The linear sampling method (LSM) offers a qualitative image reconstruction approach, which is known as a viable alternative for obstacle support identification to the well-studied filtered backprojection (FBP), which depends on a linearized forward scattering model. Of practical interest is the imaging of obstacles from sparse aperture far-field data under a fixed single frequency mode of operation. Under this scenario, the Tikhonov regularization typically applied to LSM produces poor images that fail to capture the obstacle boundary. In this paper, we employ an alternative regularization strategy based on constraining the sparsity of the solution's spatial gradient. Two regularization approaches based on the spatial gradient are developed. A numerical comparison to the FBP demonstrates that the new method's ability to account for aspect-dependent scattering permits more accurate reconstruction of concave obstacles, whereas a comparison to Tikhonov-regularized LSM demonstrates that the proposed approach significantly improves obstacle recovery with sparse-aperture data.  相似文献

In this study, poly(dl ‐lactide‐co‐glycolide)/porous silicon (PLGA/pSi) composite microspheres, synthesized by a solid‐in‐oil‐in‐water (S/O/W) emulsion method, are developed for the long‐term controlled delivery of biomolecules for orthopedic tissue engineering applications. Confocal and fluorescent microscopy, together with material analysis, show that each composite microsphere contained multiple pSi particles embedded within the PLGA matrix. The release profiles of fluorescein isothiocyanate (FITC)‐labeled bovine serum albumin (FITC‐BSA), loaded inside the pSi within the PLGA matrix, indicate that both PLGA and pSi contribute to the control of the release rate of the payload. Protein stability studies show that PLGA/pSi composite can protect BSA from degradation during the long term release. We find that during the degradation of the composite material, the presence of the pSi particles neutralizes the acidic pH due to the PLGA degradation by‐products, thus minimizing the risk of inducing inflammatory responses in the exposed cells while stimulating the mineralization in osteogenic growth media. Confocal studies show that the cellular uptake of the composite microspheres is avoided, while the fluorescent payload is detectable intracellularly after 7 days of co‐incubation. In conclusion, the PLGA/pSi composite microspheres offer an additional level of controlled release and could be ideal candidates as drug delivery vehicles for orthopedic tissue engineering applications.  相似文献

The relative importance of the Chicago/Gay urban area was investigated to determine its impact on atmospheric mercury (Hg) concentrations and wet deposition in the Lake Michigan basin. Event wet-only precipitation, total particulate, and vapor phase samples were collected for Hg, and trace element determinations from five sites around Lake Michigan from July 1994 through October 1995 as part of the Lake Michigan Mass Balance Study (LMMBS). In addition, intensive over-water measurements were conducted aboard the EPA research vessel Lake Guardian during the summer of 1994 and the winter of 1995 as part of the Atmospheric Exchange Over Lakes and Oceans Study. Atmospheric Hg concentrations were found to be significantly higher in the Chicago/Gary urban area than surrounding sites: Hg in precipitation was a factor of 2 and particulate Hg was a factor of 6 times higher. Overwater measurements found elevated Hg concentrations 19 km off shore of Chicago/Gary suggesting an enhanced near field atmospheric deposition to Lake Michigan. Meteorological transport analyses also determined that local sources in the Chicago/Gary urban area significantly impacted all of the LMMBS sites indicating a broad impact to the entire Lake Michigan basin.  相似文献

We used scanner data on cigarette prices and sales collected from supermarkets across the United States from 1994 to 2004 to test the hypothesis that cigarette prices are positively correlated with sales of cigarettes with higher tar and nicotine content. During this period the average inflation-adjusted price for menthol cigarettes increased 55.8%. Price elasticities from multivariate regression models suggest that this price increase led to an increase of 1.73% in sales-weighted average tar yields and a 1.28% increase in sales-weighted average nicotine yields for menthol cigarettes. The 50.5% price increase of nonmenthol varieties over the same period yielded an estimated increase of 1% in tar per cigarette but no statistically significant increase in nicotine yields. An ordered probit model of the impact of cigarette prices on cigarette strength (ultra-light, light, full flavor, unfiltered) offers an explanation: As cigarette prices increase, the probability that stronger cigarette types will be sold increases. This effect is larger for menthol than for nonmenthol cigarettes. Our results are consistent with earlier population-based cross-sectional and longitudinal studies showing that higher cigarette prices and taxes are associated with increasing consumption of higher-yield cigarettes by smokers.  相似文献

