The use of mass spectrometry to measure hydrogen exchange rates for individual proteins in complex mixtures is described. Incorporation of stable-isotope-labeled (SIL) amino acids into a protein of interest during overexpression in bacteria produced distinctive isotope patterns in mass spectra of peptic peptides from the labeled protein. The isotope pattern was used as a signature for peptides originating from the SIL protein. In addition, stable-isotope labeling simplified identification of the peptic peptides by providing partial amino acid composition information. Despite the complex isotope patterns associated with SIL peptides, hydrogen exchange rates could still be measured for peptides from SIL protein and were found to be the same as exchange rates for unlabeled protein. Hydrogen exchange in a single protein of interest was measured in a complex mixture of proteins, a bacterial cell lysate. This methodology, which includes easy recognition of peptic peptides from the protein(s) of interest during hydrogen exchange studies in heterogeneous systems, will permit analysis of structural properties and dynamics of large protein complexes and complex protein systems.  相似文献

Detecting and characterizing continuous changes in early forest succession using multi-temporal satellite imagery requires atmospheric correction procedures that are both operationally reliable, and that result in comparable units (e.g., surface reflectance). This paper presents a comparison of five atmospheric correction methods (2 relative, 3 absolute) used to correct a nearly continuous 20-year Landsat TM/ETM+ image data set (19-images) covering western Oregon (path/row 46/29). In theory, full absolute correction of individual images in a time-series should effectively minimize atmospheric effects resulting in a series of images that appears more similar in spectral response than the same set of uncorrected images. Contradicting this theory, evidence is presented that demonstrates how absolute correction methods such as Second Simulation of the Satellite Signal in the Solar Spectrum (6 s), Modified Dense Dark Vegetation (MDDV), and Dark Object Subtraction (DOS) actually make images in a time-series somewhat less spectrally similar to one another. Since the development of meaningful spectral reflectance trajectories is more dependant on consistent measurement of surface reflectance rather than on accurate estimation of true surface reflectance, correction using image pairs is also tested. The relative methods tested are variants of an approach referred to as “absolute-normalization”, which matches images in a time-series to an atmospherically corrected reference image using pseudo-invariant features and reduced major axis (RMA) regression. An advantage of “absolute-normalization” is that all images in the time-series are converted to units of surface reflectance while simultaneously being corrected for atmospheric effects. Of the two relative correction methods used for “absolute-normalization”, the first employed an automated ordination algorithm called multivariate alteration detection (MAD) to statistically locate pseudo-invariant pixels between each subject and reference image, while the second used analyst selected pseudo-invariant features (PIF) common to the entire image set. Overall, relative correction employed in the “absolute-normalization” context produced the most consistent temporal reflectance response, with the automated MAD algorithm performing equally as well as the handpicked PIFs. Although both relative methods performed nearly equally in terms of observed errors, several reasons emerged for preferring the MAD algorithm. The paper concludes by demonstrating how “absolute-normalization” improves (i.e., reduces scatter in) spectral reflectance trajectory models used for characterizing patterns of early forest succession.  相似文献

The authors describe a new technique for extracting the intrinsic laser-diode dynamic properties accurately. This simple technique eliminates the need for accurate microwave calibration of the test equipment and problems of microwave reflections, nonideal frequency response of laser mount, and detector. The effect of the parasitic components of the laser diode are also eliminated from the results so that measurements of important dynamic properties of the laser can be found up to high frequencies (10-20 GHz) on standard laser diodes. Blood samples from 740 Egyptian Nationals working in the tourism industry at two sites in the South Sinai governorate were screened for markers of infection with hepatitis B virus (HBV), hepatitis C virus (HCV), human immunodeficiency virus (HIV), and Treponema pallidum. Study subjects included 467 individuals from a rural seashore tourist village and 273 persons at two hotels in a well-established resort town. Subjects' ages ranged from 15 to 70 years; 99.3% were male. The prevalence of serologic markers for currently asymptomatic or past HBV infection alone was 20.7% (n = 153), of markers for past or chronic HCV infection alone was 7.4% (n = 55), and of markers for both HBV and HCV was 6.9% (n = 51). Of the 204 individuals positive for anti-HBV core antibody, 12 (5.9%) were also positive for hepatitis B surface antigen. Two individuals (0.3%) had a serologic market suggestive of an active syphilitic infection. No subject was found to be HIV-seropositive. History of prior injections and number of injections were associated with infection with HCV. Primary residence in the Nile delta and valley areas where schistosomiasis is highly endemic, was also a statistically significant risk factor for HCV, but not HBV infection.  相似文献

