and water users respond to flooding issues. In particular, the research focuses on the twin concepts of legitimacy and dialogue as a means to assess the boundaries of stakeholder engagement with these strategic change scenarios.  相似文献