The application of infrared microspectroscopy (IRMS) technology, combined with multivariate analysis, was evaluated to develop sensitive and robust methods to assess cleanability of stainless steel surfaces for the removal of dairy food residues. UHT milk samples (skim, 1%, 2%, and whole) were analyzed for total nitrogen (Kjeldahl) and fat (Babcock) contents. The coupons were manually soiled with serially diluted milk samples resulting in soils ranging from 0.1 to 428.1 μg/cm(2) for protein and 0.1 to 374.17 μg/cm(2) for fat, and then autoclaved to simulate a heated equipment surface. Reflectance spectra were collected from stainless steel coupons by using IRMS, and multivariate analysis was used to develop calibration models based on cross-validated partial least squares regression (PLSR). Statistical analysis for the prediction of protein and fat showed a standard error of cross-validation (SECV) of 0.5 and 0.4 μg/cm(2) for prediction of protein and fat, respectively, and correlation coefficients (rVal) > 0.99. To improve the sensitivity, swabbing and concentration steps were used prior to IRMS analysis obtaining SECV of 0.04 and 0.01 μg/cm(2) for the prediction of protein and fat, respectively, and rVal > 0.99. The PLSR models accurately predicted the levels of protein and fat on autoclaved stainless steel coupons soiled with milk. A simple, reliable, and robust protocol based on IRMS and multivariate analysis was developed for multicomponent characterization of stainless steel surfaces that can contribute to more efficient cleaning verification with regard to contamination on surfaces of processing equipment. PRACTICAL APPLICATION: We report the application of Fourier transform infrared microspectroscopy (FTIR) for the validation of CIP cleaning efficiency that would provide a basis for better understanding of the mechanisms involved in the removal of physical soil and food residues from different types of equipment surfaces commonly utilized in the biotech, pharmaceutical, and food industries. Reliable calibration models were generated that showed the ability to predict the amounts of dairy soils on the surface of stainless steel coupons. Including a swabbing step of the coupons before infrared spectral acquisition provided improved sensitivity and reproducibility for multicomponent cleaning verification. Results from this research project would allow designing experiments to rapidly evaluate different materials and finishes, the effects of process variables, the influence of food components, and the development of reliable and robust cleaning validation protocols to ensure the safety and quality of the product.  相似文献

The hides of cattle are the source of Escherichia coli O157:H7 that contaminates beef carcasses during commercial beef processing. Therefore, effective interventions that reduce hide contamination should reduce subsequent carcass contamination. The first objective of this study was to identify the most effective reagents for decontamination of beef hides. Cattle hides draped over barrels were used for in vitro experiments to compare the efficacy of washes using 1.6% sodium hydroxide, 4% trisodium phosphate, 4% chlorofoam, or 4% phosphoric acid, each followed by a rinse step using either water or acidified (pH 7.0) chlorine at 200 or 500 ppm. All treatments using a water rinse reduced hide coliform counts by 1.5 to 2.5 log CFU/ 100 cm2. Compared with water rinses, 200 and 500 ppm acidified chlorine rinses increased efficacy by approximately 1.0 and 2.0 log CFU/100 cm2, respectively. Vacuuming of the treated areas to remove excess liquid improved hide cleanliness by an average of an additional 1.0 log CFU/100 cm2. The second objective was to evaluate the use of an on-line hide-wash cabinet that used a sodium hydroxide wash and a chlorinated (1 ppm) water rinse. Hides sampled before entering and after exiting the cabinet had aerobic plate counts and Enterobacteriaceae counts that were reduced by 2.1 and 3.4 log CFU/100 cm2, respectively, and the prevalence of E. coli O157 on hides was reduced from 44 to 17% when the cabinet was in use. Preevisceration carcass aerobic plate counts and Enterobacteriaceae counts were both reduced by 0.8 log CFU/100 cm2, and the prevalence of E. coli O157 on preevisceration carcasses was reduced from 17 to 2% when the cabinet was in use. These results support decontamination of hides as an effective means to reduce pathogen contamination of cattle carcasses during processing.  相似文献

The 12 Principles of Green Engineering have been proposed as a framework within which to examine existing products and guide their redesign as well as to evaluate new product designs. The EcoWorx system represents a recyclable carpet tile product that is assessed using the 12 Principles of Green Engineering and cradle-to-cradle design principles to evaluate environmental, qualitative, and economic performance as compared to existing Shaw carpet tile products. The product design strategy embodies life cycle considerations of a cradle-to-cradle product that puts technical nutrient recovered materials into repeated use. EcoWorx offers a unique opportunity for Shaw to utilize the 12 Principles to analyze and optimize new products and processes for the additional capacity needed to meet rising demand for sustainable carpet products. The analysis of the EcoWorx system demonstrated the value of the 12 Principles in verifying and formalizing the experience and intuition of product designers. This suggests that the mistrial and error reliance on experience could be enhanced with this formalized approach and hence lead to more widespread success of design practices that result in sustainable products.  相似文献
