There are reports that nano-sized zero-valent iron (Fe0) exhibits greater reactivity than micro-sized particles of Fe0, and it has been suggested that the higher reactivity of nano-Fe0 may impart advantages for groundwater remediation or other environmental applications. However, most of these reports are preliminary in that they leave a hostof potentiallysignificant(and often challenging) material or process variables either uncontrolled or unresolved. In an effort to better understand the reactivity of nano-Fe0, we have used a variety of complementary techniques to characterize two widely studied nano-Fe0 preparations: one synthesized by reduction of goethite with heat and H2 (Fe(H2)) and the other by reductive precipitation with borohydride (Fe(BH)). Fe(H2) is a two-phase material consisting of 40 nm alpha-Fe0 (made up of crystals approximately the size of the particles) and Fe3O4 particles of similar size or larger containing reduced sulfur; whereas Fe(BH) is mostly 20-80 nm metallic Fe particles (aggregates of <1.5 nm grains) with an oxide shell/coating that is high in oxidized boron. The FeBH particles further aggregate into chains. Both materials exhibit corrosion potentials that are more negative than nano-sized Fe2O3, Fe3O4, micro-sized Fe0, or a solid Fe0 disk, which is consistent with their rapid reduction of oxygen, benzoquinone, and carbon tetrachloride. Benzoquinone-which presumably probes inner-sphere surface reactions-reacts more rapidly with FeBH than Fe(H2), whereas carbon tetrachloride reacts at similar rates with FeBH and Fe(H2), presumably by outer-sphere electron transfer. Both types of nano-Fe0 react more rapidlythan micro-sized Fe0 based on mass-normalized rate constants, but surface area-normalized rate constants do not show a significant nano-size effect. The distribution of products from reduction of carbon tetrachloride is more favorable with Fe(H2), which produces less chloroform than reaction with Fe(BH).  相似文献

Molecular methods have become vital epidemiological tools in the detection and characterization of bacteria associated with a foodborne outbreak. We used both culture and real-time PCR to detect a Vibrio parahaemolyticus isolate associated with a foodborne outbreak. The outbreak occurred in July 2002 in Polk County, Florida, and originated at a Chinese buffet, with one person being hospitalized. The hospital isolated V. parahaemolyticus from the patient but destroyed the sample after diagnosis. From an onsite visit of the restaurant, food samples that possibly contributed to the outbreak were collected and sent to the Florida Department of Health, Tampa Branch Laboratory. Crab legs, crabsticks, and mussel samples were homogenized and incubated according to the Food and Drug Administration Bacteriological Analytical Manual culture protocol. Three sets of primers and a TaqMan probe were designed to target the tdh, trh, and tlh genes and used for real-time PCR. This study was successful in isolating V. parahaemolyticus from a mussel sample and detecting two of its genes (tdh and tlh) in food and pure culture by real-time PCR.  相似文献

The physical, dielectric, and optical properties of hot isostatically pressed lead magnesium niobate polycrystalline ceramics modified with 1/2 mol% La2O3, Pb1–3/2 x La x x /2-(Mg1/3Nb2/3)O3, have been investigated. Methods used to characterize the ceramics included determination of the dielectric permittivity, optical transmittance, and refractive index dispersion. The materials exhibited relaxor ferroelectric type behavior with a peak dielectric constant K > 14000 and average T c ∼−35°C. Various sintering, hot isostatic pressing, and annealing conditions were examined to produce highly dense and optically transparent materials. Through the use of hot isostatic pressing, densities more than 99.5% theoretical and transmittance greater than 50% at 633-nm wavelength were obtained. Hot isostatic pressing technique appears to be a good alternative to hot uniaxial pressing without the associated problem of PbO volatility, reactivity with the pressure vessel, and geometrical constraints.  相似文献

In this paper, the inversion processing of cascade impactor data to construe continuous size distributions within fine particulate matter (PM2.5) is examined for residential oil furnace and fireplace appliance emissions. Impactor data from tests with these emissions sources are selected for the challenges they pose to comprehending the size distributions of aerosol mass and chemical species. In specific, the oil furnace aerosol offers an opportunity to apply data inversion to study a bimodal lognormal distribution in which much of the aerosol mass is impactor-penetrating nanoparticles . The fireplace emissions on the other hand cover the issue of a chemical size distribution, which is subject to particle loss and characterized by a single lognormal, accumulation mode peak. Computational steps relevant to the application of the data inversion are illustrated in detail. Evaluation of correlation coefficients (0.992) indicates that the inversion model predictions fit the impactor data well. Simulations of systematic measurement error (±10%) at each impactor stage are shown to have a negligible impact on the inversion results for test data. It is concluded that data inversion can be effective when (i) source emissions contain a portion of particles that falls outside the measurement range of cascade impactors, (ii) a mass size distribution of an individual species is determined without the knowledge of the total mass concentration for that species, or when (iii) losses in the particle charger system are significant.  相似文献

查阅迄今为止中国现有的实测地应力资料,参照霍克–布朗的地应力分析、统计和拟合方法,形成实测平均水平地应力与垂直地应力的比值随深度变化的散点图,回归实测平均水平地应力与垂直地应力的比值随深度的变化曲线并与霍克–布朗曲线进行比较,发现深度超过1 200 m以后,中国实测平均水平地应力与垂直地应力的比值统计值大于霍克–布朗曲线中值,且最大应力包线与霍克–布朗包线基本吻合,而最小应力包线的应力水平小于霍克–布朗包线的应力水平。进一步分析回归中国岩浆岩、沉积岩和变质岩等3种岩性的地应力统计结果,得到类似的回归曲线。统计结果对了解中国地应力场的宏观分布情况具有重要参考价值,对进行深埋地下结构分析计算和辅助设计具有重要的实用价值。  相似文献

