The N-acylethanolamine phospholipids (NAPE) are precursors for N-acylethanolamines (NAE), including anandamide (20∶4-NAE), which is a ligand for the cannabinoid receptors. Previously, NAPE were believed to be found only in injured tissue, e.g., after neurodegenerative insults. Neuronal injury may occur in response to seizure activity. Therefore, we investigated the effect of pentylenetetrazol (PTZ)-induced seizures in PTZ-kindled mice on the level of NAPE in the brain. Male NMRI mice were kindled with PTZ injections 3 times/wk, thereby developing clonic seizures in response to PTZ. Mice were killed within 30 min after the clonic seizure on the test day (12th injection) and the brains were collected. Eight species of NAPE were analyzed as the glycerophospho-N-acylethanolamines by high-performance liquid chromatography-coupled electrospray ionization mass spectrometry. No effect of the PTZ kindling on the NAPE levels in murine brains was observed. Total NAPE in control mice cortex (n=4) was 16.4±3.0 μmol/g wet weight of which 20∶4-NAPE accounted for 3.6 mol%, and the major species was 16∶0-NAPE, accounting for 52.1 mol%. Determination of the activity of NAPE-hydrolyzing phospholipase D and of N-acyltransferase in brain membrane preparations from adult and 3-d-old mice revealed an enzyme pattern in the adult mice that was favorable for NAE accumulation as opposed to NAPE accumulation. Thus, there was no difference in NAPE levels; at present, however, this does not exclude that NAE may accumulate during seizure.  相似文献

This paper describes the purification and characterization of microviridin J, a newly discovered metabolite of Microcystis that causes a lethal molting disruption in Daphnia spp., upon ingestion of living cyanobacterial cells. Microviridin J consists of an acetylated chain of 13 amino acids arranged in three rings and two side chains. Unlike other known isoforms of microviridin, microviridin J contains arginine that imparts a unique solution conformation characterized by proximal hydrophobic interactions between Arg and other regions of the molecule. This eventually results in the formation and stabilization of an additional ring system. Microviridin J potently inhibits porcine trypsin, bovine chymotrypsin, and daphnid trypsin-like proteases. The activity against trypsin is most likely due to Arg and its distinctive conformational interactions. Overall, the data presented for microviridin J emphasize once again the ability of cyanobacteria to produce numerous and potent environmental toxins.  相似文献

In this paper a general method for dimensional synthesis of both planar and spatial mechanisms is presented. The method addresses the problem of non-assembly. A gradient based optimization algorithm is employed and the sensitivities are calculated analytically using the method of direct differentiation. A well established and general method of computational kinematics is employed, in a slightly modified variant, to avoid the problem of non-assembly. The modification consists of a minimization of the residuals of the kinematic constraint equations, rather than equating them to zero. The point-coordinate formulation is applied. We consider ways to use monotonicity to reduce the excess width (due to dependence) of computed intervals. Use of monotonicity generally involves evaluation of a derivative. We show how monotonicity can often be used without evaluating derivatives. As examples, we show how Gaussian elimination and evaluation of slopes can be sharpened. A variable amount of extra computing is required to obtain the sharper results.  相似文献

This paper formulates the eigenvalue sensitivity of a stiffened plate with respect to stiffener location; the analysis is based on the generalized Rayleigh quotient of the combined platebeam system. The results show that the eigenvalue sensitivity is proportional to the force between the plate and the stiffener as well as to the slope of the eigenfunction at the interface between the plate and the stiffener.  相似文献

We introduce an image enhancement method referred to as the watershed-based maximum-homogeneity filter. This method first uses watershed analysis to subdivide the image into homogeneous pixel clusters called catchment basins. Next, using an adaptive, local, catchment-basin selection scheme, similar neighboring catchment basins are combined together to produce an enhanced image. Because the method starts with watershed analysis, it can preserve edge information and run with high computational efficiency. Illustrative results show that the method performs well relative to other popular nonlinear filters.  相似文献

The far held of an electric Hertzian dipole located in a borehole is expressed as the product of a transmission coefficient and the far field of a dipole in a homogeneous region. The derivation uses the method of steepest descent, and gives physical insight into the wave phenomena that are important for borehole radar. The received field inside the borehole due to a planar interface located in the vicinity of the borehole is calculated using geometrical optics and the principle of reciprocity. The resulting expression is validated by numerically comparing it with a finite-difference time-domain solution. In an idealized situation, the reflection from the oil-mater boundary in a horizontal well can be detected with a borehole radar  相似文献

Dietary sources of fatty acids were evaluated for their influence on oocyte quality and follicular development using 54 lactating cows in summer. Fat supplements were 1) sunflower oil (80% cis 18:1), 2) Ca salt of transoctadecenoic acids (57% trans 18:1), 3) Ca salt of vegetable oils (30% 18:2), and 4) linseed oil (56% 18:3 and 16% 18:2). Fats were fed at 1.35% of dietary dry matter beginning at 5 wk prior to expected calving date and at 1.5% (oils) and 1.75% (Ca salts) of dietary dry matter for 15 wk after parturition. Four days following a programmed induced ovulation, 5 transvaginal oocyte aspirations were performed 3 or 4 d apart. Three days after the last aspiration, PGF was injected, followed 3 d later by a GnRH injection and a timed artificial insemination (d 0) 16 to 20 h later. For the first 4 aspirations, oocytes grading 1 or 2 were used for in vitro embryo production. Total cell number and the proportion of terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL)-positive blastomeres were analyzed at d 8. At the fifth aspiration, the occurrence of metaphase II, group II caspase activity, and TUNEL labeling were determined after oocyte maturation. A total of 1,011 oocytes were collected. The proportion of oocytes with high caspase activity was greater for grade 3 compared with grades 1 and 2 (37.5 vs. 1.54 and 1.61%). Feeding polyunsaturated fatty acids, as compared with monosaturated fatty acids, failed to affect oocyte quality, as demonstrated by subsequent embryo development. Cows fed 18:2- or 18:3-enriched diets had a larger preovulatory follicle at insemination and subsequent volume of the corpus luteum compared with those fed cis 18:1 or trans 18:1 diets (16.8, 16.2 vs. 15.0, 14.9 ± 0.7 mm; 7,323, 8,208 vs. 6,033, 5,495 ± 644 mm3, respectively). The previously documented benefits of polyunsaturated fatty acids on reproductive performance appear to reflect actions at alternative biological windows in lactating dairy cows.  相似文献
