The spatio-temporal patterns of neural activity evoked by electrical stimuli to the antennal nerve (AN) in male cockroach antennal lobes (ALs) in vivo were analyzed by optical imaging using a voltage-sensitive dye. The response pattern was initially a depolarization on the AN and subsequently a depolarization followed by a hyperpolarization on the whole area of macroglomerulus (MG) and a part of ordinary glomerulus (OG). It was suggested by the pharmacological results that the depolarizing responses on the AL consist of both a presynaptic response, representing synchronous compound action potentials from the AN, and a postsynaptic response, representing synchronous compound excitatory postsynaptic potentials and action potentials from neurites of AL neurons, and that the inhibitory responses of GABAergic local interneurons in the AN are different in time course from that in the AL. 相似文献

Two new prototype dipole magnets for the proposed new fast cycling (f=1 Hz) synchrotron at GSI in Darmstadt have been designed, fabricated and tested. The magnets are based on a window frame iron yoke cooled by liquid nitrogen and a superconducting winding made from a hollow NbTi composite superconductor cable cooled with forced two-phase helium flow at T=4.5 K. The cold mass of the magnet is separated from the yoke by a small vacuum gap of 0.75 mm to 2.5 mm. A decrease of ac power losses by a factor of 2.3 in comparison with a standard Nuclotron dipole is obtained. The distributions of aquaporin-1 mRNA and protein were studied by hybridization histochemistry with a homologous riboprobe and immunohistochemistry, in the adult sheep kidney. Heaviest labelling occurred in the thin descending limb (DTL) of the loop of Henle in the inner stripe of the outer medulla, with apparent decreasing expression in the inner medulla, outer stripe of the outer medulla and cortex, but no quantitation was performed. Only proximal tubules (PT) (convoluted and straight) and DTL labelled. The glomerulus showed no labelling, consistent with the pattern in the rat but different to that in the human. During ontogeny, no labelling occurred in the mesonephros at 27 or 41 days of gestation (term = 145-150 days) but other structures did label at 27 days (heart, lung bud, blood vessels surrounding developing spinal cord). Labelling first occurred faintly in the metanephros at 41 days of gestation and increased throughout gestation consistent with morphological development of nephrons. 相似文献

Two-dimensional gel electrophoresis databases have been generated for a range of tissue cell and fluid types, from a number of species. A major difficulty in the development of such databases is the large number of proteins present in even a single cell type (> 10,000) and the low levels of many of these proteins. One approach to overcome these difficulties is to fractionate the cell into its organelles and generate individual databases for each subcellular component. BACKGROUND: This study was conducted to investigate the impact of the preservation method of bioprosthetic heart valve materials on calcification rates and biocompatibility of the biologic tissue. METHODS: In subcutaneous rat implants, conventionally preserved bioprosthetic heart valve material was compared with bovine pericardium that was treated with L-glutamic acid to reduce residual glutaraldehyde released from the fixed tissue. Both these methods were compared with bovine pericardium that was stabilized by a dye-mediated photooxidation reaction without glutaraldehyde. Biocompatibility of these biomaterials was tested in vitro using human endothelial cell cultures. RESULTS: Conventionally preserved bovine pericardium with a high amount of glutaraldehyde incorporated into the tissue resulted in severe calcification 63 days after subcutaneous implantation in rats (165.4 +/- 20 mg Ca2+/g dry weight). Postfixation treatment with L-glutamic acid, which reduces free, unbound aldehyde groups, showed a significant decrease in calcification (89.6 +/- 14 mg Ca2+/g dry weight). Glutaraldehyde-free preservation by dye-mediated photooxidation showed no calcification after 63 days of subcutaneous implantation (1.0 +/- 0.4 mg Ca2+/g dry weight). Regular endothelial cell proliferation was observed on photooxidized and L-glutamic acid-treated tissue, whereas conventionally treated tissue caused endothelial cell death. CONCLUSIONS: This study underlines the detrimental role of glutaraldehyde in the calcification process of bioprosthetic heart valve materials and emphasizes alternative preservation methods that reduce or avoid the use of glutaraldehyde. 相似文献

Levels of analysis issues have attracted a lot of attention in group psychology research. Despite assertions pertaining to the value of multilevel models, most researchers focus on either the individual within groups or the group as a whole, but seldom on both. A multilevel approach may be helpful to group psychologists. This article addresses levels of analysis issues that are an inherent part of group research, and a number of methods that can be used to analyze multilevel data are presented. The methods fall into 3 categories: (a) assessing the extent of agreement within a single group, (b) contrasting within-group and between-groups variance, and (c) conducting multiple-level analyses.