Malignant melanoma is a lethal skin cancer containing melanoma-initiating cells (MIC) implicated in tumorigenesis, invasion, and drug resistance, and is characterized by the elevated expression of stem cell markers, including CD133. The siRNA knockdown of CD133 enhances apoptosis induced by the MEK inhibitor trametinib in melanoma cells. This study investigates the underlying mechanisms of CD133’s anti-apoptotic activity in patient-derived BAKP and POT cells, harboring difficult-to-treat NRASQ61K and NRASQ61R drivers, after CRISPR-Cas9 CD133 knockout or Dox-inducible expression of CD133. MACS-sorted CD133(+) BAKP cells were conditionally reprogrammed to derive BAKR cells with sustained CD133 expression and MIC features. Compared to BAKP, CD133(+) BAKR exhibit increased cell survival and reduced apoptosis in response to trametinib or the chemotherapeutic dacarbazine (DTIC). CRISPR-Cas9-mediated CD133 knockout in BAKR cells (BAKR-KO) re-sensitized cells to trametinib. CD133 knockout in BAKP and POT cells increased trametinib-induced apoptosis by reducing anti-apoptotic BCL-xL, p-AKT, and p-BAD and increasing pro-apoptotic BAX. Conversely, Dox-induced CD133 expression diminished apoptosis in both trametinib-treated cell lines, coincident with elevated p-AKT, p-BAD, BCL-2, and BCL-xL and decreased activation of BAX and caspases-3 and -9. AKT1/2 siRNA knockdown or inhibition of BCL-2 family members with navitoclax (ABT-263) in BAKP-KO cells further enhanced caspase-mediated apoptotic PARP cleavage. CD133 may therefore activate a survival pathway where (1) increased AKT phosphorylation and activation induces (2) BAD phosphorylation and inactivation, (3) decreases BAX activation, and (4) reduces caspases-3 and -9 activity and caspase-mediated PARP cleavage, leading to apoptosis suppression and drug resistance in melanoma. Targeting nodes of the CD133, AKT, or BCL-2 survival pathways with trametinib highlights the potential for combination therapies for NRAS-mutant melanoma stem cells for the development of more effective treatments for patients with high-risk melanoma.  相似文献

This work focusses on a numerical and experimental analysis of unsteady forced convection in hydrodynamically developed and thermally developing laminar air flow in a circular duct, subjected to a periodic variation of the inlet temperature. The experiments were conducted over a wide range of Reynolds number (281.2 ≤ Re ≤ 1024.3) and inlet frequency (0.01 ≤ β ≤ 0.20 Hz) of the periodic heat input. In the numerical study, the non-uniform inlet temperature amplitude profile derived from the experiments, was included in the numerical model. A fully explicit, second-order accurate finite difference scheme was developed and used for the solution of the unsteady energy equation. Numerical results are obtained with the fully developed parabolic velocity profile under the boundary condition of the first kind, which was verified by the experiments. Temperature variations along the centerline of the circular duct are observed to be thermal oscillations with the same frequency as the inlet periodic heat input and amplitudes that decayed exponentially with distance along the duct. Thermal response along the wall exhibits negligible amplitude variation with changes in Reynolds number and inlet frequency. The variation in the periods and amplitudes of the thermal oscillations are observed to be a function of spacial system variables only. Satisfactory agreement between the numerical and experimental results are obtained.  相似文献

Food fraud is an intentional act for economic gain. It poses a risk to food integrity, the economy, public health and consumers’ ethics. Seafood is one commodity which has endured extensive fraudulent activity owing to its increasing consumer demand, resource limitations, high value and complex supply chains. It is essential that these fraudulent opportunities are revealed, the risk is evaluated and countermeasures for mitigation are assigned. This can be achieved through mapping of the seafood supply chains and identifying the vulnerability analysis critical control points (VACCP), which can be exposed, infiltrated and exploited for fraudulent activity. This research systematically maps the seafood supply chain for three key commodities: finfish, shellfish and crustaceans in the United Kingdom. Each chain is comprised of multiple stakeholders across numerous countries producing a diverse range of products distributed globally. For each supply chain the prospect of fraud, with reference to species substitution, fishery substitution, illegal, unreported and unregulated substitution, species adulteration, chain of custody abuse, catch method fraud, undeclared product extension, modern day slavery and animal welfare, has been identified and evaluated. This mapping of the fraudulent opportunities within the supply chains provides a foundation to rank known and emerging risks and to develop a proactive mitigation plan which assigns control measures and responsibility where vulnerabilities exist. Further intelligence gathering and management of VACCPs of the seafood supply chains may deter currently unknown or unexposed fraudulent opportunities.  相似文献

采用木聚糖酶水解麦麸制备阿魏酸低聚糖,以阿魏酸低聚糖的量为指标,研究了酶解温度、pH值、酶浓度、底物浓度、作用时间五个因素对酶解效果的影响.并在此基础上,通过中心组合试验确定最佳酶解条件.最终的优化条件是木聚糖酶浓度3%,底物浓度80 g/L,作用时间12 h,此条件下阿魏酸低聚糖提取量为0.831×10-5 moL/g麦麸.  相似文献
