A colinearly synthesized peptide consisting of a H-2d restricted T-helper cell epitope of Semliki Forest virus (SFV) and triple repeats of sequence GPGRAF, derived from the V3 domain of HIV-1 strains, was used to immunize BALB/c (H-2d) mice. Pepscan analysis of sera from peptide-immunized mice revealed that the chimaeric peptide GREKFTIRPHYGKEIGPGRAFGPGRAFGPGRAF contains three distinct antibody-reactive sequences GREKFTIR, PHYGKEI and GPGRAF. The chimaeric peptide evoked HIV-1 IIIb neutralizing antibodies in serum as measured in vitro by reduction of syncytia formation and reduction of p24 production as well. So, the T-helper cell epitope of SFV provided help to a small linear neutralization epitope of HIV-1 strains. Interestingly, the T-helper cell epitope alone might induce antibodies cross-reactive with HIV-1 IIIb specific peptide GPGRAFVTIGK which shows some homology (residues underlined) with the antibody-reactive sequence GREKTIR of SFV. 相似文献

Strains of Vibrio parahaemolyticus were isolated from clinical, marine and freshwater fish of Calcutta, West Bengal, India. Drug and metal resistance characteristics were compared for differentiation of clinical and environmental strains. Eighteen out of the twenty environmental isolates were resistant to arsenate, unlike the clinical isolates which were all susceptible. Stem cell factor (SCF) is produced by stromal cells as a membrane-bound molecule, which may be proteolytically cleaved at a site close to the membrane to produce a soluble bioactive form. The proteases producing this cleavage are unknown. In this study, we demonstrate that human mast cell chymase, a chymotrypsin-like protease, cleaves SCF at a novel site. Cleavage is at the peptide bond between Phe-158 and Met-159, which are encoded by exon 6 of the SCF gene. This cleavage results in a soluble bioactive product that is 7 amino acids shorter at the C terminus than previously identified soluble SCF. This research shows the identification of a physiologically relevant enzyme that specifically cleaves SCF. Because mast cells express the KIT protein, the receptor for SCF, and respond to SCF by proliferation and degranulation, this observation identifies a possible feedback loop in which chymase released from mast cell secretory granules may solubilize SCF bound to the membrane of surrounding stromal cells. The liberated soluble SCF may in turn stimulate mast cell proliferation and differentiated functions; this loop could contribute to abnormal accumulations of mast cells in the skin and hyperpigmentation at sites of chronic cutaneous inflammation. 相似文献

With shortage of research funds and increasing competition for medical posts, performance indicators and control instruments are being looked for in order to be able to allot research funds and make professorial appointments in relation to scientific performance. Incomprehensibly for many, the impact factor has become the decisive scientometric indicator at German universities despite of substantial systematic limitations. The impact factor is derived from the journal citation reports. Its basis of calculation entails the following problems: the editorial board of the private Institute of Scientific Information (ISI) decides on whether a journal is to be classified as a source journal. The citation index of all journals is calculated from their citations alone. Crucial means of influencing the impact factor result from self-citations and citation groups in these source journals. Languages other than English and other than Latin alphabets are appreciably disadvantaged by the citation index, which is why for example despite its international significance the rapid development of the osteosynthesis technique in German speaking countries went unnoticed by British and American orthopedic surgeons and scientists. The articles on postgraduate training necessarily published by clinicians in the respective language of their country are not cited because the addresses of such publications do not engage in research. Clinical disciplines (especially highly specialized disciplines such as trauma and hand surgery) thus attain appreciably lower impact factors for their journals than basic disciplines and interdisciplinary clinical sectors which lead the ranking of journals. The period covered in calculating the impact factor is only 2 years. Very modern and widely disseminated organs of publication with a short information halflife are favored. From the 10 objectively most often cited and most important journals for the scientific society, only 2 are to be found amongst those with the highest impact factor. The impact front-runner from 1995 has a very low absolute number of citations. The impact factor provides limited statistical information on a journal in its special field. Using it for this purpose presupposes knowledge of rules, limitations and constraints. Its uncritical use as a general currency of science is fundamentally unscientific. In addition, this leads to the specialists in the field knowledge of the universities being disregarded in favor of a pseudo-objective parameter determined elsewhere. At all events, correction factors for the impact factor have to be applied in respect to the different disciplines. The faculties should reach agreement on relevant (also on German language) organs of publication. The impact factor is not suitable as an indicator of the research activity and the quality of a researcher or an institution. Besides careful human judgement and other classical methods of decision making, the Science Citation Index can contribute to the individual evaluation. 相似文献

Multiparous sows that had been tethered during lactation were put in two different housing conditions after weaning (Day 0); the sows were either tethered by neck chain, or individually housed in a pen of approximately 6 m2. After two months, ten tethered and eleven loose housed sows were used to assess stress and reproductive parameters. Stereotypic behaviour after the afternoon feeding was assessed from Day 18 onwards; at Day 53 stereotypic behaviour tended to occur during a higher percentage of time in the tethered sows (P = 0.11) and at Day 66, the differences were significant (tethered, 78 +/- 5 vs. loose-housed, 40 +/- 10% of time (mean +/- sem); P = 0.03). At Day 35 and 55, cortisol profiles after afternoon feeding were similar for the two groups of sows (P > 0.10). Around oestrus (approximately Day 64), the profiles of oestradiol-17 beta, luteinizing hormone and progesterone were measured and proved to be similar for both treatment groups (P > 0.10). The duration of oestrus was shorter in the tethered sows (42 +/- 4 vs. 63 +/- 2 h; P < 0.001) and, consequently, the timing of ovulation during oestrus (h after onset of oestrus) was advanced in the tethered sows (28 +/- 2 vs. 41 +/- 2; P < 0.001). The duration of ovulation did not differ (tethered, 2.9 +/- 0.5 vs. loose-housed, 2.1 +/- 0.2 h; P = 0.16). The sows were sacrificed at Day 5 after ovulation; ovulation rate, fertilization rate, embryo development and embryo diversity were similar for the two groups, as were adrenal weight and size of adrenal cortex. Duration of oestrus and the levels of stereotypies at Day 60 tended to be negatively related in the tethered sows (P = 0.07), but not in the loose-housed sows (P = 0.65). In conclusion, sows that had been tethered during pregnancy and lactation, and were housed loose or were tethered again at weaning within two months differed both in stereotypic behaviour and in duration of oestrus, without apparent effects on reproductive hormones. 相似文献

This is the first case of nonprimary collapsing focal segmental glomerulosclerosis (FSGS) associated with Loa loa filariasis. Loa loa micofilariae were detected on a blood smear after a patient presented with nephrotic syndrome (NS), microhematuria, and renal failure. The renal biopsy showed a collapsing glomerulopathy variant of FSGS. Microfilariae also were identified in renal microvasculature, including the afferent arterioles and the glomerular and peritubular capillaries. 相似文献