Abstract DNA typing of four tetrameric repeat loci (HUMVWA, HUMTH0I, D21SII and HPRT) was carried out in a Chinese Han population from Shanghai (East China) and one from Guangzhou (South-East China) using a quadruplex PCR amplification and detection of the fluorescent-labeled alleles on the ALF DNA sequencer. All loci were in accordance with Hardy-Weinberg equilibrium except for D21S11 in the Guangzhou population. A test for population differentiation showed no statistical difference in the allele frequency distribution between the two populations. Comparison of the allele frequency data with other Chinese Han populations from North and South-West China for the STR loci HUMVWA and HUMTH01 revealed heterogeneity between Northern Chinese Han and Southern Chinese Han, which is in accordance with previous studies on the basis of protein markers.  相似文献

The authors examined over time the relationship between characteristics of the family caregiving system for older persons with Alzheimer's disease and the health of 136 adult offspring who were involved in care. The goal was to determine if family and offspring health relationships found at point of initial patient evaluation (Tl) remained consistent over a 12-month period (T2) and if qualities of the family environment assessed at Tl predicted a change in offspring health and well-being as the demands for care increased over time. Offspring carehours, gender, education, distress, and patient severity (Step 1); measures of family functioning (Step II); and family by distress interaction terms (Step III) were used to predict offspring health and well-being at T2. Findings indicated that (a) family characteristics associated with offspring health at Tl displayed consistent predictions of health over time and (b) family emotion management variables predicted a reduction in offspring health scores by T2. These data document the ongoing effects of the family context of care and suggest the family setting as a target for intervention. Amiodarone is a benzofuran derivative with a chemical structure similar to thyroxine. Originally introduced to treat angina pectoris, amiodarone was found to have antiarrhythmic properties, and in 1985, was approved in the United States for treatment of life-threatening ventricular arrhythmias. It is now used for various ventricular and supraventricular arrhythmias refractory to conventional first-line medications, and as a result, side effects have been observed with increased frequency. The most severe and potentially life-threatening of these side effects is the development of pulmonary toxicity. Typically, amiodarone pulmonary toxicity (APT) is manifested by acute pneumonitis and chronic fibrosis. Amiodarone-associated hemoptysis (AAH) is a rare occurrence. The authors describe a case of AAH successfully treated with cessation of drug and steroid therapy.  相似文献

This study examines whether a catheter mounted left intraventricular balloon may prevent left ventricular (LV) dysfunction following acute experimental myocardial infarction. In 10 anesthetized pigs, multiple coronary arterial ligations were applied around the apex of the heart. LV end-diastolic pressure (LVEDP), aortic flow (AF), and LV long and short axis fractional shortening (FS) were measured before and at 15 min intervals after ligations. At the 60th min after ligation, the LV long axis FS and AF decreased by 7.2 +/- 2.6% (p < 0.05) and 13.25 +/- 2.68% (p < 0.01), respectively, and the LVEDP increased by 4.3 +/- 1.1 mm Hg (p < 0.01) while no change was noted in the LV short axis FS. An intraventricular catheter mounted nonpulsating balloon was positioned over the endocardium of the infarcted area at the LV apex. Inflation of the nonpulsating balloon to an optimal volume, which was found to be equal to 8-10% of the LV end-diastolic volume, resulted in a reduction (by 3.8 +/- 1.2 mm Hg, p < 0.01) of the already increased LVEDP and in an increase (by 6.6 +/- 2.1%, p < 0.05) in the LV short axis FS while no statistically significant change was noted in the AF and LV long axis FS. It is concluded that an intraventricular catheter mounted balloon patch positioned over the endocardium of the infarcted area may ameliorate early LV dysfunction, possibly by interfering with the functional geometry of the LV contraction.  相似文献

The effect of various concentrations of ofloxacin, ciprofloxacin and lomefloxacin on the morphokinetic parameters of the cellular elements of the lung tissue of intact animals (mice, guinea pigs and dogs) was studied under the conditions of the tissue culture. It was shown that in a concentration of 100 micrograms/ml and the exposure time of 24 hours the drugs had no effect on the mobility and structure of the lung tissue cellular elements. When the drugs were used in a concentration of 100 micrograms/ml the mobility rate of the lymphocytes and macrophages proved to by markedly retarded by the end of the 24th hour. In a concentration of 1000 micrograms/ml the drugs killed the cell culture. There were detected no specific differences in the effect of the fluoroquinolones on the cellular elements of the lung tissue of the intact animals.  相似文献
